HBCD는 생물 축적성 및 독성, 지속성, 장거리 이동 특성으로 인해 2013년 스톡홀름 협약에서 시급히 규제가 필요한 지속성유기오염물질로 지정되었다. 본 연구에서는 양식장, 도심, 비도심과 같은 연안 이용 방식이 다른 해역에서 미세플라스틱의 오염 수준과 특성을 살펴봄으로써, 해역 별 인간 활동에 따른 미세플라스틱 오염과의 상관성을 파악하고 해양 기반 오염원의 중요성을 이해하고자 하였다. EPS가 대량으로 사용되는 양식해역에서 EPS 미세플라스틱의 오염과 생물 노출 현황을 파악하고 EPS 해양쓰레기에 서식하는 해양생물의 미세플라스틱 섭식과 서식 생물에 의한 미세플라스틱의 생성을 살펴보았다. 국내 연안에 분포하는 대형 및 미세 EPS 해양쓰레기에서의 HBCD 함유 수준을 확인하고, 아시아-태평양 지역 12개 국가 해안의 EPS 해양쓰레기를 확보하여 EPS 해양쓰레기를 통한 유해 화학물질 오염이 지구적인 환경 이슈가 될 수 있음을 확인하였다. EPS 해양쓰레기에 함유되어 있는 HBCD가 EPS에 부착하여 서식하는 생물로 전이되고 있음을 실환경에서 증명하였으며, 실험실에서 전이 경로를 탐구하였다. 마지막으로, 해양쓰레기 오염 외의 다른 오염원이 제한적인 청정지역(외딴 섬)을 연구지역으로 선정하여, 해양환경에서 EPS 해양쓰레기를 통한 유해화학물질의 이동과 확산을 규명하고자 하였다. 연구에 앞서, 해양환경의 다양한 매질(해수, 퇴적물, 생물, 플라스틱쓰레기)에서의 플라스틱 기원 오염물질 분석방법(추출, 정제, 기기 분석)을 확립하였다. 또한, 해수, 퇴적물, 생물 중에서의 미세플라스틱을 분석하기 위하여 분석방법(유기물제거, 밀도분리 방법)을 적립하였다. 기원오염물질과 미세플라스틱 분석은 정도 관리를 실시하여 분석방법을 검증하였다. 도심, 비도심, 양식장 지역에서의 미세플라스틱 분포 특징을 조사하여, 인간활동에 따른 미세플라스틱 오염 특성에 대해 연구하였다. 퇴적물과 해수 중 미세플라스틱 폴리머 조성이 조사 지역별로 상이하게 나타났다. 도심지역에서는 높은 폴리머 다양성을 보였으며, 이는 도심의 하천수에서의 분포 경향을 잘 반영하고 있었다. 반면 소규모의 어촌 마을이 위치한 비도심 지역에서 폴리머 다양성이 가장 낮았으며, 밧줄이나 어구에 사용되는 폴리프로필렌(polypropylene, PP)이 우점하였다. EPS 부자를 사용하여 굴 양식을 하는 해역에서는 폴리스티렌(polystyrene, PS) 미세플라스틱이 가장 많이 발견되었다. 종합하면, 해양 매체인 해수, 퇴적물에서 미세플라스틱의 조성은 해역의 이용 특성을 잘 반영하고 있으며, 각 해역에 서식하는 생물(이매패류, 갯지렁이)에 반영되어 있었다. 도심지역이 비도심지역에 비해 높은 미세플라스틱의 농도를 보였으나 양식장 지역의 농도는 도심지역과 비슷하거나 높은 수준이었다. 이번 연구 결과로 육상 기인 오염원과 더불어 해상 기인 오염원이 미세플라스틱의 주요 발생원이 되고 있음을 확인하였다. EPS 해양쓰레기의 내측과 외측에 해조류, 멍게류, 소형어류, 게, 지렁이류 등 다양한 생물이 서식하고 있음을 확인하였다. EPS 내측에 대형 갯지렁이류인 바위털갯지렁이(Marphysa sanguinea)가 굴을 파고 서식하고 있으며, 굴의 외벽에서 미세한 EPS 알갱이가 다량 관찰되었다. EPS 해양쓰레기에서 채집한 바위털갯지렁이의 체내에서 0.2-3.8 mm 크기의 EPS 조각이 개체 당 평균 131개 확인되었다. 바위털갯지렁이로 인한 EPS 미세플라스틱 생성을 확인하기 위해, 실험실에서 바위털갯지렁이를 여과한 해수와 함께 EPS 블록에 노출한 결과, 바위털갯지렁이가 EPS에 굴을 파고 들어가면서 하루 평균 1600개의 미세 EPS 조각을 생성함을 확인하였다. 본 연구 결과는 해양환경에서 자외선, 물리적 마찰과 같은 물리적 요인과 더불어 생물학적 요인이 미세플라스틱의 생성에 기여하고 있음을 규명하였다. 환경 중 EPS 해양쓰레기에 함유된 HBCD의 농도 수준을 확인하기 위해, 우리나라 해변에서 다양한 종류의 EPS 해양쓰레기(60L 소형부자, 200L 대형부자, 200L 재활용부자, 미세플라스틱)를 채취하였다. 분석 결과, HBCD가 EPS 미세플라스틱을 포함한 대부분의 EPS 해양쓰레기에서 검출되었다. HBCD를 고농도로 함유하고 있는 건축용 EPS 폐기물이 대형 양식용 부자 내측에 재사용 되고 있음을 확인하였다. 이는 건축용 EPS 폐기물이 해양 활동(e.g. 소형 선박, 양식장 등)에 재활용 될 경우 다량의 HBCD 해양 유출을 초래 할 수 있음을 시사한다. 아시아-태평양 12개 국가의 해안에서 EPS 해양쓰레기를 확보하여 분석한 결과, 모든 시료에서 HBCD가 검출되었다. 특히 알래스카 해변에서 발견된 일본 쓰나미 EPS 쓰레기에서 건축자재 수준의 높은 HBCD 농도가 확인되었다. 이는 해양쓰레기를 통해 유해 화학물질이 장거리 이동하고 있으며, 해양환경에서 해양쓰레기가 오염물질을 이동∙확산시킬 수 있음을 의미한다. 또한 EPS 해양쓰레기의 첨가오염물질 문제가 우리나라뿐만 아니라 전 세계적인 환경 이슈가 되고 있음을 보여주는 결과이다. EPS 해양쓰레기에 부착하여 서식하는 담치 중의 HBCD 농도와 이성질체 조성을 통해, EPS로부터 HBCD의 생물 전이를 증명하고자 하였다. 비교를 위하여 동일한 해역 내 여러 다른 기질(폴리에틸렌 부자, 금속부자, 자연 암반)에 서식하는 담치를 함께 채집하여 분석하였다. 다른 기질에 비해, 고농도(5160 ng/g lipidweight)의 HBCD가 EPS 서식 담치에서 관찰되었으며, EPS 부자의 HBCD 이성질체 조성(α,β,γ-HBCD)이 EPS 서식 담치에 잘 반영되어 나타났다. 본 연구를 통해, 플라스틱 해양쓰레기 첨가 화학물질이 부착하여 서식하는 생물에게 전이됨을 실환경에서 최초로 확인하였다. 플라스틱에서 첨가 물질이 생물로 전이되는 경로를 확인하고자, 두 가지 서로 다른 크기의 EPS 입자를 제조하여 실험실에서 담치에게 노출시켰다. 섭식 불가능한 크기의 EPS 알갱이(4.6±0.33 mm)를 노출시켜 EPS에서 용출된 HBCD의 해수를 통한 생물 축적을 살펴보았고, 섭식 가능한 크기의 EPS 입자(20-770 µm)를 노출시켜 EPS 입자의 생물 섭식을 통한 HBCD의 축적을 살펴보았다. 두 EPS 노출구에서 대조구보다 월등히(약 15배) 높은 HBCD가 검출되었으며, 이는 EPS에 함유된 HBCD가 해양 생물에 전달될 수 있음을 보여준다. 노출 기간 동안 두 노출구에서 해수 중 HBCD의 농도가 동일하도록 실험 조건을 설계하였기 때문에, EPS 입자 노출구에서 섭식의 효과로 인해 더 높은 HBCD 생체 축적을 예상했으나, 노출 4일, 10일 경과 후 두 그룹의 담치 중 HBCD 농도는 비슷한 수준을 나타냈다. 이는 해양무척추동물에서 물을 통한 간접적인 노출이 HBCD 생체 축적에 중요한 경로가 될 수 있음을 의미한다. “해양에서 해양쓰레기를 이동을 통해 청정해역에 오염물질이 전달되고 서식 생물에 영향을 미칠 수 있다”라는 가정을 규명하기 위하여, 해양쓰레기 오염도가 크게 다르나 인위적 오염원이 없는 두 개의 섬을 연구정점(오염지역, 비오염지역)으로 선정하였다. 해양쓰레기 조사를 통해 지역간 해양쓰레기 오염도 차이를 확인하였으며, 두 정점에서 해양쓰레기 풍도는 78±39 개/m2, 0.5±0.3 개/m2로 매우 큰 차이를 나타냈다. 오염 지역의 플라스틱 해양쓰레기 중 EPS 재질이 차지하는 비중은 개수 대비 79%로 우세하였다. 두 지역에 모두 서식하는 생물 5종(갯강구, 삿갓조개, 타래고둥, 갈고둥, 담치)을 채집하여 첨가오염물질(HBCD)의 체내 농도 수준을 비교하였다. 첨가물질과 비교를 위해 해양환경 중에 널리 분포하여 해양쓰레기로 인한 기여도가 상대적으로 낮을 것으로 추정되는 대표적 흡착오염물질인 폴리클로리네이티드비페닐(polychlorinated biphenyl, PCB)를 함께 분석하였다. PCB의 경우 정점 간 생물 내 농도 차이가 없었으나, HBCD는 해양쓰레기 오염도가 높은 지역에 서식하는 생물에서 낮은 지역의 생물에서 보다 유의하게 높은 농도를 보였다. HBCD 외의 다른 플라스틱 첨가제(산화방지제)에서도 동일한 지역간 분포 경향이 확인되었다. 본 연구는 해양환경에서 해양쓰레기를 통해 청정한 해역에 플라스틱 기인 화학물질이 이동∙확산되고 서식 생물에 전달되고 있음을 실제 환경에서 증명한 첫 사례이다.

Plastic debris is global environmental problem affecting marine life from low to high trophic levels of organisms. Plastic debris cause not only just physical effect such as ingestion and entanglement in the marine species, but also chemical effect such as bioaccumulation of plastic induced hazardous compounds which can subsequently lead to further effects. Additives are normally added in plastic product to improve performance of the polymer. Plastic products and their debris contain a complex mixture of additive chemicals at up to 70% of total volume, and some of them are endocrine disruptor chemicals (EDCs) or are listed as persistent organic pollutants (POPs). But, research regarding plastic-associated chemicals such as quantitative measure in plastic debris, sorption-desorption properties, and bioaccumulation of chemicals to organism through microplastic ingestion has mainly focused on sorbed chemicals such as polychlorinated biphenyls (PCBs) and organochlorine pesticides. Many additives that are low molecular weight and are not covalently bound to the polymer backbone, are likely to leach out of the plastic to environments. Therefore, it is concerned that plastic debris raise overall additive chemical levels in surrounding environmental media, subsequently increase the bioaccumulation of these chemicals throughout marine food webs, including humans. However, their environmental exposure, fate, and ecological impact via plastic debris pollution in the marine environment are largely unknown. Polystyrene (PS) which is mainly expanded PS (EPS) and extruded PS (XPS) is a major constituent of plastic marine debris throughout the world. Especially, EPS buoys are the most common item of beach debris (size: >2.5 cm) in South Korea, and its fragments accounted for the predominant microplastics (size range: 1-5 mm; 93% of total microplastics) along the Korean coast. EPS marine debris have high amount of additive hexabromocyclododecans (HBCDs; 0.2% to 2.4% by weight). Owing to its properties, HBCD was designated as a global elimination compound in the Stockholm Convention on Persistent Organic Pollutants (POPs) in 2013. Due to light weight, EPS debris has a great potential for long-range transport in the ocean, accompanying the movement of HBCD additive. Once EPS reach somewhere for example remote uncontaminated region, the plastic debris and their fragments are likely to have the potential to influence surrounding environments and marine organisms living there, as a source of HBCD. This study, firstly, investigated the contamination levels and characteristics of microplastics in marine matrices from three coastal regions with different coastal use pattern such as aquafarm, urban, and rural areas, and identified the importance of sea-based activity as a source of microplastics in the marine environment, particularly aquaculture activities using EPS buoys. And then, organisms inhabiting EPS debris were observed, and its interaction with EPS substrate was studied. To identify the overall levels of HBCD in EPS marine debris including microplastics, an intensive monitoring study was conducted in South Korea, and expanded to 12 countries from the Asia-Pacific region. After that, transfer of HBCD from EPS debris to inhabiting organisms was investigated in the marine environment, and the transfer routes (such as aqueous medium and particle ingestion) of HBCD from EPS to mussel were assessed through lab exposure experiment. Finally, the potential of EPS debris as a vector of additive HBCD to the pristine marine environment was evaluated through a comparative study on marine organisms inhabiting marine debris-contaminated and uncontaminated islands. Microplastic contamination of abiotic matrices (seawater and sediment) and biotic matrices (bivalves and polychaetes) in three coastal regions characterized by different types of human activity, including urban, aquafarm, and rural areas was investigated. In the abiotic matrices, different microplastic polymer compositions were observed among three sites. Diverse polymers were found in marine matrices from the urban site, implying diverse microplastic sources in highly populated and industrialized areas. Polystyrene was more abundant in the aquafarm site, reflecting the wide use of EPS aquaculture buoys. Polypropylene was more abundant at the rural site, probably due to the use of polypropylene ropes and nets in fishing activity. A marine invertebrate microplastic accumulation profile showed trends similar to those exhibited by abiotic matrices, reflecting regional variation in human activities. These results indicate that marine microplastic are generated from both marine-based sources such as aquaculture farm and fishing activities, and that the abiotic and biotic marine matrices reflect the microplastic characteristics. Organisms inhabiting EPS debris collected off the cost of Korea were observed. Polychaetes (Marphysa sanguinea) were found deep inside EPS debris. A large number of EPS particles (131 ± 131 particles/individual, 0.2–3.8 mm in length) were found in the digestive tracts of burrowing polychaetes living on EPS debris. To confirm the formation of microplastics by polychaetes and identify the quantity and morphology of produced microplastics, polychaetes were exposed to EPS blocks in filtered seawater under laboratory conditions. Polychaetes burrowed into the blocks and created numerous EPS microplastic particles, indicating that a single polychaete can produce 0.5 million microplastics per year. These results reveal the potential role of marine organisms as microplastic producers in the marine environment. To obtain a better understanding of chemical dispersion via EPS pollution in the marine environment, intensive monitoring of HBCD levels in EPS debris and microplastics was conducted in South Korea, where EPS is the predominant marine debris originated mainly from fishing and aquaculture buoys. HBCD was detected extensively in EPS buoy debris and EPS microplastics stranded along the Korean coasts, which might be related to the detection of a quantity of HBCD in non-flame-retardant EPS bead (raw material). The high detection frequency (90%) of the flame retardant in sea-floating buoys, and the recycling of high-HBCD-containing EPS waste inside large buoys (range: 424-3,880 µg/g) highlight the need for proper guidelines for the production and use of EPS raw materials, and the recycling of EPS waste. HBCD was also abundantly detected in EPS debris (0.98-14,500 µg/g) collected from the Asia-Pacific coastal region, indicating that HBCD contamination via EPS debris is a common environmental issue worldwide. Suspected tsunami debris from Alaskan beaches indicated that EPS debris has the potential for long-range transport in the ocean, accompanying the movement of hazardous chemicals. It is hypothesized that if EPS debris acts as a source of the additives in the marine environment, organisms inhabiting such debris might be directly influenced by them. Therefore, the characteristics of HBCD accumulation by mussels inhabiting EPS was investigated. For comparison, mussels inhabiting different substrates were also studied. The significantly high concentration of HBCD was detected in mussels from EPS debris (mean: 523±1090 ng/g lipid weight; l.w.) than its from such as high-density polyethylene (HDPE) buoy (60±17 ng/g l.w.), metal buoy (55±21 ng/g l.w.), and natural rock (57±21 ng/g l.w.). The high HBCD levels up to 5160 ng/g l.w. and the γ-HBCD dominated isomeric profiles in mussels inhabiting EPS strongly supports the transfer of HBCDs from EPS substrate to mussels. Furthermore, microsized EPS particles were identified inside mussels, probably originating from their substrates. The transfer routes of additive HBCD from EPS to marine organisms were investigated in lab condition, by exposing mussels to EPS of two different sizes: EPS particle (20–770 μm) and EPS bead (4.6 ± 0.33 mm). To study the uptake of leached HBCD in aqueous phase, EPS bead was exposed to mussel group (EPS leachate group; hereafter EPS-L group). At the other group, EPS particle were exposed to see the dietary (ingestion) effect together with aqueous uptake (EPS leachate and ingestion group; hereafter EPS-L·I group). Since the HBCD concentration in seawater was similar between two groups, HBCD concentration of mussel expected to be higher in EPS-L·I group than EPS-L group. However, there was no statistically difference (t-test, p>0.05) between HBCD concentration of mussels from EPS-L (4710±2490 ng/g l.w.) and EPS-L·I (5300±1070 ng/g l.w.) groups, indicating that aqueous uptake of leached HBCD from EPS is more important route than ingestion of EPS, in bioaccumulation of additive HBCD. This result highlight that aqueous exposure of leached additives from plastic is significant exposure pathway in marine invertebrate. To identify the potential of EPS debris for the transfer and dispersion of chemicals to the pristine marine environment, two islands located in the southern part of Korea with very different EPS debris pollution levels were selected as study sites. The exposure levels of HBCDs in six kinds of marine organisms inhabiting each island were assessed and compared. Along with HBCDs, PCBs, a typical sorbed chemical in plastic debris was also analyzed. No difference was observed in PCBs levels in organisms between islands, while the HBCD concentration was significantly higher in organisms inhabiting marine debris contaminated islands than those inhabiting non-contaminated island (Mann-Whitney, p<0.05). To confirm if marine plastic debris can be a source of other additives in the environment, antioxidants and benzotriazole ultraviolet (UV) stabilizers were analyzed. Antioxidants displayed similar spatial distributions with HBCD between the sties. The result of this study indicates that plastic marine debris has a strong potential for source and carrier of plastic additives chemicals in the marine environment.
