There are methods for collecting blood such as the retroorbital plexus, facial vein, saphenous vein, sublingual vein, lateral tail vein, and cardiac puncture in rodents. The most general site used for blood sampling in mice is the retroorbital plexus (RP). It is a controversial issue because the process is easy and can get a lot of blood, but it can cause terrible pain and severe sequelae in experimental animals. In addition, methods of blood collection in RP require skilled techniques and anesthesia compared to other methods of blood collection. As an alternative to the blood collection method in RP, the blood collection method in facial vein (FV) had recently being interested in for the advantage of no need to anesthesia. There has been a comparative analysis of the effects of blood sampling using the RP and FV on stress influence, but no comparative studies have been reported on experienced and novice researchers based on blood collection methods. In this study, the investigator compared and analyzed the hematological and immunological aspects of blood collection between the experienced and novice groups after sampling the RP and FV. Sixty 6weeks-old ICR mice were used for 6 experiments in 2 weeks, and body weight changes were measured according to the number of blood collections and 0.4 ml blood sampling time to determine the difference between the blood collection methods. Free cortisol levels were measured by ELISA to assess the effect of stress on blood collection and animal handling. For comparative analysis of stress and immune cells, the spleens of the mice were collected, weighed and compared after 6 blood samplings. In addition, tissue damage was analyzed on the tissues collected from both of blood collection sites. There were no different changes in body weight between the two groups (experienced and novice) compared to the control groups. Compared to the experienced group, FV is relatively faster in blood collection time compared to RP, and as the number of blood collection increases, the techniques of novice group increase rapidly. On the other hand, in RP, novice group showed great differences in damage to experimental animals due to immaturity of their techniques, and it takes a long time to master the technique. As time passed, both groups showed eye damages caused by retroorbital plexus bleeding. Free cortisol levels (a type of stress hormone) in FV were also lower in the two groups than in RP. The size and cells of the spleen are relatively low in FV than in RP. Because the distribution of granulocytes and macrophages in the blood and spleen is immunologically related to the stress, FV was relatively lower than that of RP. In conclusion, the facial vein collection method is superior to the retroorbital plexus collection method in terms of collection time, hematology analysis, immunology analysis, and the ethics. When comparing tissue damage and blood collection stress indicators, facial vein collection is suitable for experiments that require medium/long-term blood collection and blood collection times.
