Despite their temporal crop disease suppression and yield improvement roles, uncontrolled utilization of agrochemicals has unignorable negative impact on human health, animal life, qualities of soil and water, as well as the overall environment entities. So far, tremendous and intensive research works at varying levels had been undertaken to get environmentally sound and economically feasible alternative sources. The effort has shown remarkable breakthroughs in introducing sustainable farming technology. The main agents in such environmentally friendly agriculture are soil microorganisms. However, the work done to fully utilize the natural grace in the soil and address the problems at the grassroots level is at its early stage and have limited access to many. As a part of the vast natural grace utilizers, the current work has been undertaken with the overall objective of isolation and evaluation of a bacterium for its biotic and abiotic plant stress management. A gram-positive, spore-forming, halotolerant, and thermophilic Bacillus species was screened from a soil in the Muan District, the Republic of Korea. Identification of the isolate as Bacillus velezensis AR1 was made through 16s rRNA sequence analysis. Investigation of the test bacterium under invitro condition demonstrated strong volatile and agar diffusible antimicrobial activities ranging from 7.2 to 26.5mm; and 8.3 to 100% growth inhibition, respectively, against the target phytopathogens. The strain also produced several biochemicals that have a direct and indirect contribution to the growth of the host plant. Production of plant growth-regulating hormones, catalytic enzymes, siderophores, ammonia, etc. are among the most important traits of the isolate. Further analysis on the solubilization of hardly soluble plant nutrients was another important trait to qualify the bacterium as plant growth-promoter. As such, the isolate AR1 showed a remarkable solubilizing activity on insoluble phosphorus and zinc sources. A bioactive secondary metabolite was purified from the active layer of ethyl acetate and further subjected to nuclear magnetic resonance (NMR) analysis. From the analysis of the heteronuclear multiple bond correlation (HMBC), the purified compound was identified as monomethyl succinate (MMSucc). The subsequent investigation of the antifungal activity of the organic compound resulted in growth inhibition of 5.7 to 100%, 8.8 to 96.7%, and 7.2 to 71.3% on R. solani, S. sclerotiorum and B. cinerea at 25 and 550µg mL-1 MMSucc, respectively. Furthermore, the purification process conducted on the n-butanol extract resulted in an identification of a new dipeptide amino acid organic compound called 5-N-tyrosinylornithine. This metabolite has also showed strong antifungal activity. Pot experiment was conducted to assess disease control efficacy of the isolate against Alternaria sesami, a causal pathogen of sesame (Sesamum indicum L.) leaf spot/blight. The result indicated that the sole and alternate application of AR1 with fungicide significantly suppressed disease severity by reducing it to less than 10%. The probable reason could be attributed to the antifungal secondary metabolites produced by the isolate in the culture broth. Furthermore, the isolate induced plant defense-related biochemicals viz., total phenolic and flavonoid contents, and amino acid proline concentration in the leaf.
