Sclerotinia sclerotiorum and Sclerotinia minor are major fungal pathogens which cause lettuce drop disease resulting in serious losses of global lettuce production. This study focused on control of S. sclerotiorum and S. minor by Bacillus velezensis CE and Bacillus licheniformis MH48 under in vitro and in vivo conditions. Two Bacillus strains produced major metabolites for fungal inhibition such as siderophores, various lytic enzymes including chitinase, protease, β-1, 3-glucanase as well as biosurfactant. These two Bacillus strains showed phosphate solubilizing activity, an important trait for plant growth promotion. Moreover, volatile organic compounds released by these Bacillus strains highly restricted mycelial growth of target fungal pathogens. Active metabolites present in the culture filtrates of the two strains strongly affected to mycelial growth and sclerotia germination of S. sclerotiorum and S. minor. The broth cultures of the strain CE and strain MH48 were also effective to reduce Sclerotinia disease of detached lettuce leaves caused by S. sclerotiorum. Due to their ability to grow at high temperature, large scale inoculation of two Bacillus strains were successfully performed in non-sterilized low cost medium at 50℃. Application of large-scale mass cultures of two Bacillus strains significantly suppressed lettuce drop under in vivo conditions. Complete suppression of lettuce drop was observed in treatment with combined application of strain CE, strain MH48 and fungicide. Therefore, the results of our study revealed that B. velezensis CE and B. lichneformis MH48 could be considered as effective candidates for control of lettuce drop. Moreover, large-scale cultures of two Bacillus strains through onsite mass cultivation in low cost medium could be a reliable strategy for successful application of biocontrol agents under field conditions.
