본 연구에서는 부위별 뽕나무(뿌리껍질, 잎, 줄기, 열매)의 총 페놀 및 플라보노이드 함량을 확인하여 그 중 페놀 및 플라보노이드 함량이 가장 높았던 상백피(뽕나무 뿌리껍질)를 각 농도별 에탄올로 추출하여 항산화 활성을 확인하였다. HPLC 정성 및 정량분석을 통해 상백피 추출물에 함유되어 있는 플라보노이드를 확인하였으며, 과산화수소로 유도한 신경세포에 대하여 상백피 추출물 및 morusin의 세포 보호효과를 확인하였다. 부위별 뽕나무 추출물의 총 페놀 및 플라보노이드 함량을 확인한 결과 상백피에서 가장 높은 페놀(65.14 ± 1.4 µg/ GAE)과 플라보노이드 함량 (30.79 ± 2.7 µg/ NE)을 확인할 수 있었다. 농도별 에탄올(증류수, 30, 50, 70, 100%)을 가하여 각각 상백피를 추출한 뒤, 농도별 에탄올 상백피 추출물의 페놀 및 플라보노이드 함량을 확인하였고, ORAC(oxygen radical absorbance capacity) 실험, DPPH(1,1-diphenyl-2-picrylhydrazyl) 라디칼 소거 실험, ABTS(2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) 라디칼 소거 실험, FRAP(Ferric reducing antioxidant power) 실험, Fe2+ chelate 실험을 통해 항산화 활성을 확인하였다. 그 결과 70% 에탄올 상백피 추출물에서 가장 높은 페놀 및 플라보노이드를 함유하고 있었으며, 높은 항산화 활성을 나타내었다. 상백피의 플라보노이드 지표성분으로 morusin이 보고되어 있으며, 본 연구의 상백피 추출물의 HPLC 정량 분석 결과 morusin이 다량 함유되어 있는 것으로 확인되었다. 가장 높은 항산화 활성을 보였던 70% 에탄올 상백피 추출물과 지표성분인 morusin의 PC12 신경세포에 대한 보호 효과를 확인하기위해 H2O2로 유도한 PC12 신경세포에 70% 에탄올 상백피 추출물과 morusin을 각각 처리하였으며, MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) 실험을 통해 세포 생존율을, LDH(Lactate dehydrogenase) 실험을 통해 세포막 보호효과를 확인하였다. MTT 실험 결과 70% 에탄올 상백피 추출물을 25∼200 µg/mL로, morusin을 3.1∼6.25 µg/mL의 농도로 처리 했을 때 세포의 생존율이 증가하는 경향을 보였으며, LDH 실험 결과 농도 의존적으로 LDH 함량이 감소하는 경향을 보였다. 본 연구결과를 통해 상백피와 morusin의 항산화 활성 및 신경세포 보호 효과를 확인할 수 있었으며 기능성 소재로서의 가능성을 확인할 수 있었다.

Cell viability was significantly increased when treated 25∼200 µg/mL for 70% ethanol extract of rootbark of morus alba L. and 3.1∼6.25 µg/mL for morusin in MTT assay and LDH amount was decreased in dose dependant manner in LDH assay. The results of this study suggest that root bark fo Morus alba L. and morusin have antioxidative and neuronal cell protective effect and have potency to be a functional source.
