유방암은 여성의 주요 사망 원인 중 하나이며, 치료에는 암세포의 증식과 세포 자멸사의 조절이 필요합니다. 본 연구에서는 MCF-7 인간 유방암 세포에 대한 jaceosidin의 처리가 세포 증식 억제와 세포 자멸사 유도에 대한 시간 및 농도 의존적인 효과에 대해 조사하였다. 세포의 형태 학적 변화, MTT assay, western bolt을 통한 세포 자멸사 관련 단백질의 발현 측정, FACS를 이용한 Annexin V/PI 염색 및 측정을 통해 jaceosidin의 MCF-7 세포 증식 억제 및 세포 자멸사 유도 효과를 특성화 하였다. MTT 분석 결과는 jaceosidin 처리가 MCF-7 암 세포 생존을 용량 및 시간-의존적 방식으로 억제함을 보여주었다. MTT assay를 시행한 결과 jaceosidin의 처리는 MCF-7 세포의 세포 생존율을 농도 및 시간 의존적으로 감소시켰으며, 정상 세포인 EA.hy926에는 영향을 미치지 않음을 확인했다. Western blot을 통해 세포 자멸사 관련 단백질 발현을 조사한 결과, BAX/Bcl-2의 발현 비율 및 PARP의 분절이 증가했고 AKT, ERK 및 p38 MAPK의 인산화가 Jaceosidin의 처리 농도 의존적으로 증가하였다. 또한 FACS를 이용한 AnnexinV/PI 염색 및 측정 결과, jaceosidin의 처리에 의해 MCF-7 세포의 Early 및 Late 세포 자멸사가 유도된 세포 수가 증가함을 추가로 확인했다. 전반적으로, 본 연구의 결과는 jaceosidin의 처리는 MCF-7 인간 유방암 세포의 AKT 및 ERK, p38 MAPK 경로의 활성화를 통해 세포 자멸사를 유도함을 보여준다.

Breast cancer is a leading cause of death in women, and its treatment requires a balance between proliferation and apoptosis. This study investigated the apoptotic effect of jaceosidin at various concentrations (0, 10, 20 and 40 µM) on MCF-7 human breast cancer cells for 24 and 48 h treatment time. The MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay, pro-apoptotic gene expression, flow cytometry, as well as cell morphological changes were used to characterize apoptosis in the cancer cells. MTT assay results revealed that jaceosidin treatment inhibited MCF-7 cancer cell viability in a dose and time-dependent manner. However, no apoptosis was induced on human EA. hy926 endothelial cells, confirming its non-cytotoxicity. Apoptotic cell death of MCF-7 cell was induced by an increase in the pro-apoptotic gene expression of cleaved PARP and a coaction between pro-apoptotic (Bax) and anti-apoptotic (Bcl-2) members of the Bcl-2 family, Phosphorylated ERK, p38, and AKT. Apoptosis was further confirmed in the cells by a flow cytometric analysis which indicated both early and late apoptosis of the cells in varying percentages as treatment concentration increased. Overall, our results show that jaceosidin induces apoptosis in MCF-7 breast cancer cell through Erk, p38, and MAPK pathway.
