This study aimed to elucidate whether 1-methylcyclopropene (SmartFreshTM, 1-MCP) treatment delays the fruit softening mechanism associated with the fruit quality of newly released four Korean apple cultivars (‘Summer Prince’, ‘Summer King’, ‘Green Ball’, and ‘Hwangok’) during cold storage. The ethylene was produced highly in ‘Summer Prince’, moderately in ‘Summer King’ and ‘Green Ball’, and a very low amount in ‘Hwangok’ whether the fruits were stored during shelf-life or cold storage. For all apple cultivars during cold storage, the 1-MCP-treated fruits exhibited lower internal ethylene concentration, higher firmness, and higher titratable acidity compare with control fruits, in association with less fruit softening. Especially, the 1-MCP-treated of ‘Green Ball’ fruits significantly delayed fresh weight loss and reduction of soluble solids content. And fruit peel color alterations (L*, a*, and b*) were more variables in sunny-side than background regions in all cultivars. For cell wall metabolism, slower degradation of cell wall components (water-soluble pectin, CDTA-soluble pectin, sodium carbonate-soluble pectin, hemicellulose, and cellulose) was also observed in 1-MCP-treated fruits in comparison to the control fruits. Similarly, the enzymatic activities (of polygalacturonase, pectin methylesterase, cellulase, β-galactosidase, and α-L-arabinofuranosidase) that cause cell wall degradation were relatively lower in 1-MCP-treated fruits than in the control fruits for all cultivars. The solubilization of uronic acid content and its molecular weight was highly inhibited in ‘Green Ball’, less affected in ’Summer King’ and ‘Hwangok’, and not affected in ‘Summer Prince’ by 1-MCP. But, 1-MCP was not affected on total sugar content in cell wall. The levels of non-cellulosic neutral sugar compositions (rhamnose, arabinose, xylose, mannose, galactose, and glucose) in middle lamella of cell wall were delayed by 1-MCP, but these effects were differed by soluble fraction and each cultivar. Furthermore, the transcriptional analysis of the cell wall-related genes (MdPG1, MdPME1, Mdβ-GAL1, Mdβ-GAL2, and Mdα-ARF2) encoding the enzymes was further proved that the expression levels of these genes were higher in the control fruits than in the 1-MCP-treated fruits. Overall, the results suggested that the usage of 1-MCP is useful to delay fruit softening of four apple cultivars during cold storage by delaying the degradation of cell wall components, enzymatic activities of cell wall hydrolysis and low expression levels of cell wall-related genes.
