However, modern consumers, especially in developed countries, are reluctant to eat the edible insects majorly due to food neophobia. It also encountered some limitations because of their different functionality such as poor crosslinking properties from conventional animal-origin proteins. Therefore, it is important to improve the protein quality and to modify functionality of edible insect to increase utilization. In a series of study, the optimum extraction condition of edible insect proteins was investigated. Then, the interactions of enzyme and myofiblillar proteins with the extracted proteins were investigated to improve the protein functionality of the edible insect as a novel food ingredient. Firstly, quality changes of protein during extraction from edible insects are crucial for food application. Therefore, this study was conducted to investigate the changes of amino acid composition and protein physicochemical functionality extracted from three different edible insects (Tenebrio molitor, Allomyrina dichotoma, Protaetia brevitarsis). Three steps used for protein extraction were: grounding, de-fatting, and extraction. Essential amino acid index was increased undergone protein extraction steps. A. dichotoma had the best value at all steps and P. brevitarsis was followed. pH values of the extract increased with each step except for T. molitor. Further, Lightness, redness, and yellowness decreased generally. Protein molecular weight differed according to species and P. brevitarsis had the widest protein bands after extracting. Through extraction steps, foaming capacity and foam stability were improved and emulsifying capacity and emulsion stability did not improve dramatically. In conclusion, P. brevitarsis was the most suitable edible insect to use for protein sources and protein extraction can be useful way to improve protein physicochemical functionality of edible insects. Secondly, the effect of aqueous fat separation and defatting using organic solvents (99% of methanol, ethanol, and n-hexane) on the characteristics and functionality of proteins extracted from P. brevitarsis was investigated. The defatting efficiency, amino acid composition, protein solubility, and physicochemical properties were the highest when proteins were defatted using n-hexane. Proteins defatted using ethanol were similar in foaming capacity and emulsifying capacity. Surface hydrophobicity decreased when using organic solvents, and excessive fat content disrupted the functional properties of the extracted proteins. Proteins extracted using the different solvents displayed different pH values. The pH of the aqueous extract was the lowest. CIE L*a*b* color values also differed with the use of different extraction methods. Although n-hexane might be the most efficient solvent for defatting the proteins extracted from edible insects, ethanol could also be used to obtain similar foam and emulsifying capacities. Thirdly, the effect of pH and NaCl on protein quality and physicochemical functionality of P. brevitarsis was investigated. pH values of extracted protein were increased as pH of solvent and NaCl concentration increased. Foaming capacity was the highest at 0.5 M and 1 M NaCl and their stability was low. Emulsifying capacity and emulsion stability was increased away from pH 4. Although increase of NaCl concentration interrupted emulsion formation, their emulsion stability was higher than low salt concentration. Therefore, protein of P. brevitarsis had high functionality extracted at alkaline condition and high concentration of NaCl. However, NaCl could be not proper to form emulsion phase. Fourthly, the functional properties of protein solutions extracted from P. brevitarsis was improved by use of transglutaminase (TG) as a cross-linking agent. After various incubation times (10, 20, 30, 60, and 90 min) with TG, the protein solutions were assessed with regard to their amino acid composition, protein nutritional quality, pH, color (yellowness), molecular weight distribution, thermal stability, foam ability (capacity and stability), and emulsion ability (capacity and stability). Incubation with TG changed the amino acid composition of the proteins and shifted the molecular weight distribution towards higher values, while improving the rest of the aforementioned properties. Since the incubation time for 90 min decreased the protein functionality, the optimum incubation time for cross-linking protein with TG will be 60 min. This study was conducted to estimate the effect of the partial replacement of porcine myofibril protein (PP) with protein extracted from P. brevitarsis (IP) in protein-containing solutions and gels. The protein concentration was regulated at 30% and 1% for gels and solutions, respectively; replacement percentages increased by 20%. Gels containing IP showed a lower viscosity and thermal stability; the texture and shape of the gels were negatively impacted as the replacement percentage increased. Therefore, heating is not an appropriate method to induce the gelation of IP containing solutions. However, the replacement of PP with IP increased the foaming capacity and emulsifying capacity in solutions. In conclusion, this study showed that the partial replacement of PP with IP impacts positively on protein solutions but impairs their gelation. In conclusion, the improvement of functional properties of edible insect proteins and the replacement effect of pork protein by insect protein was verified.
