Laphet is a fermented green tea leaves in Myanmar, made from tea leaves (Camellia sinensis) under limited oxygen condition and it has been consumed over 100 years. This study aimed to investigate the microbial community, phytochemical composition, and prebiotic activity of Laphet. For these goals, Laphet samples were collected from different parts of Myanmar and their microbial diversities were analyzed by culture dependent and culture independent methods. Next, phytochemicals composition such as total polyphenol content, total flavonoid contents and antioxidant activities of Laphets were analyzed. Finally, prebiotic activity of Laphet was investigated by in vitro digestion and fermentation experiments. In microbiota analysis of Laphet, a combination of culture-dependent and culture-independent analyses revealed diverse bacterial and fungal population structures in 14 Laphet products. Amplicon-based sequencing analysis showed Lactobacillus and Acetobacter were the most abundant bacterial genera and Candida, Pichia, Cyberlindnera, Dipodascus, and Debaryomyces were common yeasts. In culture dependent analysis, eight lactic acid bacteria (LAB) and nine yeasts were isolated, and the dominant isolates were Lb. plantraum and Lb. collinoides in bacteria and Pichia manshurica, Candida boidinii, and Cyberlindnera jadinii in yeast. Most LAB and two yeast strains belonging to Debaryomyces hansenii exhibited positive tannase enzyme activity. In phytochemicals analysis, most of the Laphet samples showed considerable amount of total polyphenol, total flavonoid contents and antioxidant activities. In HPLC analysis, Laphet sample contain higher flavonols and lower flavon-3-ols content compared to raw tea leaves and other processed teas. Ethanol and acetate were detected in highest concentrations in Laphet together with low level of other organic acids. In prebiotic effect analysis of Laphet, Laphet sample was firstly subjected to in vitro digestion analysis. Then, in vitro digested Laphet sample was fractionated into polyphenol (P), polysaccharide and fiber (F) and mixture of P and F and those three fractions were used as substrate for in vitro fermentation analysis.Total polyphenol and flavonoid contents were relatively stable through the in vitro digestion but antioxidant activity greatly decreased during intestinal phases. At genus level, Bacteroidetes and Phascolarctobacterium compositions were increased by polysaccharide of Laphet after in vitro fermentation. Clostridium sensu stricto 1, Fusobacterium, Escherichia-Shigella were found to be suppressed by polyphenol of Laphet. After 24 hr, all samples tested showed higher content of acetate than control. Butyrate and propionate productions were favored by polysaccharide of Laphet. Polyphenol of Laphet accelerated the formate and succinate production. Ethanol content was increased after fermentation and the contents of samples tested were higher than control. Taken together, Laphet could be regarded as a health promoting fermented food as it has unique microbiota with beneficial bacteria, contains health promoting phytochemicals, and shows prebiotic properties.
