Although Mongolian shilajit has been documented to be effective against anti-Alzheimer’s disease (AD), there is a lack of proof. In addition, the identification of origin plant and the structure elucidation of the compounds is insufficient, and there is no quality control standard. In this study, the anti-AD-related activities of Mongolian shilait extract and fractions (Fr.) were examined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging, acetylcholinesterase (AChE), butyrylcholinesterase (BChE), β-site amyloid precursor protein-cleaving enzyme 1 (BACE1), and advanced glycation end-product (AGE) formation assays. The results showed that AD inhibitory activity was identified in the n-butanol (n-BuOH) Fr., so we performed to isolate the active compounds from n-BuOH Fr. by bioassay-guided isolation. Through proton (1H)- and The carbon (13C)-nuclear magnetic resonance (NMR), electrospray ionization (ESI)/hybrid linear trap-quadruple (LTQ)-orbitrap-high-resolution mass spectrometry (HRMS), compounds of bioassay-guided isolation identified syringaldehyde (1), benzoic acid (2), phthalic acid (3), hippuric acid (4), urolithin B (5), and (+)-usnic acid (6). They evaluated inhibitory effects on anti-AD assay. Hippuric acid (4) showed AChE and BChE inhibitory activity. Syringaldehyde (1) showed inhibitory effects on BACE1 activity and AGE formation. Syringaldehyde (1), phthalic acid (3), and urolithin B (5) showed a better effect than aminoguanidine (AG) hydrochloride, a typical AGE formation inhibitor.
