Human norovirus (HuNoV) is a major cause of acute gastroenteritis and is highly contagious. The virus is transmitted through contaminated food, water and person to person via contact with infected people. The main symptoms include acute diarrhea and vomiting, particularly resulting in dehydration in young children and elderly people. Studies regarding norovirus such as its life cycle, control, vaccination and medicine have mostly used surrogate cell lines such as feline calicivirus and murine norovirus due to lack of cell lines or small animal models. Firstly, in this study, we have developed a small animal model, tried to distinguish between active and inactive forms of HuNoV, and also created a method to replicate HuNoV in zebrafish using an integrated transcriptomic and proteomic approach. In total 28,355 transcripts and 233 proteins were expressed in HuNoV-injected zebrafish, and sixteen biomarkers were identified as cytokines, transmembrane transporters and chaperones. We found that HuNoV replicated in juvenile zebrafish 30 days post-hatching that were reared at 15°C for 10 days. Secondly, foodborne viruses were detected in feces resultant from acute diarrhea using a column made with concanavalin A (Con A)-linked polyacrylate. HuNoV (GI.2, GII.1, GII.4, GII.13), rotavirus and HAV were detected in the samples using the column with Con A-linked polyacrylate. Finally, we observed the efficacy of hand soap with included Canavalia gladiata extracts to remove HuNoV from hands. In these studies, we proved that zebrafish is an effective small animal model for HuNoV through the host response in adult zebrafish and the replication of HuNoV in juvenile zebrafish. We also developed a diagnostic method to rapidly and accurately detect foodborne viruses. Lastly, we developed and evaluated a new product that can remove norovirus from hands through human clinical trials.
