Synaptic plasticity was investigated by recording the pyramidal cells after placing stimulating electrodes in the schaffer collateral pathway in the cornu ammonis area 1 region of an acute hippocampal slice. LTP was induced by high-frequency stimulation (HFS), and then the slice was incubated for 30 min with NRG1-inducing and ErbB-blocking drugs. In addition, another form of synaptic plasticity depotentiation (DP) induced the HFS-stimulated LTP with theta-pulse stimulation within a few minutes following HFS stimulation. To investigate the memory differences between the WT and Tg groups, the novel object recognition test was used. The expression of NRG1/ErbB was evaluated by on Western blot analysis. PV + interneurons from the mouse hippocampal tissue were stained immunohistochemically. Statistical significance was determined using GraphPad Prism (GraphPad Software Inc.), and independent student's t-test for unpaired data (p <0.05).
Results: PV interneurons did not differ between the 6-month 5XFAD and WT groups, but decreased significantly in the 5XFAD 11- month 5XFAD and WT groups. When comparing between 5XFAD and littermate WT mice at 6- and 11 months, LTP for the fEPSP slope of 5XFAD mice decreased at 6 months, but reappeared at 11 months.
