ADH1C 유전자형은 비타민 A 제한 및 근내지방 침착과 관련이 있는 것으로 보고되었지만 비타민 A 추가 급여 및 근육 발달과 관련된 연구는 아직 미흡하다. 따라서 본 연구의 목적은 ADH1C 유전자형에 따른 비타민 A 추가급여가 근육 발달에 미치는 영향을 조사하는 것이다. 챕터 2에서는 in vitro 실험에서 비타민 A 처리가 근육 유래 SV 세포의 근원성 분화에 미치는 영향과 ADH1C 유전자형 간의 근원성 분화 능력의 차이를 조사했다. mRNA 발현의 결과를 따르면 성장 단계에서의 비타민 A 처리가 48 시간 후 MYF5의 발현을 유의적으로 (p < 0.05) 증가시켰으며 분화 6일 후의 근원성 mRNA (MYF5, MYOD, MYOG, MYOG)의 발현을 유의적으로 (p < 0.05) 증가시켰다. 마찬가지로, 분화 단계에서의 비타민 A 처리에 의해 분화 6일에서 근원성 mRNA (MYF5, MYOD, MYOG, MYF6)의 발현이 유의적으로 (p < 0.05) 증가되었다. 그러나 분화 6일에서 ADH1C TT타입과 TC타입의 세포 간 MYOG와 MYF6의 mRNA 발현에는 유의적인 차이가 보이지 않았다. 상기 실험결과들을 통하여 비타민 A 처리는 SV 세포의 근원성 분화를 촉진할 수 있으나, ADH1C 유전자형이 근육 분화에 미치는 영향은 in vivo 대사 상태와 함께 조사해야 할 필요가 있음을 확인하였다. 챕터 3에서는 비타민 A의 추가급여와 ADH1C 유전자형이 한우 이유 송아지의 근육 발달에 대한 효과를 조사하였다. 20 마리의 수컷 송아지 (90 일령)를 선택하여 비타민 A 추가급여 (기간: 3 개월; 대조군: 10,000 IU/kg of as-fed; 처리군: 40,000 IU/kg of as-fed) 및 ADH1C 유전자형 (대조군: TT 타입 5마리 및 TC 타입 5마리; 처리군: TT 타입 5마리 및 TC 타입 5마리)에 따라 무작위로 두 그룹으로 분리하였다. 그 결과, 비타민 A의 혈청 농도는 4 개월령과 6 개월령의 처리군에서 유의적으로 증가 (p < 0.05) 하였다. 그 중 TC 타입 보다 TT 타입 송아지의 혈청 비타민 A 농도가 더 높았다 (p < 0.05). 처리군의 6 개월령 혈청 트라이글리세라이드 및 비에스테르형지방산 농도는 대조군에 비해 유의적으로 (p < 0.05) 증가하였다. 그러나 체중, 일당 증체량 및 사료 섭취량은 유의적인 차이를 보이지 않았다. 또한 배최장근의 mRNA 발현 결과에 따르면 비타민 A 처리군에서 PAX7 이 증가되었다 (p < 0.05). 한편, 간에서의 ADH1C 및 ALDH1A1 mRNA 발현은 모두 비타민 A 추가급여에 의해 감소되었으나, TC 타입에서 TT 타입보다 높은 수준의 ADH1C mRNA 발현을 보였다. 상기 실험 결과들은 한우 이유 송아지에 대한 비타민 A 추가 급여는 근육에서 PAX7 mRNA 발현을 증가시켜 위성 세포의 저장을 촉진시킬 수 있으며 TC 타입 송아지의 비타민 A 대사 능력이 더 클 수 있음을 시사한다. 이러한 점을 미루어 보아, 본 실험은 비타민 A 추가급여가 근육 발달을 촉진시킬 수 있는 잠재력을 확인하였고, ADH1C 유전자형은 육량 증진을 위한 유전자 맞춤형 사양 관리에 사용될 수 있음을 밝혔다.

For beef production, the promotion of muscle development (hyperplasia of myofibers and hypertrophy of existing myofibers) is particularly critical in beef cattle. Although the alcohol dehydrogenase 1C (ADH1C) genotypes has been shown to be associated with vitamin A restriction and intramuscular fat, there have been no studies related to vitamin A supplementation and muscle development. Therefore, this work aimed to investigate the effects of vitamin A supplementation and ADH1C genotypes on muscle development in vitro and in vivo. In chapter 2, the effect of in vitro treatment with vitamin A on the myogenic differentiation of muscle-derived SV cells and the differences in myogenic differentiation ability between ADH1C genotypes were investigated. The results of mRNA expression showed that vitamin A treatment (1 μM) during the growth phase significantly increased (p < 0.05) the expression of MYF5 after 48 h and myogenic mRNA (MYF5, MYOD, MYOG, and MYF6) at D6 of differentiation. Similarly, treatment (0.1 μM) at the differentiation phase significantly up-regulated (p < 0.05) the expression of myogenic mRNA (MYF5, MYOD, MYOG, and MYF6) at D6 of differentiation. However, there was no significant difference in mRNA expression of MYOG and MYF6 between ADH1C TT type and TC type cells at the D6 of differentiation. Thus, it was confirmed that vitamin A treatment promotes myogenic differentiation of SV cells, whereas the effect of ADH1C genotype on muscle differentiation would need to be investigated together with metabolic status in vivo. In Chapter 3, the effects of in vivo supplementation with vitamin A and ADH1C genotypes on muscle development in weaned Korean native cattle were examined. Twenty male calves (90 days of age) were selected and randomly separated into two groups based on vitamin A treatment (duration: 3 months; control: 10,000 IU/kg of as-fed vs. treatment: 40,000 IU/kg of as-fed) and ADH1C genotype (control: 5 TT type and 5 TC type; treatment: 5 TT type and 5 TC type). As a result, serum concentration of vitamin A was significantly increased (p < 0.05) in the vitamin A treatment group at 4 and 6 months of age. TT type calves had greater serum concentration of vitamin A (p < 0.05) than the TC type calves. Serum triglyceride and non-esterified fatty acid concentrations increased (p < 0.05) in the treatment group compared to the control at 6 months of age. However, bodyweight, average daily gain and feed intake showed no differences between the groups. Besides, mRNA expression results of the longissimus dorsi muscle showed that PAX7 was increased (p < 0.05) under vitamin A treatment. Both ADH1C and ALDH1A1 mRNA expression were down-regulated in liver by vitamin A supplementation. Meanwhile, the mRNA expression of ADH1C in TC type was higher than that in TT type. Our findings suggest that vitamin A supplementation to weaned calves may promote the storage of satellite cells by elevating PAX7 mRNA in the muscle, and that of the TC type calves may have more metabolic capacity to vitamin A. In Summary, our results suggest that vitamin A supplementation (in vitro: 0.1 and 1 μM; in vivo: 40,000 IU/kg, as-fed) had the potential to promote muscle development and that the ADH1C genotypes can be used in genetic customized feeding management to maximize beef production.
