Allergic asthma is a chronic disease which characterized by inflammatory responses of the airways. Liriope platyphylla has a variety of biological properties, including anti-inflammatory, antioxidant and other medicinal effects. In this study, we aimed to evaluate the effects of Liriope platyphylla ethanol extract on ovalbumin induced allergic asthma, and further to examine its anti inflammatory effect in vivo and in vitro. Mice were sensitized and challenged with ovalbumin to build allergic asthma model in vivo. Bronchoalveolar lavage fluid (BALF) was collected for inflammatory cells analysis and left lobe lung tissues were examined for histopathological examination. The levels of IL-4, IL-5, IL-13, IgE, TNF-α, IFN-γ were determined by the respective ELISA kits. The NFκB signaling pathways were examined by western blot analysis. The results showed that Liriope platyphylla ethanol extract significantly ameliorated ovalbumin induced pathologic changes of lungs, such as decreasing massive inflammatory cell infiltration and mucus hypersecretion, and reduced the number of inflammatory cells in BALF. Liriope platyphylla ethanol extract also decreased the ovalbumin induced levels of IL-4, IL-5, IL-13, TNF-α and IgE. Furthermore, Liriope platyphylla ethanol extract inhibited ovalbumin induced phosphorylation of RelA(p65), IκBα in vivo. These anti-inflammatory effects of Liriope platyphylla ethanol extract has also been confirmed in LPS-stimulated RAW 264.7 macrophages in vitro. In conclusion, above results indicate that Liriope platyphylla ethanol extract exhibits a potent anti-inflammatory activity in allergic asthma through modulating NFκB signaling pathways, which would be used as a promising therapy agent for allergic asthma.
