To provide appropriate treatment and reduce the rates of transmission, viruses have to be diagnosed with sensitive and rapid techniques, such as reverse transcription polymerase chain reaction (RT-PCR). The present study aimed at developing a multiplex real–time RT-PCR (rRT-PCR) detection method targeting 14 respiratory viruses known to have high disease severity and infectivity, namely: influenza A and influenza B viruses, parainfluenza virus (PIV) 1, PIV2, PIV3, PIV4, human metapneumovirus (hMPV), adenovirus (ADV), human rhinovirus (HRV), respiratory syncytial virus (RSV), and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Thus, 14 different primer pair and probe sets were designed based on specific genes (with varying criteria). The method employed a portable PCR device capable of point-of-care testing (POCT) with a multiplex biochip to reduce the PCR test duration and reduce the amount of solution required per reaction by half. To confirm the detection limit of the primer pair and probe sets for each target virus, standard curves were constructed. rRT-PCR experiments were conducted using nucleic acid samples from nasopharyngeal swabs of positively diagnosed patients. The rRT-PCR method was capable of detection at 1.0 × 101 – 1.0 × 102 copy number/rxn of synthetic samples for all primer pair and probe sets, and could effectively detect most nucleic acid samples. This study demonstrated the potential for rapid and accurate respiratory virus detection using the POCT platform based multiplex rRT-PCR analysis method. The innovative assay method described in this study is expected to provide rapid, low-cost, early diagnosis of respiratory viruses.
