A novel Gram-negative black-pigmented obligate anaerobic rod-shaped bacterium was isolated from canine dental plaque. Because of the similar morphological characteristics, the strain was first assumed to be one of the most commonly isolated black-pigmented anaerobic periodontal pathogens in dogs, Porphyromonas gingivalis and Porphyromonas gulae. In addition to the similarity of colony morphology, the novel bacterium had shared cultural and biochemical characteristics including proteolytic activity and asaccharolytic properties. However, using comparative analyses of 16S rRNA gene sequences, it was unable to identify the novel isolate to the genus and species levels. Whole genome sequencing (WGS) was performed using the Illumina MiSeq and Oxford Nanopore MinION sequencing and the genome of the novel isolate had a size of 3.0 Mb with a DNA GC-content of 46.9%. Phylogenetic analyses using 120 bacterial orthologous genes revealed that the novel isolate represented a distinct phylogenetic lineage in the order Bacteroidales. On the basis of the low 16S rRNA sequence similarities (<86.5%) and distant relationships between the novel isolate and members of the order, the isolate is considered as a novel genus within the novel family in the order Bacteroidales. The genes involved in iron uptake and storage, proteolysis, hemolysis, type IX secretion system (T9SS) were found in genome annotation and these genes play an important role in the pathogenesis of P. gingivalis.
