Currently, plant-based meat analogue has attracted much attention, and various processed meat products including hamburger patties, ham, sausages, meatball, nuggets as well as minced beef or pork have been imitated using plant-based ingredients. Still major ingredients to produce the meat analogue products are limited to the texturized vegetable protein (TVP) mainly consisting of soy protein isolate (SPI) and wheat gluten. The limited plant protein sources manifest unattractive consumers’ preference, and it is necessary to investigate various potential ingredients from plant origin for improving sensorial satisfaction of meat analogue products. To manufacture meat analogue products with similar texture to actual meat products, main proteins and hydrocolloids were reformulated, and the roles of each ingredient were analyzed based on water binding properties and texture profile analysis (TPA) with meatball as control. In this study, SPI and mung bean protein isolate (MPI) were selected as the main protein sources for meat analogue. Based on gel electrophoresis, SPI was consisted of two factions indicated as glycinin (legumin fraction) and β-conglycinin (vicilin fraction) whereas MPI was composed of only vicilin globulin. For that reason, SPI exhibited two endothermic peaks at 82.5°C and 101°C which were evidenced by unfolding of conglycinin and glycinin, respectively. Alternately, MPI showed single peak at 96.3°C, and these thermal characteristics affected thermal gelling behavior. According to oscillatory shear rheology, both proteins showed similar changes in storage (G′) and loss (G″) moduli during double heating and cooling stage. However, SPI could form a very weak gel despite of lower denaturation temperature of β-conglycinin comparing to MPI. It was expected that SPI would limit texturization of meat analogue unless high thermal processing such as extrusion cooking was applied, and it could be a simple strategy to replace SPI to MPI for better texturization of the products under normal food cooking condition (~90°C). To find optimal SPI to MPI ratio, meat analogue containing total 27% protein was prepared using SPI alone or replacing MPI from 19 to 74%. The sample treated with SPI alone showed higher cohesiveness compared to the control (meatball), but the replacement of MPI showed an effect of lowering the cohesiveness, thereby improving textural imitate of the meatball. MPI also tended to increase the adhesiveness of meat analogue, but the change was not statistically significant comparing to the treatment prepared by SPI alone. Protein powder tended to increase hardness, adhesiveness, cohesiveness, and springiness as the amount of protein was increased, but there was no difference in cohesiveness and springiness in 27% and 31% protein contents. When replacing SPI with MPI, moisture content and cooking loss were not changed and expressible moisture was increased indicating that the water holding capacity was insufficient compare to SPI. However, when MPI was increased up to 20%, it still showed a lower value than that of meatballs. Five materials (potato starch, κ-carrageenan, methylcellulose, konjac, potato protein) were evaluated as additives for meat analogue. The additives decreased the expressible moisture comparing to non-additive treatment and methylcellulose and konjac showed lowest values among additives. In texture profile analysis, potato starch and κ-carrageenan were negatively evaluated as they acted to further strengthen the hardness of the protein powder. Methylcellulose was helpful by lowering the excessively high hardness and springiness of the protein and increasing the insufficient tackiness. Konjac showed the greatest value in decreasing springiness and increasing adhesiveness while maintaining hardness. When methylcellulose and konjac were mixed, it showed a positive effect of raising the adhesiveness to a level similar to that of meatballs. Potato protein decreased cohesiveness and springiness using alone and when it was mixed with methylcellulose and konjac in combination, contributed to improving the textural of meat analogue.
