This study was carried out to develop a new and environmental-friendly bio-fungicide to control powdery mildew using Rhus javanica and Cedrela sinensis.
1. Establishment of optimal concentration test of control efficacy in Rhus javanica and Cedrela sinensis extracts In order to develop an environmental-friendly control agent against strawberry powdery mildew, a total of 30 medicinal crop extracts were tested for control of powdery mildew. As a result, seven kinds of crops including Rhus javanica, Artemisia iwayomogi, Artemisia annua, Cedrela sinensis, Reynoutia sachalinensis, Dendropanax morbifera, Helianthus tuberasus were selected. Among the selected crops, the control value of the two most effective R. javanica and C. sinensis was in the range of 50-70%, lower than that of general disinfectants. Therefore, the extracts of selected crops and potassium silicate which are reported to be effective in controlling powdery mildew, were mixed and sprayed. As a result, the control effect was increased to 86% of similar level to that of the disinfectant. The result of single treatment according to the potassium silicate concentration (0.3%, 0.5%, 0.7%) showed that control effect was the highest at 0.7% concentration, followed by 205%, 0.3% concentration. But the treatment of 0.7% concentration of potassium silicate was formed in white powder on the surface of leaf. So, the optimal concentration was set to 0.5%. To find out the control effect according to the concentration of medicinal crop extracts, 1,000, 5,000 and 10,000 times were mixed with 0.5% potassium silicate and sprayed. The control effect was best at extracts 1,000 times with 0.5% potassium silicate, so this was set as the final concentration of natural control agents. In order to determine the control effect according to the number of spraying, one treatment and two treatments were performed. As a result, when sprayed once, the control effect lasted for 13~14 days, when sprayed twice at an interval of 3~4 days, it lasted about 30 days, indicating that the control effect was superior to that of one treatment. Therefore, the control effect of 0.5% potassium silicate and R. javanica and C. sinensis extract (1,000 times) was more than 75% when spraying twice at 3~4 days intervals, and the control period was more than 30 days. It is considered suitable as an eco-friendly control agent against strawberry powdery mildew.
2. Analysis of the effective ingredients of the control agent using Rhus javanica and Cedrela sinensis. LC-MS/MS analysis was conducted to investigate the components of R. javanica and C. sinensis having an inhibitory effect on powdery mildew. As a result of the main components of the natural extract, ellagic acid was the highest at 3.03mg/g and 3.24mg/g in R. javanica and C. sinensis, respectively. Quercetin-3-O-glucoside and quercetin contained in the range of about 0.56mg/g~0.60mg/g in both R. javanica and C. sinensis, and kaempferol contained in the range of 0.13mg/g~0.19mg/g, and naringenin contained in the range of about 0.001mg/g~0.003mg/g. SEM and EDS analysis were performed to investigate the amount of control agent remaining on the surface of strawberry leaves. The result shows that potassium silicate spraying was shown to inhibit the occurrence of powdery mildew by forming a physical film on the strawberry leaves, and it was found that the effect gradually disappeared as the concentration of potassium silicate decreased as time passed after spraying. Therefore, mixed spraying of medicinal crop extracts and potassium silicate is presumed to have a synergistic effect against strawberry powdery mildew with the formation of a physical film of potassium silicate and the antibacterial components of the medicinal crop extracts.
