The specific molecular mechanisms by which the dopaminergic neurons are injured remain to be elucidated and thus there has been no cure till now. It has been shown that BDNF is a key neurotrophic factor that restores the dopaminergic neurons in the variant PD models. Therefore, BDNF induction is a decent therapeutic strategy to treat PD. Simvastatin has been used to prevent or treat cardiovascular diseases by lowering cholesterol synthesis in the liver. Emerging evidence suggests that the drug owns a neuroprotective property attributable to its pleiotropic effects. This study aimed to assess the neuroprotective effect of simvastatin via inducing BDNF. Firstly, I tested its protective effects against MPP+ toxicity using SH-SY5Y neuroblastoma cells. I found that simvastatin rescued the cells from MPP+ toxicity at 10 nM. The neuroprotection seemed to come from the upregulation of BDNF as simvastatin increased BDNF and its downstream signaling molecules in the cells, and the protective action was significantly counteracted by BNDF siRNA or TrkB antagonist (K252a). Importantly, I found that simvastatin upregulated BDNF via the PPAR α activation. Silencing the PPAR α gene attenuated the simvastatin-induced BDNF upregulation and the cell viability upon exposure to MPP+. Next, I evaluated its neuroprotective effects in the PD animal model mediated by MPTP (15 mg/kg x 4 times with 1.5 hrs intervals, i.p.). I found that pretreatment with simvastatin for 2 weeks prevented the MPTP-caused loss of the nigrostriatal dopaminergic neurons as determined by immunohistochemical stainings for tyrosine hydroxylase. In parallel with the results, simvastatin attenuated the decrease in the striatal dopamine level and behavioral impairment elicited by the neurotoxicant. Consistent with the in vitro study, simvastatin upregulated BDNF expression and restored the BDNF level in the MPTP-treated brain. These findings suggest that simvastatin's neuroprotection may be linked to the upregulation of BDNF via activation of PPAR α and it could be a possible therapeutic intervention for the treatment of PD.
