Cytotoxicity and cell proliferation were evaluated to exclude the effect of dermokine on macrophages themselves, and various concentrations of recombinant dermokine were added to osteoclasts in osteoclastic medium containing RANKL and M-CSF and differentiated into osteoclasts. To evaluate the molecular biologically effect of dermokine on osteoclasts, mRNA expression of NFATc-1, TRAP, cathepsin K, DC-stamp, and c-Fos, known as differentiation markers, was confirmed through RT-PCR, and then phosphorylation of ERK, JNK, and p38 was evaluated with western blots to find a specific pathway.
