PUE의 단회 경구독성시험에서 LD50 cut-off가 2,000mg/kg로 GHS Category 5로 안전한 물질로 확인되었다. 조직병리학적 소견에서 정상대조군(CMC군)은 위 점막의 조직 형태는 정상 소견이었다. 상피세포는 정상적인 형태로 염증세포 침윤이 없었다. 음성대조군 (Injured group)에서 위 점막은 염증세포 침윤, 고도의 퇴행성 세포, 세포질 점액, 점막위축이 관찰되었다. Lansoprazole 투여한 양성대조군 (LPZ) 그리고 PUE를 투여한 저용량 (PUE250) 및 고용량 (PUE500) 투여군은 경증 혹은 중등도의 세포 변성 및 염증을 보였다. PUE250과 PUE500 투여군이 Injured군보다 혈장 PGE2의 수준이 높았다. cAMP 수준은 CMC군, LPZ군, PUE250 군, PUE500군이 NC군보다 작았다. 위의 결과를 종합하면 상피 세포 내의 프로스타글란딘 (PGE2) 경로를 통한 점액 및 중탄산염 분비를 증가시켜 1차 및 2차 위 점막 완전성을 향상시키고 H+/K+ ATPase 펌프 억제를 통해 정수리 세포로부터 산 분비를 감소시켰다. 항지방간 효과를 보기 위하여, 3T3-L1 세포에서 FPUE 및 PUE에 대한 지질 축적시험을 실시하였다. FPUE 및 PUE는 3T3-L1 세포에서 지질 축적을 억제하였다. 고지방식을 섭취하는 마우스에서 FPUE 및 PUE의 투여는 체내 지질 농도의 수준, 간 손상 및 내분비펩티드를 정상 수준으로 유지시켰다. 마우스에서 FPUE의 투여는 PPARγ, C/EBP-α, SREBP-1c, aP2, ACC1 및 GLUT4의 유전자 발현을 정상 수준으로 유지시켰다. FPUE의 투여는 장내 미생물총의 변환 활성을 통하여 건강한 미생물총으로 유지시키고 Firmicutes의 감소와 지질 수준을 감소시켰다. 마우스의 알코올성 지방간 질환 모델에서 FPUE는 간에서 항산화, 항염증 활성 및 위장 영역에서 미생물총 조절 효과를 통해 간 손상을 보호하는 것으로 나타났다. Pyrus ussuriensis Maxim has been cultivated all over the world, including Korea. It is a fruit that contains many nutrients important to health. In previous studies, PUE was known to be effective in the prevention and treatment of various systemic and chronic diseases. In this study, based on the reported evidence, Pyrus ussuriensis Maxim extract (PUE) and Fermented Pyrus ussuriensis Maxim extract (FPUE) with lactic acid bacteria were prepared for three kinds of chronic diseases (gastritis, obesity and fatty liver). The present study was conducted to investigate the preventive and therapeutic effects. The result are as follows: Before the anti-gastritis effect evaluation, GC/MS analysis of 50% ethanol PUE was performed. As a result, the main components were 3,5-dimethyl-1H-pyrazole, 4H-pyran-2,3-dihydro-3,5-dihydroxy-6-methyl, 5-hydroxymethylfurfural, 1,5-anhydro-arabino-furanose, beta-D-Glucopyranose, 4-O-beta-D-galactopyranosyl, Thymol, Hexitol, D-Mannitol and vitamin E. In a single oral toxicity test of PUE, the LD50 cut-off was 2,000 mg/kg, and it was confirmed as a safe substance in GHS Category 5. In histopathological findings, in the normal control group (Uninjured group), the tissue morphology of the gastric mucosa was normal. Epithelial cells were normal and there was no inflammatory cell infiltration. In the negative control (Injured group), inflammatory cell infiltration, highly degenerative cells, cytoplasmic mucus, and mucosal atrophy were observed in the gastric mucosa. The positive control group (LPZ) administered with lansoprazole and the low-dose (PUE250) and high-dose (PUE500) groups administered with PUE showed mild or moderate cell degeneration and inflammation. Plasma PGE2 levels were higher in the PUE250 and PUE500 groups than in the injured group. The cAMP level was smaller in the Uninjured, LPZ, PUE250, and PUE500 group than in the injure group. Summarizing the above results, it increased primary and secondary gastric mucosal integrity by increasing secretion of mucus and bicarbonate through the prostaglandin (PGE2) pathway in epithelial cells, and decreased acid secretion from parietal cells through inhibition of H+/K+ ATPase pumps. To determine the lipolytic effects, a lipid accumulation test was performed for FPUE and PUE in 3T3-L1 cells. FPUE and PUE inhibited lipid accumulation in 3T3-L1 cells. Administration of FPUE and PUE in rats fed with a high-fat diet maintained normal levels of body lipid concentrations, liver damage and endocrine peptides. Administration of FPUE in rats maintained the gene expression of PPARγ, C/EBP-α, SREBP-1c, aP2, ACC1 and GLUT4 at normal levels. Administration of FPUE maintained a healthy microflora through the modulation activity of the intestinal microflora, and decreased the number of Firmicutes and lipid levels. In the ethanol induced fatty liver mice model, FPUE was shown to protect against liver damage through antioxidant, anti-inflammatory activity in the liver and microbiota-modulating effects in the gastrointestinal tract. As a result of analyzing the components of FPUE by LC/MS-MS, chlorogenic acid was identified as the main material before fermentation, and beta-hydroxyphenyl-lactic acid, dl-beta-Phenyllactic acid, and beta-indole lactic acid after fermentation. As a result of GC/MS analysis of FPUE, the main components were acetic acid, (R, R)-Butane-2,3-diol, L-Lactic acid, Butyrolactone, 2-Pyrrolidinone, and Tetramethyl-Pyrazine. In the alcohol-treated group, slight balloon degeneration and a large amount of fat accumulation were observed. The FPUE administration group was more effective in preventing fatty liver than the PUE administration group. Aspartate Aminotransferase (AST), Alanine Aminotransferase (ALT) and Triglyceride (TG) activities were dose-dependently decreased in the FPUE-administered group. In gene expression studies, alcohol dehydrogenation (ADH and ALDH), antioxidant enzyme activity (SOD) and the expression of related genes were upregulated in the FPUE group compared to the NC group, but not significantly for ADH and SOD. The FPUE-administered group showed a clear change in the composition of the gut microbiota than the NC group. This is consistent with the PCoA results, although normal microflora was disturbed by ethanol intake, but returned to normal by FPUE administration. Taking all the test results, Non-fermented Pyrus ussuriensis extract (PUE) and Fermented Pyrus ussuriensis extract (FPUE) fermented with lactic acid bacteria were effective in preventing and treating three types of chronic diseases (gastritis, obesity and fatty liver). In particular, PUE was effective in gastritis, and FPUE was effective in preventing and treating high fat-diet induced obesity and alcoholic fatty liver.
