이와 같은 마이크로파에 의한 변성전분이나 차아염소산나트륨에 의한 산화전분에 관한 연구가 진행되고는 있으나 아직 미진한 편이며 기름골 전분에 관한 연구는 거의 없는 실정이다. 따라서 본 연구는 기름골 전분이 마이크로파에 의해 가열된 물리적 변성전분과 차아염소산나트륨에 의해 산화된 화학적 변성전분의 이화학적 특성과 호화특성에 대하여 조사하였다.
제안 방법
기름골 산화전분은 Konoo 등의 방법(15)을 일부 변형하여 제조하였다. 즉 35% 전분현탁액을 교반기가 부착된 반응관에 넣고 1 N NaOH를 가하여 pH를 10.
기름골 전분의 미세구조를 조사하기 위해서 알카리 침지법에 의하여 추출한 전분을 전자주사 현미경 (scanning electron microscope, S-2380N, Hitachi Ltd, Japan)으로 500배와 4000배로 확대하여 관찰하였다.
기름골의 물리적 변성전분의 팽윤력 (Swelling power)과 용해도(Solubility)는 Schoch의 방법 (21)을 변형하여 측정하였고, 산화전분은 Leach 등(22)의 방법을 변형하여 55° C에서 차아염소산나트륨의 농도별로 측정하였다
알카리 침지법에 의해 제조한 기름골 전분을 마이크로파로 가열하여 물리적 변성전분을 제조하였다. 마이크로파 가열은 200 mL 비이커 내부를 여지로 둘러싸고, 전분 100 g을 넣어 여지로 덮은 뒤 polyethylene film으로 밀봉하여 전자레인지 (Samsung, RE-IH700)의 중앙에 위치하도록 한 다음 60초, 120초, 180초 동안 각각 가열하여 방냉한 후, 80 mesh 체를 통과시키고 각 시료를 데시케이터에 보관하면서 실험에 사용하였다.
전분현탁액이 40°C가 되면 차아염소산나트륨을 각각 1.5%, 3%, 6%(15, 30, 60 mg/g starch)을 첨가하면서 3시간 반응시킨 후 7% HCl로 중화하였다(pH 5.0±0.2)
즉 50°C에서 1분간 유지한 다음 95° C까지 1분당 12°C로 가열하고 95°C에서 2.5분간 유지시킨 다음 1분당 12°C/min의 속도로 냉각시켜 2분간 유지시키면서 점도를 측정하여 호화 특성을 조사하였다.
대상 데이터
본 실험에 사용된 기름골(Cyperus esculentus L. var sativus Boeck)은 국내에서 재배, 생산된 것을 수확한 후괴경을 수세, 건조하여 시료로 사용하였다.
알카리 침지법에 의해 제조한 기름골 전분을 마이크로파로 가열하여 물리적 변성전분을 제조하였다. 마이크로파 가열은 200 mL 비이커 내부를 여지로 둘러싸고, 전분 100 g을 넣어 여지로 덮은 뒤 polyethylene film으로 밀봉하여 전자레인지 (Samsung, RE-IH700)의 중앙에 위치하도록 한 다음 60초, 120초, 180초 동안 각각 가열하여 방냉한 후, 80 mesh 체를 통과시키고 각 시료를 데시케이터에 보관하면서 실험에 사용하였다.
이론/모형
기름골 전분의 RVA 측정은 Bason 등(23)과 Ross 등의 방법(24)에 의하여 기름골 전분 3 g을 증류수 25 조에 분산시키고, 신속점도측정계 (RVA, Rapid Visco Analyzer, Model 3D, Newport Scientific PTY., Ltd, Australia)를 이용하여 측정하였다. 즉 50°C에서 1분간 유지한 다음 95° C까지 1분당 12°C로 가열하고 95°C에서 2.

기름골 전분의 분리는 시료를 수세한 후 직접 실험용분쇄기를 이용하여 건식 분쇄한 다음 알카리 침지법(14)을 이용하여 분리하였다.
기름골의 물리적 및 화학적 변성전분의 일반성분(수분, 회분, 조지방, 조단백질)은 AOAC법 (16)에 따라 정량하였다. 기름골 전분의 색도측정은 색차계(color and color diffenaice meter, Model CR-300, Minolta, Japan)를 이용하여 L, a, b 값을 측정하여 변화된 값을 비교하였는데 이때L, a, b값은 각각 96.

전분의 호화온도와 호화 엔탈피는 Donovan 등의 방법(25)에 따라 전분 4 mg을 aluminium sample pan에 취하고 전분의 2배량에 해당하는 증류수를 microsyringe로 가하여 밀봉한 후에 실온에서 4시간 방치한 다음 DSC(Differential Scanning Calorimetry, TA 2000, USA)를 사용하여 10 & deg; C/min의 속도로 30°C부터 110°C까지 가열하여 흡열 peak를 얻었다
성능/효과
기름골 변성전분을 DSC를 이용하여 측정한 호화개시온도, 최대호화온도, 호화종결온도 및 호화엔탈피는 Table 6과 같다. DSC 측정 결과 기름골 변성전분은 일정한 엔탈피를 갖는 1개의 흡열반응 곡선을 보였다. 마이크로파에 의한 변성전분의 DSC 결과 가열시간이 증가함에 따라 호화개시 온도는 낮아지고 호화엔탈피는 점차 증가하였다.
기름골 전분의 팽윤력과 용해도는 마이크로파에 의한 변성전분의 경우 생전분에 비하여 60°C 이하에서는 가열시간이 길수록 약간 높았으나 60°C 이상에서는 약간 감소하였다
일반적으로 용해도와 팽윤력은 전분 입자내의 미셀 구조의 결정성과 밀접한 관련이 있으며, 아밀로오스와 아밀로펙틴 함량, 전분의 열처리 방법, 전분의 지방산의 결합 정도 및 전분의 이화학적 변성 등에 영향을 받으며 산화전분은 일정온도 이상에서는 팽윤 없이 용해된다(37,38). 따라서 기름골 전분은 차아염소산나트륨에 의해 전분의 C-C결합이 분해되는 동시에 전분의 미셀구조가 파괴된 것으로 생각된다.
전분의 점도는 가용성 아밀로오스 함량, 결합된 지질 함량, 입자의 팽윤정도 팽윤된 입자의 열이나 전단력에 의한 저항정도에 영향을 받는데(40) 마이크로파 가열에 의해 나타난 위의 결과는 팽윤력의 감소, amylose-lipid complex 형성 등의 요인에 의한 것으로 보여진다(33). 또 산화전분의 경우에는 최고점도가 267-129 RVU로 나타나 물리적 변성전분과는 달리생전분에 비해 낮았으며 setback 및 breakdown도 현저하게 낮았다.
기름골 변성전분을 신속점도계를 이용하여 측정한 호화특성으로부터 얻은 각 측정치는 Table 5에 나타내었다. 마이크로파에 의한 물리적 변성전분은 생전분에 비해 호화개시 온도가 약간 높았고 최고점도는 416-488 RVU로 매우 높았다. 노화의 경향을 반영하는 setback은 마이크로파 가열시간이 길어짐에 따라 증가하는 경향을 보였는데Uchijima와 Kurihaia(39)도 마이크로파 가열에 의해 쌀과고구마 전분의 노화가 촉진된다고 하였다.
마이크로파에 의한 변성전분의 DSC 결과 가열시간이 증가함에 따라 호화개시 온도는 낮아지고 호화엔탈피는 점차 증가하였다
기름골 변성전분의 물 결합능력은 Table 4에 나타내었다. 물리적 변성 전분의 경우 마이크로파 처리 시간에 따라 물결합능력이 점차 증가하여 마이크로파 1분 처리시 86.13%, 2분 처리시 89%, 3분 처리시에는 91.46%로 증가하였다. 일반적으로 전분에 결합된 물은 전분 입자 및 전분 표면에 흡착되는 것으로 전분 입자의 치밀도와 밀접한 관련이 있으며 (21) 밀가루(34), 고구마 가루(35)의 경우에도 마이크로파 가열에 의하여 물 결합능력이 증가한다고 보고되었다.
기름골 변성전분의 일반성분은 Table 1과 같다. 물리적 변성전분의 마이크로파 가열시간에 따른 수분함량의 변화는 가열시간이 길어짐에 따라 기름골 전분의 수분 함량이 점차 감소하여 180초 동안 마이크로파 가열을 하였을 때기름골 전분의 수분함량은 3.39%까지 떨어졌다. 일반적으로 마이크로파에 의한 전분의 온도와 수분함량 차이는 시료의 중량, 재질, 형상, 크기 뿐만 아니라(10) 마이크로파의 에너지 및 수분함량에 기여하는 것으로 알려져 있다(26).
3과 같다. 산화전분의 경우에는 차아염소산나트륨을 1.5% 처리한 경우와 3% 처리한 시료는 큰 차이를 보이지 않았으나 처리량을 6%로 증가시켰을 경우 용해도와 팽윤력이 급격히 증가하는 경향을 보였다. 일반적으로 용해도와 팽윤력은 전분 입자내의 미셀 구조의 결정성과 밀접한 관련이 있으며, 아밀로오스와 아밀로펙틴 함량, 전분의 열처리 방법, 전분의 지방산의 결합 정도 및 전분의 이화학적 변성 등에 영향을 받으며 산화전분은 일정온도 이상에서는 팽윤 없이 용해된다(37,38).
마이크로파 처리한 기름골 전분은생전분과 같이 둥근형 또는 타원형으로 나타났다. 생전분에 비해 마이크로파 처리한 전분의 표면에 약간의 변형이 관찰되었으나 마이크로파 가열방법과 가열시간에 따른 기름골 전분의 외관상 형태변화는 크지 않았다. 한편 차아염소산나트륨으로 처리한 기름골 산화전분은 치환도가 증가할수록 매끄러운 입자표면이 분해되었으나 원래의 기름골생전분의 입자 모양을 거의 그대로 유지하고 있는 것으로 보아 산화에 의한 전분입자의 변화는 매우 작다고 생각된다.
이는 찰옥수수 산화전분의 이화학적 특성을 연구한 Chung 등(31)의 연구와 비슷한 결과였다. 이와 같은 결과로 보아 차아염소산나트륨이 기름골 전분과 반응하는 동안 전분의 백도값에 영향을 미침을 알 수 있었다.
생전분에 비해 마이크로파 처리한 전분의 표면에 약간의 변형이 관찰되었으나 마이크로파 가열방법과 가열시간에 따른 기름골 전분의 외관상 형태변화는 크지 않았다. 한편 차아염소산나트륨으로 처리한 기름골 산화전분은 치환도가 증가할수록 매끄러운 입자표면이 분해되었으나 원래의 기름골생전분의 입자 모양을 거의 그대로 유지하고 있는 것으로 보아 산화에 의한 전분입자의 변화는 매우 작다고 생각된다.
