제안 방법
추출과 농축을 위하여 Sep-Pak C18 카트리지를 이용하여 시료를 용출시킨 후 이 용액을 다시 질소농축기로 농축하였다. 메탄올 500 μ1로잔사를 녹여 0.22 μm syringe filter를 가지고 여과시 킨 후 2 ml 갈색 vial에 옮겨 LC/ESI-MS/MS로 분석하였다.
이러한 결과의 차이는 실험에 사용된 과실의 품종, 숙성정도, 분석물질의 추출 및 정제 공정, 분석시료의 전처리 과정, 분석 방법 및 기기 감도 등 다양한 원인에 의해 나타나는 것으로 생각된다. 본 연구에 사용한 석류추출물은 과육 중의 에스트로겐 성분을 최대화하기 위하여 주정을 이용한 정제과정을 거쳤으며, 분석 시에 산가수분해와 Sep-pak 여과 과정을 거쳐 좀 더 간편한 분석법을 시도하였다. 기존의 에스트로겐 분석방법 중 GC-MS를 이용한 방법은 hydroxyl기를 유도체화 해야 하는 번거로움이 있었으며, LC를 이용한 분석방법의 경우 물질의 정성분석이 불가능하여 정확한 분석이 불가능하였다.
본 연구에 사용된 LC/MS는 시료 자동주입기(Agilent 1200 series G1313A, Autosampler) 가 장착된 Agilent 사 (Palo Alto, CA, US A) 의 Agilent 1200 series HPLC를 사용하였으며, 분리된 각 물질의 분자량 확인을 위해 Agilent 6410 Triple-quadrupole tandem LC/MS/MS (Agilent Technologies, Palo Alto, CA,USA)를 사용하였다. 에스트로겐 분석을 위해 시료는 추출, 정제, 농축하였으며, LC/MS/MS ESI negative mode에서 Extend C18 컬럼을 이용하여 분석을 실시하였으며 분석조건은 Table 1과 같다.
