4.43, 4.57, 5.24, 6.17, 6.55, 7.41, 7.57, 8.10분의 각 peak가 mass peak(m/z)와 상대면적비(relative area)로 보아 각각 α-pinene이 21.55%, camphene이 6.36%, β-pinene이 9.32 %, myrcene이 0.46%, limonene이 13.05%, terpinolene이 8.39%, α-terpineol이 1.07%, bornyl acetate이 5.01%, α-copaene이 2.05%, caryophellene이 11.43%, humulene이 4.35%로 주요 구성성분이라고 추정되었다(Fig. 1A and 1B).
DPPH를 이용하여 essential oil의 항산화작용을 DPPH 라디칼 변화율을 측정한 결과, 소나무 잎과 가지 정유는 1000 µg/ml 농도에서 대조군에 비교하여 각각 71.1±4.36(p<0.01) 및 67.6±4.95%(p<0.01)를 보여 유의성있는 DPPH 라디칼 소거작용이 있음을 나타내었다(Fig. 4).
Elastase는 control에 비하여 112.3%의 변화율을 보였고 소나무 잎과 가지는 100 µg/ml에서 각각 106.1±4.6(p<0.01), 101.3±1.0(p<0.01)%의 변화율로 억제하는 경향을 보였다(Fig. 8).
Pine twigs(PT)와 위 5성분들은 대조약물인 Vitamin C가 100 µM에서 31.7±11.0% (p<0.01)의 억제작용을 보이는 것보다 hyaluronic acid가 가수분해되는 것을 더 강하게 억제하였다(Table I, II).
구성성분 중 myrcene과 phellandrene은 100 µM 에서 hydroxyl radical 생성이 각각 76.1±11.1, 74.2±7.5%로 억 제되었고, silica에 의한 세포내 hydroxyl radical 생성을 농도의존적으로 억제하였다.
구성성분 중 terpinolene은 10 µM에서 83.7±4.5%, α-pinene, myrcene, phellandrene은 100 µM에서 각각 92.5±6.9, 80.1±2.2, 76.6±7.9%로서 유의성있는 억제작용이 있었다(Fig. 11).
구성성분들 중 α-pinene은 10 µM에서 40.2±9.2%(p<0.01), β-pinene과 myrcene은 1 µM에서 각각 39.2±6.7(p<0.01), 39.4±12.3%(p<0.01), phellandrene과 terpinolene은 100 µM에서 각각 45.4±7.9(p<0.05), 40.3±7.7%(p<0.01)로 유의성있는 억제효과를 나타냈다.
대조약물인 Vitamin C 100 µM에서 103.4±1.9(p<0.05)%의 변화율과 비교하였을 때 거의 유사하게 elastase 활성을 억제하였다 (Fig. 8).
또, 소나무 잎과 가지의 정유는 RAW 264.7 세포에서 silica 자극에 의해 증가된 세포내 H2O2 생성과 hydroxyl radical 생성을 유의성있게 억제하였고, 그 약효성분은 β-pinene과 terpinolene, myrcene, phellandrene 등이었다.
또한 그의 성분들 12종류 중 phellandrene, terpinolene 및 myrcene이 1000 µM에서 각각 84.1±1.54(p<0.01), 87.5±0.99 및 95.6±2.56%로 유의성있는 항산화작용이 있었을 뿐 나머지 9종의 정유들은 1000 µM에서도 유의성있는 변화가 없었다(Fig. 5).
서로 불용인 혼합물 증기의 전 압력은 순성분 각각의 증기압의 합과 같으므로, 정유의 증기압과 물의 증기압의 합이 증류조 내의 압력과 같아지면, 정유는 수증기와 함께 비등하여 유출하는 steam distillation 방법으로 채취한 소나무의 잎과 가지를 음건 세절한 후 분쇄하여 100℃에서 6시간 동안 추출하여 잎 (needles) 4 kg에서 9ml, 가지(twigs) 1.7 kg에서 3ml의 각각 0.225%(v/w), 0.176%(v/w) yield의 essential oil을 얻었다.
소나무 가지 정유는 1, 10, 100 µg/ml에서 superoxide radical의 소거율이 각각 91.2±2.05, 91.3±3.41, 87.9±1.97%로서 유의성있는 농도의존적 항산화작용이 있었다(Fig. 6).
소나무 잎과 가지 정유와 그의 구성성분들의 silica에 의한 세포내 H2O2 생성을 관찰한 결과, silica 1 mg/ml과 RAW 264.7 세포를 배양하면 H2O2 생성을 121.4% 증가시켰으나, 소나무 잎과 가지 정유는 10 µg/ml에서 각각 84.1±6.1, 83.2±5.9%의 H2O2 생성을 보여주어 미약한 항산화작용이 있었다(Fig. 10).
소나무 잎과 가지 정유와 그의 구성성분들의 silica에 의한 세포내 Hydroxyl radical 생성을 관찰한 결과, silica 1 mg/ml은 RAW 264.7 세포에서 hydroxyl radical 생성을 539.3% 증가시켰으나, 소나무 잎과 줄기 정유는 10 µg/ml에서 hydroxyl radical 생성이 각각 89.8±1.7, 88.0±5.7%로서 유의성있는 억제효과가 있었다(Fig. 12).
