1. Extraction of protein-bound polysaccharide and identification of components An anticancer substances were prepared by extraction of hot water from the Lentinus edodes(fruiting body, mycelial), Pleurotus ostreatus(fruit body, mycelial & wasted mycelial). The yield were 0.9, 0.7, 2.2, 1.1% and 1.4%
1. Extraction of protein-bound polysaccharide and identification of components An anticancer substances were prepared by extraction of hot water from the Lentinus edodes(fruiting body, mycelial), Pleurotus ostreatus(fruit body, mycelial & wasted mycelial). The yield were 0.9, 0.7, 2.2, 1.1% and 1.4% respectively. The results of clonogenic assay showed that protein-bound polysaccharides from the Pleurotus ostreatus, Lentinus edodes & Cordyceps sinensis mycelial had anticancer activity against Raji above 10mg/㎖ concentration, and did not exhibit anticancer activity against cell lines such as SF-188, 3LL, KB and HEP-2 up to 10mg/㎖ concentration. This result suggests that protein-bound polysaccharides has specially anti-lymphoma cancerous effect. The optimum conditions for cultivation of Lentinus edodes mycelial & Cordyceps sinensis mycelial were elucidated to be incubation temperature of 25℃, initial pH 4.0 & 5.0, agitation of 300 & 200rpm, inoculation ratio of 10.0 & 1.0%(v/v), and aeration of 1.0 & 1.4 volume of air/volume of medium/minute in medium. At that time, yield of Lentinus edodes & Cordyceps sinensis mycelial was 18.5/L and 9.1g/L respectively. In addition, the amino acid analysis of the protein-bound polysaccharides from the Lentinus edodes fruit body and Lentinus edodes mycelial showed proline contents 22.61% and 22.67%, respectioely, the Pleurotus ostreatus fruit body & Cordyceps sinensis mycelial showed asparatic acid contents 18.34% and lysine 22.69%, respectively. At the mineral analysis, the major mimeral, protein-bound polysaccharides from the Lentinus edoses fruit body and Lintinus edodes mycelial were K, and Na. And the Pleurotus ostreatus fruit body & Cordyceps sinensis mycelial were P, K, Na and Mg. 2. Anti-cancer activity The in vivo experiments were carried out to test as follows: 1)anti-cancer activities on leukemia($L_1210$), Hepatlicus cancer($H_22$), Sarcoma180($S_180$) and 2)the effect on immune system through changes in intestine weight and the number of hemolytic plague forming cells. Every protein-bound polysaccharides in the present study showed anti-cancer activities on $L_1210$ and fruit body of Lentinus edodes 25mg/kg treatment and group showed the highestinhibition rate(86%) among these. Except for Lentinus edodes mycelial 5mg/kg treatment, every treatment group inhibited cancer on $H_22$ and Pleurotus ostreatus medium of post cultivate 25mg/kg treatment and fluit body of Lentinus edodes 25mg/kg treatment group showed the highest inhibition rate (86% and 71%, respectively) among these. The inhibited rates of fruit body and mycelial of Lentinus edodes 25mg/kg treatment group on $S_180$ a were 33.9%, respectively. Every samples at final conc. of 50, 100, 200, 400㎍/㎕ on in vitro cell toxicity test did not show significantly different cell death rates at P<0.05. In immune test, weights of liver and spleen were increased according to increase in conc. but were not significantly different at P<0.05. The weights of thymus were higher in fruit body and mycelial of Lentinus edodes treatment group then other but were not significantly different at P<0.05. Hemolytic plague forming cell which are capable of antibody formation were significantly high in fruit body and mycelial of Lentinus edodes treatment. Consequently, the anti-cancer activity of Lentinus edodes was thought to be mainly generated through immune-promoting mechanism. 3. effect of protein-bound poly-saccharides on blood glucose level and energy matabolism The hypoglycemic effects of 2 mushrooms, Pleurotus ostreatus and Lentinus edldes, on streptozotocin(STZ) induced diabetic rats were investigated in this study. Diabetes mellitus was induced in male Sprague-Dawley rats by the injection of STZ into the tail vein at a dose of 45mg/kg. Sprague-Dawley male rats(200-500g) were assigned to one control and three STZ-diabetic groups. Diabetic groups were assigned to STZ-control, Pleurotus ostreatus and Lentinus edodes groups. All groups were fed a AIN-76 diet. The two experimental groups were fed with each protein-bound polysaccharide(150 mg/kg BW) for 14 days and with carboxymethyl cellulose for STZ-control group. The body weight gain was monitored and the blood levels of glucose and cholesterol were measured, level of protein, triglyceride and the free fatty acid in plasma were analysed. Serum aminotransferase activity as also measured. The body weight gain was lower in the all diabetic groups than in the of normal group. The weight of spleen was reduced by administration of the Lentinus edodes protein-bound polysaccharides. The extent of blood glucose decrements in rats that fed Lentinus edodes protein-bound polysaccharides was signeficantly greater than found in the STZ-control group. The plasma levels of cholesterol and protein were not influenced in diabetic rats. The free fatty acid levels and alanine aminotransferase(ALT) activity were significantly decreased by administration of Lentinus edodes protein-bound polysaccharides. The result suggest that orally administered Lentinus edodes protein-bound polysacharides exhibited phpoglycemic effect in STZ-induced diabetic rats and that these protein-bound polysaccharides may be useful for the management of diabetes mellitus. 4. development of Health food (liquid extracts) The liquid products were to use as various beverages (tea) or pharmaceutical ingredients by vacuum concentration or by seasoning after extracting, the ingredients which have important phamacological function, mainly soluble polymers from fungus (e.g. Agaricus subfunereus). Products process : Washed into the fungus over 100% of its weight, extracted it for 30 minutes at 120℃, extracted juices, filtered, concentrated the concentrates in the reduced-pressure and made a product commercialization by adding sugar and seasonings with the concentrates (10 BX°)
