NOS는 3종의 isoform, 즉 $Ca^{2+}$에 의존적이며 constitutive form인 endothelial NOS와 neuronal NOS, $Ca^{2+}$에 비의존적이고 indicible form인 iNOS가 있으며, COX 또한 constitutive form인 COX-1과 inducible form인 COX-2가 존재한다. 염증반응에서 iNOS와 COX-2의 발현이 증가되며, 이로 인한 과량의 NO와 PG는 염증반응을 매개한다.
NF-kB는 면역반응을 조절하는 인자의 전사조절인자로서 잘 알려져 있으며, 이는 inflammatory cytokine(IL-1, IL-2, IL-6, IL-8, TNF-a 등)의 전사를 조절하고 있다. 또한 iNOS와 COX-2 유전자 promoter 부위에 NF-kB binding site가 존재하고 있어, 이 두 효소 또한 NF-kB에 의해 전사가 조절되고 있다. 따라서 iNOS 및 COX-2 발현 및 NF-kB 활성화 등은 항염증제 개발도구로서 널리 활용되고 있다.
본 과제에서는 이를 항염증제 개발도구로 활용하여 350종의 생약에 대하여 활성검사를 샐행하여 4종에서 7종의 단일화합물[β-carboline alkaloid (M-1, M-2; 고련피), Sequiterpene lactone (C-1, C-2 ; 긴담배풀, E-1; 선복화, N-1, N-3; 단풍취)]를 분리하여 그 작용기전을 규명하였다. 이러한 화합물은 macrophage cell line에서 세포 독성에 관계없이 농도 의존적으로 nitrite 및 PGE₂의 분비를 억제하였으며, Western blot와 Northern blot를 이용하여 iNOS 및 COX-2의 protein 및 mRNA의 expression을 억제함을 확인하였다. 또한 IkB의 분해를 억제하여 iNOS 및 COX-2의 중요한 transcription factor 인 NF-kB에 대한 핵 내로의 이행을 억제하여 DNA binding activity를 감소시켰다. 특히 exomethylene group를 가지는 sesquiterpene lactone류는 NF-kB의 cystein 잔기의 alkylation을 유도하여 NF-kB의 activity를 저해하였다. β-carboline alkaloid는 NF-kB 및 iNOS promoter activity를 억제함을 reporter gene를 이용하여 확인하였다. 위의 결과에서 4종의 천연물에서 분리한 7종의 화합물은 NF-kB의 activity를 억제함으로써 iNOS 및 COX-2의 activity를 감소시키고, NO와 PG의 생성을 억제하여 항염증작용을 나타내는 작용기전으로 설명할 수 있다. 따라서 본 연구는 β-carboline alkaloid 및 sequiterpene lactone을 새로운 항염증약물 개발의 lead compound로 활용하여 새로운 항염증약물개발 개발가능성을 제시한다.

Three isoforms of NOS exist, constitutively expressed neuronal NOS (nNOS) and endothelial NOS (eNOS), which are both $Ca^{2+}$ calmodulin-dependent, and an inducible isoform (iNOS) which is $Ca^{2+}$ calmodulin-independent. Two isoforms of COX exist, constitutively expressed COX-1 and an inducible isoform COX-2. iNOS and COX-2 are up-regulated in response to inflammatory and proinflammatory mediators, and their products are known to be an important mediator of acute and chronic inflammation.
Nuclear factor-kB (NF-kB) is a transcriptional factor that acts as a central mediator of the human immune response and regulates the transcription of various inflammatory cytokines such as IL-1, IL-2, IL-6, IL-8 and tumor necrosis factor-a as well as genes encoding cell adhesion molecules, immunoreceptors, hematopoetic growth factor and growth factor receptors. There are indications that activation of NF-kB is involved in iNOS and COX-2 expression because there is an NF-kB consensus site in the upstream promotor region of the iNOS and COX-2 gene. Therefore, iNOS and COX-2 expression, and NF-kB activation have been used as tools for the screening of anti-inflammatory activity.
Using iNOS and COX-2 expression, and NF-kB activation as a molecular target, we screened active compounds from many traditional herbal medicines (350 species) and found that β-carboline alkaloid (M-1 and M-2 from Melia azedarach) and 5 sesquiterpene lactones [C-1, C-2 (from Carpesium devericatum), E-1 (from Inula britannica), N-1 and N-3 (from Ainsliaea acerifolia) inhibited iNOS activity in RAW 264.7 macrophages. Here, we studied the suppression mechanism of NO PGE2, inflammatoty principles, by these compounds in Raw 264.7 cells: these compounds suppressed NO and PGE2 synthesis in RAW 264.7 macrophages through the inhibiton of iNOS and COX-2 gene expression, respectively. And, the inhibition of iNOS and COX-2 gene expression was, in large part, associated with the inhibition of nuclear translocation of NF-kB through stabilizing the NF-kB/IkB complex as well as the direct modification of active NF-kB. Furthermore, the ability to inhibit iNOS and COX-2 gene expression may account, in part, for the anti-inflammatory propoeties of these compounds. Thus, β-carboline alkaloids and sesquiterpene lactones appear to be a potential therapeutic agent for the treatment of inflammation or a lead compound for development of anti-inflammatory drugs.
