최근 리포좀, 양이온성 고분자, 입자를 포함한 비바이러스성 계를 활용한 유전자 치료 방법이 유전자 전달 분야에 연구되고 있다. 특히 양이온성 고분자를 활용한 유전자 전달계는 비특이적 방법으로 외부의 유전 물질을 세포에 전달하는 수단으로 각광받고 있다. 생분해성 고분자로 제조된 나노입자는 비활성, 생체적합성, 크기와 표면 변화에 의한 생물학적 요인의 조정, 고분자 담체의 분해속도의 변화에 의한 방출 조절의 용이성 등의 장점을 가지고 있다. 최근 리포좀, 양이온성 고분자, 입자를 포함한 비바이러스성 계를 활용한 유전자 치료 방법이 유전자 전달 분야에 연구되고 있다. 특히 양이온성 고분자를 활용한 유전자 전달계는 비특이적 방법으로 외부의 유전 물질을 세포에 전달하는 수단으로 각광받고 있다. 생분해성 고분자로 제조된 나노입자는 비활성, 생체적합성, 크기와 표면 변화에 의한 생물학적 요인의 조정, 고분자 담체의 분해속도의 변화에 의한 방출 조절의 용이성 등의 장점을 가지고 있다. DNA와 다가 양이온에 의해 형성된 복합체의 전이 효율은 다가 양이온과 DNA의 전하 비에 의존적이라고 알려져 있다.
따라서 나노입자의 장점을 살린 다가 양이온 함유 생분해성 고분자로 제조된 나노입자는 유전자의 전달체로서 효율적이라 생각되어 연구하였다. 여기서 다가 양이온으로 Polyethylene imine (PEI)을 사용하였고, 모델 plasmid DNA로는 유전자 발현 표준물질로 사용하는 PME 185/β-gal 및 Enhanced Green Fluorescence Protein (pEGFP)을 가지고 실험하였다. 생분해성 고분자로는 Polylactic acid (PLA), Poly(lactide-co-glycold) (PLGA)를 사용하였고, 이들 고분자를 사용한 나노입자를 제조하여 유전자 전달체로의 사용 가능성을 연구하였다.
이러한 연구의 일환으로 나노입자의 제조 조건 확립, 나노입자의 물리화학적 특성, 양이온성 고분자와 plasmid DNA의 복합체 형성 및 in vitro transfection 등에 대하여 연구하였다.
투석법 및 용매 증발법에 의해 제조된 나노입자는 구형이며 매끈한 표면 특성을 가지고 있었다. 나노입자 내에서 PEI의 양이 증가함에 따라 제조된 나노입자의 평균 직경은 감소 하였다. 동결건조 전 후의 평균 입자 크기의 차이는 PEI가 포함된 나노입자의 경우 작았다. 나노입자 현탁액의 안정성을 탁도 측정에 의해 평가하였다. PEI가 있는 나노입자 현탁액은 저장 15일까지 좋은 분산상 안정성을 보이고 있는 반면, PEI가 들어있지 않은 경우 3일 내에 입자의 빠른 침강을 보이는 불안정한 분산 안정성을 갖고 있었다. 이것은 사용된 고분자의 종류와 무관하게 나타났었다.
입자의 제타 전위는 PEI가 없는 PLA 또는 PLGA 나노입자는 (-)값을 보이고 있었으며 PEI의 첨가에 따라 제타 전위 값의 부호는 (-)에서 (+)로 바뀌었다. 나노입자 표면의 (+) 전하와 DNA에 있는 인산의 (-) 전하간의 상호작용에 의해 복합체가 형성 될 수 있었다. 나노입자와 DNA간의 복합체 형성은 겔 전기영동 실험에 의해 조사되었다. 나노입자의 양을 증가시키면 DNA의 이동에 감소를 일으키고 나노입자 내의 PEI와 plasmid DNA와의 무게 비 는 30:1 정 도에서 부터 free DNA가 나타나지 않았으며 , 이 이 상에서 는 모든 DNA가 나노입자의 표면에 붙잡힘을 의미하였다. 사용된 고분자의 종류에 무관하게 모든 결과가 같았으며 이로보아 DNA와의 복합체 형성은 양이온성 고분자의 농도에만 의존함을 알 수 있었다. 또한 복합체 형성은 나노입자의 표면에 있는 양이온성 고분자의 (+) 아민기와 plasmid DNA의 (-) 인산기의 전하 상호작용으로 일어났으며 이것은 복합체가 형성됨에 따라 제타전위의 값이 (+)에서 0으로 감소하는 것으로 알 수 있었다. plasmid DNA가 나노입자 표면에 흡착되어 있어 표면 전위가 감소하였고 흡착 현상은 일반 흡착 현상인 Langmuir 형 흡착을 보이고 있었다.
plasmid DNA와 결합하는데 중요한 인자인 양이온성 고분자의 종류 및 농도에 따른 물리 화학적 특성 및 in vitro transfection을 연구하였다. PEI의 분자량을 800, 25000, 750000 세 종류를 사용하였다. 분자량에 관계없이 모두 우수한 나노입자를 제조할 수 있었다. 다만 분자량이 750000인 경우 안정성이 낮았고 표면의 제타전위 값도 (+)를 유지하지 못하고 있어 in vitro transfection 효율이 가장 낮았다. transfection 효율은 PEI가 들아있지 않거나 농도가 낮은 경우 비교적 낮았으며 이 결과는 겔 전기영동 실험을 통해 복합체 형성 여부를 판단한 결과와 일치하였다. 또한 입자의 크기, 제타 전위, 분산상의 안정성에 따라 transfection 효율이 달라지는 것으로 보아 안정한 나노 입자의 제조가 가장 중요한 인자라는 것을 알 수 있었다.
이상의 결과로부터 안정한 분산성을 갖는 PEI 함유 PLA 또는 PLGA 나노입자는 DNA 전달체로서 사용 될 수 있다고 결론지었다.

Recently, gene therapeutic approach using nonviral system including liposome, cationic polymers and particles has been studied in the field of gene transfer. Gene delivery system using biodegradable polymers, especially, was in the highlighted as a means to deliver the exogeneous genetic materials to cells in nonspecific manners. Nanoparticles prepared with biodegradable polymers have several advantages such as inertness, biocompatibility, easy control of biological factors by changing the size and surface, and control of release by varying the degradation rate of polymer matrix. The transfer efficiency of complex formed by DNA and polycations are known to be dependent on the charge ratio of polycations and DNA. Therefore, the studied the nanoparticles prepared with biodegradable polymers containing polycations as a DNA carrier.
Here, we reported the preparation and characterization of the polylactic acid (PLA) and polylactide-co-glycolide (PLGA) nanoparticles containing polyethyleneimine (PEI) as a polycation and PME 185/β-gal as a model DNA. The effect of the PEI on the physiochemical characteristics of nanoparticles and adsorption efficiency of DNA were also examined.
Nanoparticies prepared by a diafiltration or evaporation method had a spherical shape and a smooth surface characteristics. When the PEI amount in polymers was increased, the mean diameter of the result nanoparticies was decreased. The difference of average size between the nanoparticies before lyophilization and those after lyophilization was small in the case of the nanoparticies containing PEI. The stability of the nanoparticles suspension was evaluated by turbidity measurement.
The nanoparticles suspension with PEI exhibited good dispersion stability until 15 days of incubation, while the suspension without PEI had poor dispersion stability showing a fast setling of particles within 3 days. The zeta-potential of PLA or PLGA nanoparticles without PEI showed a highly negative value and the sign of the zeta-potential valueschanged from minus to plus in addition of PEI. By the charge interaction between the (+) charge of nanopcuticle surface and (-) charge of phosphate in DNA, the complex could be formed. Complex fommtion bet\veen the nanoparticles amount caused a gradual decrease in movement of DNA This indicated that all of DNA was trapped on the surface of the nanoparticles. This observation was independent of the type of polymers and was only depend on the concentration of PEL Transfection efficiency was measured using absorbance detection of total β-galactosidase activity or cell counting of fluorescence imaging cells. Generally, very little transfection achieved in cells treated with polylactic acid nanoparticles without PEI. Complexes formed using PLA/PEI nanoparticles promoted a greater transfection efficiency than that obtained using PLA nanoparticles. PEI 800 or PEl 25000 system was more efficient tools of in vitro transfection of DNA in cells than PEl 750000 system. We found that in vitro transfection efficiency was related with the size, zeta potential and dispersion stability of nanoparticles.
From these results, such as good preparation of nanoparticles, stable dispersion and easy binding of DNA with PEI, we concluded that PLA or PLGA nanoparticles with PEI can be used as a carrier for plasmid DNA.
