본 연구에서는 HPV 및 KSHV의 특성연구에 대한 새로운 접근 방법으로서, 바이러스 단백질과 상호 작용하는 host factor들의 동정 및 특성을 밝히고, 이를 통하여 바이러스 생활사에 결정적인 factor를 찾아내고 종양유발 단백질의 발암 메카니즘을 규명하고자 하였다. 또한 바이러스 DNA replication system을 확립하고, 복제에 중요한 factor를 다양한 방법으로 조사하게 되었다. 이러한 접근 방법을 통해 HPV의 복제와 전사에 중요한 단백질인 E2가 세포내 단백질 CBP, pCAF, PARP과의 기능적으로 결
본 연구에서는 HPV 및 KSHV의 특성연구에 대한 새로운 접근 방법으로서, 바이러스 단백질과 상호 작용하는 host factor들의 동정 및 특성을 밝히고, 이를 통하여 바이러스 생활사에 결정적인 factor를 찾아내고 종양유발 단백질의 발암 메카니즘을 규명하고자 하였다. 또한 바이러스 DNA replication system을 확립하고, 복제에 중요한 factor를 다양한 방법으로 조사하게 되었다. 이러한 접근 방법을 통해 HPV의 복제와 전사에 중요한 단백질인 E2가 세포내 단백질 CBP, pCAF, PARP과의 기능적으로 결합한다는 사실을 밝혀내고, 종양 유발 단백질인 E7과 E2F1의 기능적 결합을 분자적 수준에서 이해함으로써 E7의 종양 유발 메카니즘 이해와 이와 관련된 자궁경부암의 기작 연구에 획기적 돌파구를 제시할 것으로 기대된다. 아울러 KSHV의 다양한 lytic 유전자들(Rta, bZIP, vIRF)이 CBP와 p53과 같은 세포내의 중요한 전사 조절인자들과 기능적으로 상호작용하고 있다는 사실을 분자 수준에서 이해함으로써, 바이러스의 발암 기작과 바이러스 증식 및 세포 조절의 메카니즘의 이해에 크게 기여하였다고 평가된다. 아울러 replication level에서의 HPV El, E2 와 KSHV의 LANA의 새로운 기능을 규명하고, LANA와 ORC간의 상호작용을 규명함으로써, 숙주세포내에서 바이러스 복제기작 연구와 항바이러스제제 개발에 크게 기여할 것으로 평가된다.

Kaposi′s sarcoma-associated herpesvirus, also called human herpesvirus 8 (KSHV/HHV8), is the likely etiological agent involved in the development of Kaposis sarcoma and several lymphoproliferative diseases including primary effusion lymphoma/body-cavity based lymphoma (PEL/BCBL), and some cases of multicentric Castlemans disease (Cao et al., 1996; Soulier et al., 1995; Nador et al., 1995). The viral genome contains more than 80 open .ending frames (ORFs) and expresses distinct sets of viral proteins during latent and lytic phase of viral infection. One of the viral proteins expressed during latent infection is the latency-associated nuclear antigen (LANA), which is encoded by ORF73 of viral genome. LANA mediates the maintenance of viral genome by tethering it to host chromosome. To identify LANA-interacting proteins of host cells, we performed yeast two-hybrid assay using full-length LANA as a bait and found activating transcription facto. (ATF4) / cAMP-response element binding protein (CREB) 2 as a cellular binding-partner of LANA. LANA bound to ATF4/CREB2 in vivo and in vitro, and the binding of LANA to ATF4/CREB2 results in the repression of transcriptional activity of ATF4/CREB2 in a transient reporter assay.
KSHV ORF50, one of viral immediate early genes expressed after lytic cycle induction, is the key regulator of the switch between latent and lytic cycle of viral infection. We found CREB-binding protein (CBP) and histone deacetylase (HDAC) 1 as a positive and negative regulator of the transcriptional activity of ORF50 in a transient reporter assay, respectively. In an in vivo and in vitro binding assay, ORF50 interacted with CBP and HDAC1, and colocalized in overexpressed 293T cells. The transcriptional regulation of ORF50 correlated with direct binding to CBP and HDAC1 in a transient reporter assay, and consequently the lytic cycle induction of KSHV-infected BCBL-1 cells by ORF50 was positively or negatively regulated by CBP or HDAC1, respectively We also found the CBP is targeted by other viral proteins including LANA and K8 of KSHV. LANA interacted with the C/H of CBP in an in vivo and in vitro binding assay, and interfered with the interaction between CBP and cellular factor in a transient reporter assay and a competitive in vitro binding assay. The transcriptional activity and in vitro histone acetyltransferase activity of CBP was negatively regulated by LANA. Similarly, K8 was also identified as a negative regulator of CBP by direct binding to the C/H3 region of CBP. These results suggest that, during viral infection, several viral proteins of KSHV target CBP, the signal integrator, to utilize as a transcriptional coactivator or regulate the transcriptional network of host cells. Virus should inhibit the programmed cell death of host cells to survive and proliferate. Therefore, many virus evolved several strategies deregulating apoptotic activity of host cells. One of them is to inhibit the activity of p53, the tumor suppressor, involved in cell cycle regulation, apoptosis, DNA damage response,and differentiation.
