본 연구에서는 누에고치 단백질의 가수분해물에 대하며 당뇨병 억제활성 및 암예방 활성을 규명하고 활성을 나타내는 고활성 펩타이드 분획을 생산하는 것을 목표로 하였다. 당뇨병 억제활성 규명을 위해서는 혈당강하효과 및 lipolysis 억제활성을 조사하였으며, 암예방 활성 규명을 위해서는 항유전독성, 항산화활성, quinone reductase(QR) 유도활성, 면역활성증강효과 등의 생리활성을 조사하였다.

본 연구에서는 누에고치 단백질의 가수분해물에 대하며 당뇨병 억제활성 및 암예방 활성을 규명하고 활성을 나타내는 고활성 펩타이드 분획을 생산하는 것을 목표로 하였다. 당뇨병 억제활성 규명을 위해서는 혈당강하효과 및 lipolysis 억제활성을 조사하였으며, 암예방 활성 규명을 위해서는 항유전독성, 항산화활성, quinone reductase(QR) 유도활성, 면역활성증강효과 등의 생리활성을 조사하였다.
정선된 누에고치로부터 피브로인을 용해시켜낸 후 산 또는 효소에 의해 생산된 가수분해물에 대하여 각 생리활성을 측정하였다. 확인되는 활성에 대해서는 가수분해물을 한외여과 또는 Sephadex G-25 gel chromategraphy에 의해 고활성 분획을 분리해내었다. 우선 직접발암원인 MNNG(N-methyl-N'-nitro-N nitroso-guanidine)에 대한 피브로인 가수분해물의 항유전독성을 실험하기 위해 해양세포를 이용한 Comet assay를 사용하였다. 각 가수분해물에 대하여 농도에 따른 항유전독성을 측정하였고, 이어서 각 가수분해물을 gel filtration chromatography를 이용해 분리한 각각의 분획에 대한 항유전독성을 측정하여 가장 활성이 우수한 펩타이드 분획을 얻고자 하였다. 한편 가수분해물이 나타내는 항유전독성의 기작을 알아보기 위한 실험도 병행하였다. 피브로인의 각 가수분해물은 처리농도를 증가시킴에 따라 농도의존적으로 DNA 손상 억제효과를 나타내었으며 특히 10 mg/ml의 농도로는 거의 negative control 값에 근접하는 억제효과를 보여줌으로써 매우 높은 항유전독성을 가지고 있음을 확인할 수 있었다. 그 중 산가수분해물과 Alcalase 가수분해물이 가장 높은 활성을 보엿으며 이로부터 가장 높은 활성 펩타이드 분획을 얻을수 있었다. 결국 각 가수분해물에서 분자량 1000 이하의 비교적 작은 분자량의 펩타이드 분획에 항유전독성 펩타이드가 집중적으로 포함되어 있음을 알 수 있었다. 활성 펩타이드들은 3∼7의 아미노산을 가지는 작은 3기로 추정되었으며 펩타이드내 glycine과 alanine의 양도 활성에 중요한 영향을 미치는 것으로 추정되었다. 또한 활성기작을 분석한 결과 펩타이드 성분이 MNNG와 결합하여 alkylating agent로서의 역할을 방해하는 기작 뿐만 아니라 세포와의 직접적인 반품을 통해서도 항유전독성을 나타냄을 알 수 있었다. 한편 피브로인 가수분해불의 항산화 활성을 측정하기 위하여 MNNG 대신 hydrogen peroxide를 사용하여 동일한 실험을 실시한 결과 hydrogen peroxide의 DNA damage에 대해서는 보호효과가 나타나지 않음으로써 항산화 활성을 확인할 수 없었다. 또 다른 항암활성으로서 가수분해물들의 quinone reductase(QR) 유도활성을 측정한 결과 모든 가수분해물이 비교물질인 t-butylhydroquinone 및 β-naphthoflavone에 비하여 QR 유도활성이 매우 낮았으며 또한 처리농도와 QR 활성 증가치 간의 유의성도 관찰되지 활성이 거의 없음을 확인할 수 있었으며 가수분해물에 대한 80% methanol 추출 펩타이드 분획 역시 활성이 나타나지 않았다. 마지막으로 면역활성 촉진을 통한 암예방 활성의 탐색을 위하여 피브로인의 각 가수분해물을 대식세포에 처리하여 nitric oxide(NO) 생성량을 측정한 결과 NO 생성을 촉진시키는 활성이 있음을 발견하였다. 활성은 peptic hydrolysate가 가장 높았고 tryptic-, Alcalase hydrolysate 순이었다. 이러한 활성의 차이는 가수분해물 내의 고분자량 펩타이드 분포가 많을수록 활성이 높다는 관계에 기인하였으나 산 가수분해물의 경우에는 예외적으로 나타났다. 각 가수분해물의 아미노산 조성을 분석한 결과 arginine, lysine의 함량이 높을수록 활성이 높으며 alanine의 glycine에 대한 비율이 커질수록 활성이 높아졌다. 산 가수분해물의 경우에는 낮은 분자량의 펩타이드들이 많이 분포하지만 arginine 및 alanine의 함량이 높아 비교적 높은 NO 생성촉진활성을 나타내는 것으로 확인되었다. 이어서 피브로인 산 가수분해물의 in vivo 항당뇨 작용을 조사한 결과 인슐린 의존형 당뇨병 모델동물(streptozotocin(STZ)에 의해 당뇨유발시킨 Sprague-Dawley(SD) rat)에서는 체중 증가량의 감소현상을 완화시켜주는 한편 혈당치 상승이 유의적으로 억제되었으나 인슐린 비의존형 당뇨병 모델동물(C57BL/6-db/db 마우스(db/db))에서는 체중의 변화에도 영향이 없었으며 혈당강하효과도 나타나지 않았다. 한편 두 모델동물에서 동일하게 피브로인 가수분해불의 급여에 의해 혈중 leptin 농도가 증가하며 또한 급여농도에 따라 증가량도 높아지는 현상이 관찰되었다. 이러한 결과로부터 피브로인 가수분해물의 인슐린 분비촉진 효과를 확인할 수 있었으며 두 모델동물에서 각각 체중의 변화와 혈당강하효과가 다르게 관찰되는 것은 해당 당뇨병의 기작이 다르기 때문인 것으로 판단되었다. 또한 항당뇨 활성의 다른 측면으로서 lipolysis 억제활성을 측정하엿다. SD rat의 고환 주변 지방조직으로부터 분리해낸 mature adipocyte에 대하여 피브로인 가수분해물을 처리하고 그 배양액 중의 glycerol 량을 측정하여 lipolysis 활성의 변화를 조사하였으나 피브로인 가수분해물은 아무런 변화도 일으키지 못함을 확인하여 lipolysis 억제를 통한 항당뇨 활성은 확인할 수 없었다.
이상과 같이 누에고치 단백질 주성분인 피브로인으로부터 가수분해물을 제조하여 항당뇨, 항암활성을 조사한 결과 항당뇨, 항유전독성, NO 생성촉진활성 등이 확인되었으며 고활성의 펩타이드 분획이 분리되었고 활성에 영향을 미치는 영향들을 규명하였다. 이들 고활성 펩타이드 분획들에 대해서는 항당뇨 및 암예방 기능성 소재로서의 응용이 가능할 것이다. 또한 추가적인 activity guided fractionation 과정을 거쳐 주효 활성의 단일 펩타이드(군)을 분리해냄으로써 의약소재로도 응용이 가능하다. 즉 본 연구를 통해 현재 식용으로도 이용되고 있는 천연소재인 피브로인의 새로운 응용분야가 제시됨으로써 안전성이 높은 기능성 식품 또는 의약품의 개발이 가능해질 수 있게 되었다.

In this study, the antidiabetic and/or chemopreventive activities of cocoon protein hydrolysates were investigated and the active peptide fraction(s) were separated. Studies were focused on in vivo antihyperglycemic and antilipolytic effects for antidiabetic activity and antigenotoxic, antioxidizing, quinone-reductase(QR) inducing, and NO-production stimulating effects for chemopreventive activity.
Fibroin, the major protein of silk, was extracted from cocoon and hydrolyzed by acid or industrially applicable proteases. In the case that any activity was detected from a hydrolysate, the active peptide fractions(s) were separated by ultrafiltration and/or Sephadex G-25 gel chromatograpy. A study was conducted to examine in vitro antigenotoxic effects of the peptides derived from the fibroin hydrolysates in mouse embryo 3T3 cells. Antigenotoxicities were determined by measuring the reduced levels of DNA damage using the Comet assay. The acid- and Alcalase hydrolysates showed higher antigenotoxic activities than peptic- and tryptic hydrolysates. Active peptide fractions were separated from acid- and Alcalase hydrolysate by gel filtration chromatorgrapy. It was deduced from the chromatograms and amino acid analyses that the size (3∼7 amino acids) and the glycine plus alanine content of peptides might be the important factors for their activities. In addition, it was found that the antigenotoxic effect of peptides was due both to the protective interactions between cells and peptide molecules and to the direct inactivation of the mutagen, MNNG by peptides. Considering the availability and safety of silk fibroin and the superior antigenotoxic effects of produced peptides, the results of this study showed the possibility of utilizing fibroin as a source for chemopreventive functional peptides.
In addition, it was found that the peptides originated from the hydrolysates of fibroin have in vitro immunostimulating effects in murine macrophage RAW264.7 cells. The stimulation effects on nitric oxide (NO) production resulted from treatments of acid or enzymatic hydrolysates were measured. Even though the sole treatment of acid hydrolysate stimulated the NO production in dose-dependent pattern, a part of its activity was found to be caused by the contaminated endotaxin, LPS. When each endotoxin-free hydrolysates obtained by filtering it through an ultrafiltration membrane of molecular weight (MW) cut-off 10,000 to eliminate LPS was used, the peptic hydrolysate with lowest degree of hydrolysis showed the highest activity. The fractions of peptic hydrolysate with MW ranges of 1,000∼10,000, 500∼l,000 and below 500 also showed a higher MW-higher activity correlation. From the analyses of amino acid composition of each hydrolysate, it was found that the contents of arginine, lysine, alanine and glycine residues affected the activity level of hydrolysate. The results of this study showed a possibility of utilizing fibroin as a source for immunostimulating (chemopreventive) functional peptides.
Antidiabetic effects of the acid hydrolysate of fibroin were investigated by oral administration to animal models for diabetes mellitus. Feeding of the fibroin hydrolysate resulted in a significant recovering effect on reduction of body weight gain and a lowering effect on blood glucose gain in streptozotocin-induced diabetic Sprague-Dawley rats (STZ rats) which were used as an insulin-dependent diabetic animal model. But the body weight and blood glucose level in C57BL/KsJ-db/db mice (db/db mice), an non-insulin-dependent diabetic animal model, were not changed significantly by the feeding. On the other hand, plasma leptin levels increased according to increased feeding amount of the hydrolysate in STZ rats and db/db mice in common. It was concluded from the results that the fibroin hydrelysate night stimulate the insulin secretion by recovering or activating pancreatic cells and result in the increased plasma leptin level. It was also deduced that the antidiabetic improvements in body Weight and blood glucose gain in STZ were thought to be due to the increased insulin secretion, but in db/db mice of which the diabetic symptoms were caused by insulin resistance, the stimulated secretion of insulin was unlikely to be able to change body weight and blood glucose level significantly.
