현재 시중이나 한의원, 한약방등에서 이용되는 한약 소재 자원류는 동양의약학 분야에서 큰 비중을 차지하고 있다. 또한 전통적인 식품의 원료로 이용되는 야생 식물 자원류 중의 일부는 현재 기능성 식품의 원료로 이용되기도 한다. 특히 식물 자원류는 동양의약학적 재료와 식품재료로 혼용되고 있으며 일부는 이 재료들이 각 업종에 종사하는 사람이나 학문적으로 구분할 수 있는 뚜렷한 근거가 없고 국내의 경우는 다만 고서 자료를 근거로 각 이익 집단의 소유권 분쟁의 대상도 되고 있다. 이러한 식품 및 한의약의 재료들은 그 사용량이나 성분이 확실히 밝혀져 있지 않은 것도 많고, 그 기능이 과학적인 근거가 불확실한 것도 있다. 더욱이 그 자원류는 인체에 위해성이 우려되는 것등이 있을 수 있으므로 이 자원류 를 국 내외적 유용자원으로 활용하기 위하여는 그 위해성의 확인이 필수적이다. 이번 연구에서는 1차년도에 우선 이러한 위해성이 우려되고 사용빈도가 많은 원자재를 선택하여 독성 여부를 조사하기로 하였다. 연구방법은 동양 한의약 원자재외 기능성 식품원료로 이용되거난 또는 혼용되는 원료중에서 국내 농가에서 재배하거나 또는 국내 야생 자원을 채집한 것을 현장에서 확인하여 구입하여 사용하였다. 이들 재료중 약 30 여종을 선택하여 in vitro 방법으로 조사하기로 하였다. 사용된 방법은 Salmonella typhimurium TA 100 및 TA 98을 이용하는 Ames test, Salmonella typhimurium 균을 이용하여 독성 시험용으로 개발한 SOS/umu test, 동물세포를 이용하여 독성을 조사하는3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide(MTT) 법 과 유사한 수단인 비색 SRB(Sulforhodamine B ) 법, 동물세포의 DNA 손상에 의하여 독성을 조사하는 Comet assay 등을 이용하여 조사하였다. 이상의 실험에서 대체로 유사한 결과를 얻었으며 일부의 시료는 방법에 따라 다소 차이가 있는 것도 있었다. Ames test에서는 TA 100에서는 만병초가 11mg/plate의 농도에서가장 높은 돌연변이원성을 나타내었으며, 황기, 뽕, 겨우살이 등에서 높은 독성나타났다. TA 98에서는 백출이 28.6mg/plate의 농도와, 황기가 11.4mf/ml의 농도에서 다른 시료에 비하여 높은 독성이 나타났다. 암세포에 대하여서는 만병초, 작약, 가시오가피 줄기, 산마늘 등이 0.5mg/ml 이상의 농도에서 SRB와 MTT에서 높은 독성을 나타내었으며, 구기자, 오미자 등은 낮은 생육억제능을 보였다. comet assay 실험시 sample의 투여 농도는 각 sample의 IC50을 정하고 그 농도를 최대로 하여 50%씩 dilution하여 3가지 농도로 처리하였으며, 이중 10여종의 시료에서 50%이상의 세포사멸이 관찰되어져 comet assay를 실시한 결과, 오가피와 뽕잎, 지치 등에서 유의적인 OTM치를 나타내어 이들이 NIH 3T3 세포의 핵에 독성을 나타낼 것으로 사료되어진다.

Although the toxicological stabilities and constituents of these plants were not studied scientifically, those materials have been used as traditional medicinal and functional food materials in many oriental medicinal doctors and functional food companies. Many consumers have been concerned about the toxic stability of the materials. The toxicological stabilities of the oriental medicinal and functional food materials have been investigated primary in vitro screening methods. The materials were bought from farmers and farmer's sale guilds in the country(mainly Kangwondo) in Korea. The samples were prepared as follows: The materials were washed with water, and dried in the room. These were extracted with 70% ethylalcohol, and frationationed several organic solvent. The extractans freezedried were used to sample for the screening tests. Screening methods employed were Ames test with Salmonella typhimurium TA 98 and TA 100, SOS/umu test with Salmonella typhimurium (TA1535/pSK1002), and 3-[4,5-dimethylthiazol-2-yl] -2,5-diphenyltetrazolium bromide(MTT) test with animal cells, Sulforhodamine B(SRB) test with animal cells, comet assay(single cell gel electrophoresis test) to detect DNA damage in animal cells. Many samples showed a mutagenicity and toxicological positive in the screening tests, and the positive samples were similar to the test systems. Ames test was a positive at higher concerntrations than that in MTT test. The results of Ames test were provided that extracts of Rhododendron brachycarpum, Atractylodes rhizoma, Astragalus membranaceus showed mutagenic activities. MTT and SRB tests with animal cells were very sensitive at low concentrations. About 79% of the growth of A549 and 73% of Hep3B and 90% of MCF-7 was inhibited by SRB test in adding 1.0mg/ml of the ethanol extracts of Rumex crispus L., Allium victorialis (Ullnug) and Acanthopanax senticosus Branch, respectively. About 90% of the growth of A549 and 73% of Hep3B and 90% of MCF-7 was inhibited by MTT test in adding 1.0mg/ml of the ethanol extracts of Paeoniae radix, Acanthopanax senticosus Root and Rhododendron brachycarpum, respectively. The results of comet assay were provided that extracts of Lithospermum erythrorhizon, Acanthopanacis Cortex, Lithospermum erythrorhizon. showed 7~8 OTM.
