천연물을 이용한 기능성 화장품에 대한 평가기술을 개발하기 위하여 복분자 딸기 열매, 오리나무 그리고 왕느릅나무 중 pedunculagin, oregonin 그리고 taxifolin-glycoside를 각각 유효지표성분으로 선택하여 1차적으로 각각의 식물 중에서의 함량분석법을 확립하였다. 2차적으로는 이러한 성분을 이용한 화장품 제제를 만들었을 때에 유효성분을 검출할 수 있는 방법을 확립하기 위하여 실제로 식물 추출물을 이용하여 화장품 제제를 만들어서 유효지표성분을 검출할 수 있는 분석방법을 확립하였다. 그 결과, 각 식물의 유효지표
천연물을 이용한 기능성 화장품에 대한 평가기술을 개발하기 위하여 복분자 딸기 열매, 오리나무 그리고 왕느릅나무 중 pedunculagin, oregonin 그리고 taxifolin-glycoside를 각각 유효지표성분으로 선택하여 1차적으로 각각의 식물 중에서의 함량분석법을 확립하였다. 2차적으로는 이러한 성분을 이용한 화장품 제제를 만들었을 때에 유효성분을 검출할 수 있는 방법을 확립하기 위하여 실제로 식물 추출물을 이용하여 화장품 제제를 만들어서 유효지표성분을 검출할 수 있는 분석방법을 확립하였다. 그 결과, 각 식물의 유효지표성분의 함량 분석법을 확립한 후, 실제 함량 분석을 실시한 결과 복분자 딸기열매 엑스 중 pedunculagin 1.313%, 오리나무 수피 엑스 중 oregonin 6.308% 그리고 왕느릅나무 수피엑스 중 taxifolin-glycoside의 함량 7.510%가 포함되어 있음을 알 수 있었다. 그리고 더 나아가 가수 분해성 탄닌을 유효지표성분으로 하였을 때 쉽게 분해되어 제제화가 어려운 복분자 딸기를 제외한 나머지 식물 추출물 1%를 포함한 화장품을 제제화하여 그 안에 유효지표성분의 함량 분석법을 확립하였다. 실제 함량 분석을 실시한 결과 오리나무의 지용성 화장품 제제 중의 0.0548%, 수용성 화장품 제제중의 0.048%의 oregonin이 분석되었으며, 왕느릅나무의 지용성 화장품 제제 중의 0.062%, 수용성 제제중의 0.059%의 taxifolin-glycoside의 함량이 분석되었다 (제1세부과제). 또한 복분자 딸기 열매, 오리나무 그리고 왕느릅나무 중 pedunculagin, oregonin 그리고 taxifolin-glycoside를 각각 유효성분으로 선택하여 미백, 항산화 및 주름개선 생리활성을 검색하였다. 2차적으로 화장품 제제에서의 추출법을 확립하기 위해 식물 추출물을 이용한 추출분석법을 검색하였고 화장품 제제 추출물의 미백, 항산화 및 주름개선 생리활성 평가지표를 마련하기위한 실험을 통해 평가 기초자료를 제공하였다. 즉, 각 유효성분으로부터 미백, 항산화 효과가 있음을 확인하였고, 화장품제제에서의 생리활성 실험 결과 화장품제제에서 tyrosinase inhibitory activity을 측정하는 경우에는 최소한 1g 이상의 화장품 (수용성 및 지용성 base)으로부터 추출하여, 25㎕/㎖ 농도에서 그 효능을 비교하는 것이 바람직하다고 판단되었다. DPPH를 이용한 항산화 실험에서는 1% UBA 와 UBB에서 화장품을 많이 취득할 수록 IC50치가 낮게 나타나고 오리나무의 경우 0.5g, 느릅나무의 경우 1.0g 이상의 화장품으로부터 추출하여 DPPH 실험을 통한 항산화 효능을 비교하는 것이 바람직하다고 판단되었다. 화장품제제에서의 melanin 생성억제와 tyrosinase 활성억제효과가 있었으며, elastase 활성억제효과가 나타났다 (제2세부과제).

To establish the herbal whitening cosmetics as a international competitive goods in the cosmetic industry, we selected phenolic compounds as a whitening-effect cosmetics material and HPLC analysis about each phenolic compound of three plant. The selected phenolic compounds are 1 tannin compounds (pedunculagin) from Rubus coreanum, 1 diarylheptanoid (oregonin) from barks of Alnus japonica and 1 flavonoid from barks of Ulmus macroparca. The results of HPLC analysis showed that the percentage of pedunculagin from Rubus coreanum is 1.313 %, the percentage of oregonin is 6.308 % and the percentage of taxifolin-glycoside is 7.510%. And, we studied futherer experiment about oil and water based cosmetics. These oil and water based cosmetics had made by extract of Alnus japonica and Ulmus macroparca. However, Rubus coreanum was hard to make because pedunculagin was hydrolysed tannin easily dissembled. The HPLC analysis results of oil and water based cosmetics exhibited that the percentage of oregonin from oil-based cosmetics is 0.0548 % and that from water-based cosmetics is 0.048 %. Also, in case of taxifolin-glycoside from Ulmus macroparca, the percentage of taxifolin from oil-based cosmetics is 0.062 % and that water-based cosmetics is 0.059 %. From 2 details, we studied whitening-effect, antioxidant-effect and wrinkles improvement-effect by selected phenolic compounds - pedunculagin, oregonin and taxifolin-glycoside. Secondary, to establish the extract method in herbal cosmetics, we was searched extract analysis. And to establish a basic valuation of whitening-effect, antioxidant-effect and wrinkles improvement-effect of extracts of herbal cosmetics, we studied tyrosinase inhibitory activity assay and DPPH assay. We established the extract method in herbal cosmetics and the results of these assay showed that was acquired more than 1g in cosmetics (Alnus and Ulmus) at tyrosinase inhibitory activity assay, was acquired more than 0.5g(Alnus) and 1g(Ulmus) in cosmetics at DPPH assay. Also, melanin content and tyrosinase activity of the B16 melanoma cells were reduced by the addition of these samples. In elastase inhibitory activity assay, these samples inhibited the porcine pancreatic elastase (PPE) activity significantly.
