현대산업사회의 산업 화학독성물질의 증가, 조합화학 (Combinatorial chemistry), High throughput screening system (HTS) 등의 도입으로 인한 다수의 신약후보물질의 등장으로 독성 실험의 수요가 급증하고 있으며 간독성을 조기 검색할 수 있는 새로운 시험법의 확립이 절실히 요구되고 있는 현실임. 물질의 독성과 안전성 여부를 조기에 예측하고 판정하여 부작용을 극소화하기 위해서는 독성유전체학을 이용한 간독성 관련 특이 유전자의 발굴과 데이터베이스 구축을 통한 새로운 독성 시험법이 절실히 요구됨. 본 연구에서는 전형적인 간독성을 보이는 약물인 타목시펜을 이용하여 동물 간에서의 유전자 발현 변화 양상을 분석함으로써 약물의 안전성과 관련된 특이적인 유전자를 DNA microarray 법으로 발굴하여 독성관련 유전자 데이터베이스를 구축하고 간독성 진단 Chip의 개발과 신약개발을 위한 인프라 구축에 기여하고자 함. 다음과 같은 실험방법이 성공적으로 수행 되었음. ① 시험약물: 유방암치료제인 타목시펜을 간독성 약물로 선정 ② 실험동물 (C57BL6 mouse)에 대조군, 저용량 (10 mg/kg), 중간용량 (50 mg/kg), 고용량 (100 mg/kg)으로 투여하고 각각 2, 4, 8, 24시간 후 간 적출함. ③ 혈액 생화학적 변화 및 간 조직의 조직병리학적 변화 조사 a. 혈액 생화학적 변화 조사 b. 간 조직의 조직병리학적 변화 조사 ④ 유전자 발현 변화 분석: DNA microarray (GeneChipTEX>$^{\circledR}$ Mouse Genome 430A 2.0 Array, Affymetrix, USA) ⑤ 유전자 발현 데이터의 분석: 유전자 프로파일링, 클러스터링과 생물학적 경로 분석 본 연구의 성공적인 수행으로 얻은 결과는 국내에서 독성유전체학의 성공적인 도입과 확립에 기여 할 것이며 안전성 관련 유전자의 데이타베이스를 구축하고 공유하기 위한 국제적인 표준화 protocol의 확립에 기여할 수 있을 것임. 타목시펜의 간독성 유발에 관련된 유전자 발굴과 분자생물학적 기전규명을 통하여 유방암 치료 및 예방용 신약개발에 기여하여 국내 제약 산업의 활성화에 기여할 수 있음. 또한 간독성을 신속하게 예측 평가할 수 있는 DNA chip 개발에 필요한 유전자 자료를 제공함.

Recently, due to the increases in number of chemical pollutants generated from industry and drug candidates developed from combinatorial chemistry and high throughput screening system (HTS), examinations on toxicity, especially for hepato-toxicity, for these compounds are heavily required. Toxicogenomics is an emerging field combining genomics and bioinformatics to identify and characterize mechanism of toxicity of compounds. In the present investigation, we performed DNA microarray experiments that detect changes in gene expression upon tamoxifen treatment, to identify discrete gene expression changes associated with a hepato-toxic response and to elucidate mechanisms of the hepato-toxicity of this drug. For the purpose, the following experimental condition was employed. ① Test Drug: Anti-breast cancer agent, Tamoxifen ② Experimental Animal (C57BL6 mouse): Vehicle control, low dose (10 mg/kg), medium dose (50 mg/kg), high dose (100 mg/kg) were orally administered for 2, 4, 8, 24 hours. ③ Examination of pathological and biochemical changes in blood and liver. a. Examination of biochemical changes in blood: GOT, GPT b. Examination of pathological changes in liver tissue ④ DNA microarray (GeneChip$^{\circledR}$ Mouse Genome 430A 2.0 Array, Affymetrix, USA) ⑤ Data Analysis: Profiling of genes responsible for the tamoxifen-induced hepato-toxicity and analysis of the associated biological pathways were provided. The results obtained from this investigation would contribute to successful adaptation and establishment of toxicogenomics in our country. Further, our efforts would contribute to establish an international standard protocol that builds a world-wide predictive database regarding genes that induce hepato-toxicity. It also provides valuable information about the molecular mechanisms on tamoxifen-induced hepato-toxicity and related genes, which may lead development of new drugs and strategies for human breast cancer treatment. In addition, our results may contribute to development of a DNA chip which evaluates the hepato-toxic response of compounds in a rapid and sensitive manner.
