본 연구과제는 퇴행성뇌질환의 지표물질과 프리온 단백질 (PrP)의 연관성을 조사하는 것으로, PrP 항체와 PrP에 결합할 것으로 추정되는 단백질에 대한 항체를 이용한 differential ELISA 기술을 이용하여 혈액 내 존재하는 PrP-단백질 복합체를 정량하였다. 또한 기존의 항체 epitope 탐색 기술인 pepspot을 응용하여 PrP의 결합부위를 탐색할 수 있는 modified pepspot 기술을 개발하였다. 알부민이 PrP와 결합하여 존재하는 것을 확인하였고, balb/c 쥐와 PrP Knockout 쥐의 plasma를 differential ELISA 기술을 이용하여 PrP-albumin 복합체의 존재를 증명하였다. 그 결합 부위를 PrP pepspot을 이용하여 탐색해본 결과, PrP의 아미노산 (KKRPKPGGWNTGGSRYPGQG)과 결합하였고, 138-151번 아미노산 (IIHFGSDYEDRYYR)과 결합하는 것을 확인할 수 있었다. β-amyloid (Aβ)의 경우도 혈액 내에서 PrP와 결합하여 존재하는 것으로 확인되었다. 다양한 질병 그룹에서 PrP-Aβ 복합체를 정량하였지만, 특정 질병과의 연관성을 찾지 못하였다. Aβ가 PrP에 결합되는 부위를 검사하였으며 그 결과 PrP 단백질의 23-42번, 89-104번, 147-161번, 244-246번과 결합하는 것으로 추정되었다. 혈액의 응고와 용혈과정에 관여되는 plasminogen (Pgn)은 PrP와 결합하는 것이 많은 논문에서 보고되었으며, 우리의 differential ELISA로 확인이 가능하였다. Pgn이 PrP에 결합하는 부위를 검사해본 결과 PrP의 23-30번, 86-100번, 141-165번의 아미노산 부위와 결합하는 것을 확인할 수 있었다.
본 실험을 통하여 알부민, Aβ, Pgn이 공통적으로 결합되는 PrP의 아미노산 서열을 유추할 수 있었으며, 그 PrP의 부위는 23-40번, 147-151번 부위임을 확인할 수 있었다. 따라서 PrP의 이러한 부위의 특성에 대한 분석과 질병에서의 역할, 그리고 다른 단백질과의 연계성, 수용체로써의 역할 등의 PrP의 생리학적 기능에 관련성이 주목된다.

Aim of the current research was to investigate the interactions between prion protein (PrP) and biomarker proteins in blood from other neurodegenerative diseases in order to understanding underlying pathway of PrP involvement in various neurological diseases. Differential ELISA technique was adopted to capture the PrP/protein complex by using two different antibodies, one for PrP and the second antibody for the complexed protein. The modified pepspot was introduced to find the critical amino acid sequences for binding epitope. Forty-five peptides of PrP(23-230) were synthesized in soild-phase peptide synthesis (SPPS) way for the use of modified pepspot, where cysteine residue was introduced at N- or C-terminus for the cross-link onto the surface.
We found that PrP could bind to albumin. Hence, when the differential ELISA of capturing the PrP/Albumin complex by using anti-PrP antibody as capturing antibody and anti-albumin antibody as the detection antibody, we could capture and detect PrP/Albumin complex in plasmas from human, bovine and mouse (balb/c), not in PrP-knockout mouse. Based on PrP pepspot, albumin binding site on PrP were 23-KKRPKPGGWNTGGSRYPGQG-42 and 138-IIHFGSDYEDRYYR-151. PrP could also bound to Aβ and PrP-Aβ complex could be detected from human plasma. However, among many groups of diseases, the plasmas from chronic renal failure diseases revealed elevated levels of PrP/Aβ complex. Again, based on PrP pepspot, Aβ binding sites on PrP were determined: 23-KKRPKPGGWNTGGSRYPGQG-42, 89-WGQGGGTHSQWNKPSK-104, 147-DRYYRENMHRYPNQY-161, 244-AYY-246. In addition, plasminogen (Pgn), a zymogen of plasmin in fibrinolysis, was suggested to bind PrP. Hence, we tested to capture and detect the complex by using differential ELISA with anti-PrP antibody and anti-plasminogen antibody. PrP/Pgn complex was found in many plasma samples from both normal and patients with various diseases. Therefore, no correlation could be drawn between a disease and the levels of the PrP/Pgn complex. Pgn also bound to PrP on the particular amino acid sequences, as following, 23-KKRPKPGGWNTGGSRYPG-30, 86-GGGWQGGGTHSQWN-100 and 141-FGSDYEDRYYRENMHRYPNGVYYRP-165.
From our research, common PrP amino acid sequences for the binding interactions with albumin, Aβ and Pgn were 23-40 and 147-151. Therefore, pivotal role of these regions of PrP could play for their physiological functions such as disease progressions, PrP amplifications, and cell signaling.
