인간 염색체 21q22.2 지역에서 발현하며 다운증후군 병인 유전자로 제시된 Dyrk 및 Tprd 단백의 기능 및 활성화기전을 연구하는 본 과제는 다음과 같은 4개의 세부 연구테마로 다운 증후군의 분자적 병인 기전을 분석하고 다운증후군 질병의 동물모델을 만들어 임상적 응용의 이론적 토대를 제공 하고자 한다. 연구개발 내용의 세부제목 및 실제 실험 방법들을 간추려 보면 (1) 인간 21번 염색체에서 다운증후군 유발 원인유전자로 알려진 Dyrk 와 Tprd 단백의 기능 연구을 위해 신경세포에서 구축한 cDNA library와 yeast
인간 염색체 21q22.2 지역에서 발현하며 다운증후군 병인 유전자로 제시된 Dyrk 및 Tprd 단백의 기능 및 활성화기전을 연구하는 본 과제는 다음과 같은 4개의 세부 연구테마로 다운 증후군의 분자적 병인 기전을 분석하고 다운증후군 질병의 동물모델을 만들어 임상적 응용의 이론적 토대를 제공 하고자 한다. 연구개발 내용의 세부제목 및 실제 실험 방법들을 간추려 보면 (1) 인간 21번 염색체에서 다운증후군 유발 원인유전자로 알려진 Dyrk 와 Tprd 단백의 기능 연구을 위해 신경세포에서 구축한 cDNA library와 yeast two hybrid 분석 방법을 이용하여 신경계에서 Dyrk와 Tprd 단백과 상호 결합하는 단백을 확인하고 확인된 유전자중 기능이 알려진 단백을 확인하여 그들과의 연관된 기능 분석을 통해 두 다운 증후군 모델 단백의 이들 세포 내에서 역할을 분석하였다. (2) 일차 및 신경세포주 모델에서 tprd 및 dyrk 유전자들의 태아 및 성인세포의 성장, 분화, 사멸에 미치는 효과 및 활성화 신호전달 기전을 연구하기 위해 dual-specific protein kinase을 암호화하는 Dyrk 단백의 경우 3가지 세포들에서 기능 및 활성화 기전을 kinase-inactive mutant expression vector와 다양한 신호전달 경로상의 화학적 방해제 및 활성화단백의 항체를 이용하여 확인하고 tprd 유전자의 경우 단백질을 과발현 시켰을때 세포의 성장과 분화, 사멸에 미치는 효과를 검색하였다. (3) 다운증후군 병인단백의 과발현 세포모델에서 신호전달 경로의 이상유무 검색 및 검색법 개발을 목적으로 다운증후군 연구모델 정립을 위한 Dyrk1A 단백의 과발현 세포주를 구축하고 구축 Dyrk1A 단백의 과발현 세포주에서 헌팅톤병 및 알쯔하이머병의 병인단백과의 연관성을 규명하였음 (4) 다운증후군의 후보 유전자중 Tprd 단백의 형질변환 동물모델의 개발에 필요한 벡터구축을 시도하고 추후 후속 연구를 통해 유전학적, 세포생물학적 실험방법을 이용, 후보 유전자의 돌연변이 효과를 비교할 예정이다.

(1) Introduction and Research Aims: Down syndrome (DS), the most common chromosome aberration, is caused by trisomy of chromosome 21. DS manifests complex phenotypes, including a characteristic facial appearance, mental retardation, neonatal hypotonia, congenital heart defects, acute leukemia, and abnormalities of the gut and endocrine system. In addition, all individuals with DS exhibit Alzheimer disease-like neuropathology by age 30-40 years. The cytogenetic and clinical correlations of patients with partial trisomy 21 indicate that one or multiple gene(s) in a region of 2-4 Mb in 21q22.2, designated as the Down syndrome-critical region (DCR), may contribute to the pathogenesis of DS. Thus, the characterization of genes in the DCR are important for elucidating the molecular basis of DS. Minibrain (Mnb) is a mutant of Drosophila whose presence is exhibited by a specific and marked size reduction of the optic lobes and central hemispheres in the adult brain. The Mnb gene encodes a Ser/Thr protein kinase that possesses a Tyr-X-Tyr sequence in the activation loop. At least 7 closely related homologous mammalian kinases have since been isolated and a novel superfamily of protein kinases called Dyrk(s) have been established. Dyrks possess Ser/Thr phosphorylation activity as well as autophosphorylation activity on Tyr residues, suggesting that Dyrk seems to be a dual specificity kinase. The kinase activity of Dyrk is dependent on the Tyr-X-Tyr motif in the activation loop, suggesting the existence of a phosphorylation-dependent activation mechanism of Dyrk by certain upstream kinases. Interestingly, human Dyrk1A is mapped to the DS critical region on chromosome 21, and thus could be a candidate gene responsible for the mental retardation of DS patients. Thus, from Drosophila to humans, it is suggested that Dyrk/Mnb is a key regulator of normal embryo neurogenesis and brain development. Although the exact cellular function of the Dyrk kinases is still unknown, an understanding of the physiological role(s) of this protein kinase family will prove to be very relevant. TPRD, which belongs to the tetratricopeptide repeat (TPR) gene family from the DCR, is expressed in all the human fetal and adult tissues examined. TPRD encodes a putative protein (TPRD) of 2,025 amino acid residues. Although the physiological function of TPRD has not been determined, protein database searches implied the TPRD gene encodes a matrix protein having some function in the nucleus and that the overexpression of the mtprd gene may be involved in several morphological anomalies observed in DS. The participation of Dyrk1 or other Dyrk-related kinases in any particular signal transduction pathway has not been elucidated so far.
(2) Results: The selective recognition of the correct substrate by protein kinase is an important biochemical mechanism that underlies the specificity of cellular responses to various stimuli. It is therefore important to determine the nature of Dyrk substrate in order to understand its function in the regulation of cellular responses. Previously, Dyrk was shown to be required for the proliferation of distinct neuronal cell types during postembryonic neurogenesis in Drosophila. Mutant flies with a reduced Dyrk1 expression have a reduced number of neurons in distinct areas of the adult brain and exhibit specific behavioral defects. In the present study we examined the cellular Dyrk1 substrate and its functional role during neuronal differentiation in hippocampal H19-7 cells. We have shown that Dyrk1 is activated by neurogenic bFGF, but not by mitogenic EGF, and that active Dyrk1 stimulates CREB phosphorylation and subsequent CRE-mediated gene transcription. Our data strongly suggest that Dyrk1 may play an important role during neurogenic factor-induced differentiation in CNS neuronal cells. Such a role was further supported by the finding that the expression of kinase-deficient Dyrk1 in a transient manner remarkably attenuates the formation of differentiated cells. The Dyrk1 protein contains several striking structural features, such as a bipartite nuclear localization signal, a PEST region, repetitively present 17-serine/threonine residues and an activation loop (YQY motif) between subdomain VII and VIII. Dyrk1 is localized to the cell nucleus, hence gaining the potential to control the expression of other genes. A GFP-Dyrk1A fusion protein was found in the nucleus of transfected COS-7 or HEK293 cells.
