(2) 해마의 새로운 신경세포 생성에 미치는 효과 측정 BrdU 염색법으로 새로이 생성되는 해마 신경세포를 선택적으로 염색하여 PPD, PPT가 새로운 해마 신경세포 생성에 미치는 영향을 측정하였다. (3) 공간학습에 미치는 효과 측정 4가지 다른 농도의 PPD, PPT를 실험용 생쥐에 투여한뒤 Morris water maze에서의 공간학습에 미치는 영향을 측정하였다. (4) 해마 신경세포 생리에 미치는 효과 측정 해마 절편과 intracellular recording 방법을 이용하여 기본적인 신경생리적 지표에 미치는 영향을 측정하였다. 막전위, 막저항, membrane time constant, spike threshold, spike size, spike adaptation, EPSP 크기, IPSP 크기, NMDA response의 크기, paired-pulse facilitation에 대한 PPD와 PPT의 효과를 규명하였다. (5) 해마 시냅스 가소성에 미치는 효과 측정 뇌절편과 extracellular field potential recording 방법을 이용해 PPD와 PPT가 고빈도 자극에 의 한 시냅스 장기강화(long-term potentiation)에 미치는 영향을 측정하였다. <연구결과> (1) PPD와 PPT는 해마의 새로운 신경세포 생성에는 유의한 효과를 미치는 않았으나 기존 신경계의 시냅스 재생을 유의하게 촉진함을 발견하였다(농도 1 mg/Kg). (2) PPD와 PPT를 동물에 투여했을때 공간학습 능력이 유의하게 증가하였다(농도 1 mg/Kg). (3) PPD와 PPT는 모든 투여군에서 막저항 등 세포생리 지표를 변화시키지 않았으나, 1 mg/Kg 이상의 농도에서 input-output curve의 증가, 그리고 paired-pulse facilitation의 감소를 유도함이 밝혀졌다. (4) PPD와 PPT는 강한 고빈도 자극을 사용했을 때 시냅스 장기강화(long-term potentiation)에 유의한 영향을 미치지 않았다. (5) 이상을 종합할 때 실험용 생쥐의 경우 1 mg/Kg의 농도에서 별 부작용 없이 신경계 재생 및 학습능력을 증진시킨다고 결론지을 수 있다.

Memory deficit among senile population is a significant socio-economic problem in Korea which has become an aging society. Cognitive enhancers that are available or being developed treat symptoms rather than enhancing functions of the hippocampus which is an essential structure for explicit memory. On the other hand, because ginseng extracts are known to promote synaptogenesis and enhance cognitive capacity, it is possible to develop new cognitive enhancers by investigating ginseng effects. I have previously found that two ginsenosides, protopanaxydiol (PPD) and protopanaxytriol (PPT), readily pass blood-brain barrier while enhancing hippocampal synaptogenesis and behavioral learning. The aim of the present project is to systematically investigate effects of PPD and PPT on hippocampal synaptogenesis, hippocampus-dependent learning, and neurophysiology, so that these components can be developed as new drug candidates. (1) Effect on hippocampal synaptogenesis We injected 4 different concentrations of PPD and PPT, and then quantified hippocampal synaptic density by measuring synaptophysin levels. (2) Effect on hippocampal neurogenesis We quantified hippocampal neurogenesis by counting the number of BrdU-staining cells in the hippocampus. (3) Effect on spatial learning We measured spatial learning of mice on a Morris water maze after injecting PPD and PPT. (4) Effect on hippocampal cellular physiology We measured effects of PPD and PPT on physiological indices of hippocampal neurons including resting membrane potential, membrane resistance, time constant, spike threshold, spike size, spike adaptation, EPSP size, IPSP size, NMDA response size, and paired-pulse facilitation. (5) Effect on synaptic plasticity We measured effect of PPD and PPT on high-frequency stimulation-induced long-term potentiation in hippocampus using field potential recordings. (1) PPD and PPT did not enhance hippocampal neurogenesis, but increased syantaptogenesis at 1 mg/Kg. (2) PPD and PPT enhanced spatial learning at 1 mg/Kg. (3) PPD and PPT did not alter cellular physiological indices at all concentrations, but, at concentrations equal to or higher than 1 mg/Kg, increased input-output curve and decreased paired-pulse facilitation. (4) PPD and PPT did not affect high-frequency stimulation-induced long-term potentiation. (5) These results lead to the conclusion that PPD and PPT enhance hippocampal synaptogenesis and learning ability without much side effects at 1 mg/Kg dose.
