연구의 목적 및 내용:
ATP 의존성 단백질 분해 조절 시스템에 대한 구조-기능 연구를 통해 원핵세포 및 진핵세포에서의 단백질 분해 시스템의 활성 조절, 기질 인지 및 작용 메커니즘의 이해와 단백질 상호작용의 인지메커니즘의 규명을 목표로 함. 원핵생물의 주 연구 대상은 B.subtilis HslUV(CodWX), S. aureus HslUV 등 Pre-mature protease와 H. pylori, E. coli, T.maritima FtsH 등 Membrane-bound protease이며, 진핵생물에서는 UBA, NZF,
연구의 목적 및 내용:
ATP 의존성 단백질 분해 조절 시스템에 대한 구조-기능 연구를 통해 원핵세포 및 진핵세포에서의 단백질 분해 시스템의 활성 조절, 기질 인지 및 작용 메커니즘의 이해와 단백질 상호작용의 인지메커니즘의 규명을 목표로 함. 원핵생물의 주 연구 대상은 B.subtilis HslUV(CodWX), S. aureus HslUV 등 Pre-mature protease와 H. pylori, E. coli, T.maritima FtsH 등 Membrane-bound protease이며, 진핵생물에서는 UBA, NZF, UIM 및 VHS 도메인 등을 포함한 poly-ubiquitin binding protein에 대한 구조-기능 연구를 수행하였음
연구결과:
■ Pre-mature protease에 대한 구조-기능 연구
○ B. subtilis CodW와 S. aureus HslV의 부분 또는 전체 구조 분석: 활성 부위의 구조-기능적 특성 규명
○ H. pylori, E. coli, T. maritima 전체 FtsH의 분리 정제, H . pylori FtsH의 ATPase, T.maritima periplasmic domain의 구조 분석: 막 단백질의 분해 기전 규명
■ Poly-ubiquitin binding protein에 대한 구조-기능 연구
○ E2-25K 단백질의 단독 또는 ubiquitin / UBB+1 단백질과의 복합체 구조 및 활성 분석:
각 단백질의 전체 및 결합 부위의 구조적 특성, 단백질 간 상호작용이 E2-25K의 ubiquitin 인지 기전과 신경 독성에 미치는 영향 등 규명
○ Poly-ubiquitin의 합성: di-ubiquitin의 분리 정제에 성공함으로써 E2-25K에 의한 polyubiquitination 기전 연구의 교두보를 마련하였으며, 현재 E2-25K와의 복합체 결정화 진행 중
○ STAM 단백질의 VHS 도메인의 분리 정제: ubiquitin 결합 도메인 중 VHS 및 UIM 도메인의 ubiquitin 인식에 대한 구조-기능 연구를 가능케 하였으며, 현재 복합체 생성 진행 중
연구결과의 활용계획:
본 연구를 통해 규명된 세포 내 ATP 의존성 단백질 분해 효소의 활성화, 표적 분해 및 활성 조절 기전과 ubiquitin-proteasome 시스템에서의 기질 인지 메커니즘은 새로운 단백질 분해 및 조절 기전을 분자 수준에서 이해하는 데 지대한 영향을 끼칠 것으로 판단되며, 원핵생물 중 H . pylori, S. aureus 등의 병원성 미생물이 매개하는 질병 또는 인체내 비정상적인 단백질의 축적에 의해 발생하는 각종 질병을 치료할 구조-기반 신약 개발에 대한 중요한 정보로 유용하게 활용 가능함

Purpose & contents:
We performed structural and functional studies of ATP-dependent protease system in order to reveal activities, recognitions of substrates, regulations, and protein-protein interactions related to the targets as pre-mature protease from B.subtilis and S. aureus, membrand-bound protease FtsH from H. pylori, E. coli, and T. maritima, and poly-ubiquitin bound proteins having UBA, NZF, UIM, and VHS domain such as E2-25K.
Result:
■ Structural and functional studies on pre-mature proteases
○ Determination of structure and function of active site of CodW from B. subtilis and HslV from S. aureus
○ Elucidating protein degradation mechanism of membrane-bound protease FtsH from H. pylori, E. coli, and T. maritima using ATPase and periplasmic domain
■ Structural and functional studies on poly-ubiquitin binding protein
○ Analyzing structures and activities of E2-25K single protein and in complex with ubiquitin and UBB+1: determining features of overall structures, modes of binding, reconition of ubiquitin and neurotoxicity via UBB+1
○ Synthesis of poly-ubiquitin: basis of mechanism of poly-ubiquitination (crystallization of E2-25K in complex with di-ubiquitin is ongoing)
○ Purification of VHS and UIM domain of STAM : now complex formation is ongoing
Expected Contribution:
Structural and functional informations related to mechanisms of acvitities, degradation of substrate, regulation, of recognition of substrate in ubiquitin-proteasome system are expected to bring insights of novel protein degradation and its regulatory mechanism. Moreover, these informations will be used to develop new drug using structure-based drug design against various diseases caused by malfunction of ubiquitin-proteasome system and pathogenic bacteria including H. pylori and S. aureus.
