Buckwheat is grown throughout a large area of Asia and Southeast Asia as a crop. Buckwheat has a well-balanced amino acid composition that is complementary to cereal grains. Fagopyrum is a genus of about 15 species. common buckwheat (Fagopyrum esculentum L.) and tatary buckwheat (Fagopyrum tataricum G.) are the most important species in the genus. Many studies on tatary buckwheat have been trying recently, especially for improving higher nutritional value of common buckwheat. Tatary buckwheat probably originated in northwestern Yunnan in China, and this species is now cultivated in the high-altitude mountainous areas of Asia and to a much lesser extent elsewhere in Europe. The flowers are homomorphic and self-fertile, with pollination occurring before the flower opens. Studies on tatary buckwheat were increased due to tatary buckwheat containing many micro-elements, vitamins and vitamin B, and being especially rich in antioxidants, such as rutin and quercetin. Rutin in buckwheat is a kind of flavonol glycoside compounds used in preventing edema, haemorrhagic diseases, and stabilizing high blood pressure due to its effectiveness in controlling blood vessel. This experiment was to select tatary buckwheat lines with high yield and rutin content. We evaluated 75 tatary buckwheat lines(or varieties) at lowland and highland on summer and autumn, respectively. There were a positive correlation between branch number and flower clusters and yield. Onthe other hand, plant height and days to flowering were shown negative correlation with yield. 9 lines among the selected 12 lines with high yield were shown higher rutin content than standard cultivar. 36 F1 hybrids obtained at 125 crossing combinations. After evaluation of agronomical traits of F1 of 36 crossing lines, 9 lines of summer type and 14 lines of autumn type.
Phytochemical constituents were isolated from the seeds of Fagopyrum tataricum (Polygonaceae) by open column chromatograph. Their structures were elucidated as β-sitosterol, β-sitosterol-3-O-glucoside, oleanolic acid, kaempferol, quercetin, kaempferol-3-O-rutinoside, and quercetin-3-O-rutinoside. The protective effects from oxidative stress of fractions and rutin from Fagopyrum tataricum were evaluated. The treatment of n-hexane, methylene chloride, ethyl acetate, and n-butanol fractions from F. tataricum showed over 90% scavenging effect against DPPH at 500μ g/ml. In addition, the fractions showed over 80% ㆍOH scavenging activity at the concentration of 50μg/ml. Moreover, rutin showed over 80% scavenging effect against DPPH at 50μg/ml. ㆍOH scavenging activity of rutin showed over 80% at 10μg/ml. Furthermore, the treatment of H2O2 to C6 glioma cell induced oxidative stress, however the fractions from F. tataricum and rutin increased significantly the cell viability. Protective effects of Tartary buckwheat (TB) and common buckwheat (CB) on amyloid beta (Ab)-induced impairment of cognition and memory function were investigated in vivo in order to identify potential therapeutic agents against Alzheimer’s disease (AD) and its associated progressive memory deficits, cognitive impairment, and personality changes. An in vivo mouse model of AD was created by injecting the brains of ICR mice with Ab25–35, a fragment of the full-length Ab protein. Damage of mice recognition ability through following Ab25–35 brain injections was confirmed using the T-maze test, the object recognition test, and the Morris water maze test. Results of behavior tests in AD model showed that oral administration of the methanol (MeOH) extracts of TB and CB improved cognition and memory function following Ab25–35 injections. Furthermore, in groups receiving the MeOH extracts of TB and CB, lipid peroxidation was significantly inhibited, and nitric oxide levels in tissue, which are elevated by injection of Ab25–35, were also decrease. In particular, the MeOH extract of TB exerted a stronger protective activity than CB against Ab25–35-induced memory and cognition impairment. The purpose of this study is to elucidate the molecular mechanism of anti-inflammatory effect of the fractions (n-hexane, methylene chloride, ethyl acetate, and n-butanol) and active compound, rutin, which was isolated from Fagopyrum tartaricum. We investigated the protective activity of the fractions and rutin against NO-induced oxidative damage by sodium nitroprusside (SNP) in RAW264.7 macrophages. We further investigated the effects of the fractions and rutin on the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and nuclear factor- κB (NF-κB) under RT-PCR in SNP-stimulated RAW264.7 cells. The fractions and rutin suppressed the NO production and down-regulated the levels of iNOS, COX-2 and NF-κB mRNA expressions. The present study indicate that the fractions and rutin from F. tartaricum showed protective activity against oxidative damage induced by NO hrough regulation of its related mRNA expressions such as iNOS, COX-2 and NF-κB.
