<연구개발 목적>
국내외 생산된 버섯에 대한 유통기준을 설정하기 위하여 버섯 수확후 관리기술 및 수출수송-유통 환경관리 설정을 위한 기초자료 확보, 버섯품질 및 등급 결정에 필요한 기초자료 작성, 수출버섯의 안전성 향상을 위한 유해물질 안전관리기준설정, 생산에서 소비까지의 환경관리 매뉴얼화 및 프로그램 개발 대한 연구를 수행하였다.
<필요성>
우리나라의 버섯은 대량생산 체계가 도입된 상태로 국제경쟁력이 강화되었으며, 특히 병재배 버섯(팽이, 큰느타리)이 버섯수출에 주도적 역할을 하고 있으며, 2009년 수출액 36,585천$ 중에 60.7%를 점유하고 있다. 하지만 이 수출버섯 품목이 수출과정에서 저장상에 문제가 되어 근본적인 문제를 해결하기 위한 저장기간 결정요인 분석 및 재배법 개선에 의한 저장기간 증대에 필요한 가술의 개발이 요구되고 있다. 특히 버섯수확후 관리기술 및 수출수송-유통 환경관리 설정과 버섯품질 및 등급을 결정할 수 있는 기초자료의 확보와 수출버섯의 안전성 향상을 위한 유해물질 안전관리기준설정 보완, 수출주도 버섯의 국제경쟁력 확보 및 경영비 절감을 위한 생산에서 소비까지의 환경관리 매뉴얼 및 프로그램 개발이 절실한 상황이다.

Ⅳ. 연구개발결과
1. 버섯 저장기간에 따른 변화 요인 분석 및 미생물 상에 관한 연구
○ 저장요인별 자실체의 경도 및 형태적 변화
팽이버섯
저장기간에 따라 형태적으로 갓과 대길이는 증가하는 경향이며, 갓과 대경도는 감소하는 경향을 보였다.
큰느타리버섯은
대경도는 2주까지는 증가하였다가 감소하였으나, 갓경도는 저장기간에 증가함에 따라 감소하였다.
형태적
특징에서는 큰 차이는 없었으나 대굵기, 대길이 등에서 품종 및 저장기간에 따라 차이가 있었다.
자실체의
갓과 대의 색은 큰 변화가 없었으며, 갈변 증상이 부분적으로 발생하였다.
팽이버섯은
저온처리에 의한 생장 억제방법을 사용하면 수량 및 개체수가 증가하였으며, 형태적 특성은 처리에 따른 변화를 확인할 수 없었다.
팽이
및 큰느타리버섯은 품종에 따라 저장성의 차이가 있으며, 큰느타리버섯은 ASI 2887 균주가, 팽이버섯은 F4과 F10번이 우수하였고, F1, F8, F9 균주는 불량하였다.
○ 저장요인별 자실체의 미생물상 조사 및 분리미생물의 분류동정
버섯자실체에서
발생하는 세균중에서 병원성을 나타내는 균주는 Ewingella americana, Pseudomonas tolaasii, Pseudomonas fluorescens이며, 이 세균은 저장중의 자실체 갈변의 주요인으로 추정된다.
그
외에 저장중의 버섯에는 Chryseobacterium indologenes, Stenotrophomonas maltophilia, Serratia grimesii 등이 발생빈도가 높았으며, 이들은 토양, 물 등 주변에서 쉽게 발견할 수 있는 미생물로 알려져 있다.
2. 버섯의 품질 및 등급 규정 수립을 위한 연구
○ 팽이버섯의 품질 규격
유통되고
있는 팽이버섯의 등급별 품질규격은 품질등급이 높을수록 갓의 직경이 작았다.
구입한
팽이버섯의 경우 A등급과 B등급은 갓의 지름이 1.0cm 이상인 버섯이 없어 특품에 해당되었으나, C등급은 갓의 지름이 1.0cm 이상이 평균 6개로 상품에 해당되었다.
○ 양송이의 품질 규격
유통되고
있는 양송이버섯의 등급별 품질규격은 A등급의 자실체의 평균무게가 19.3g, B등급은 12.2g, C등급은 10.4g 이었다.
양송이버섯
수확시 자루절단에 따른 평균 중량 감소율은 12.6%였으며, 절단 길이비율은 60.8%이었다.
○ 느타리버섯의 품질 규격
유통되고
있는 느타리버섯의 등급별 품질등급은 균상재배에서 갓의 직경과 두께는 38.9mm과 4.5mm였으며, 병재배 버섯은 22.2mm와 3.9mm이었다.
균상재배
버섯은 등급내에서 갓의 모양, 고르기 등에 차이가 없고 포장시 배열이 일정하게 되어 있었다.
○ 큰느타리버섯의 품질 규격
큰느타리버섯
재배농가에서 고품질 생산을 위한 품질관리 기준은 특품의 평균개체중은 90g 이상, 대굵기는 50mm 이상, 대길이/대굵기 비율은 1.6∼1.7, 갓직경/대굵기 비율은 1.1∼1.2이며, 상품의 평균개체중은 45g 이상, 대굵기는 40mm 이상, 대길이/대굵기 비율이 1.8∼1.9, 갓직경/대굵기 비율이 1.2∼1.4 이었다.
3. 버섯 장기 저장 실용화 기술개발 및 버섯수확후 처리기술의 매뉴얼화 연구
○ 큰느타리버섯 수확후 장기 선도유지 기술 확립 : 3 ⇒ 5주

4. 버섯 재배방법 별 유해요인 분석 및 관리기준 설정
○ 양송이버섯에서의 유해미생물의 변화
양송이
배지의 발효에 따른 미생물상은 후발효후 고온성 세균과 형광성 Pseudomonas속 그리고 방선균의 수가 증가하였지만 사상균과 푸른곰팡이균는 검출되지 않았다.
- 양송이 배지에서 분리한 미생물을 동정한 결과 Bacillus 속이 대부분을 차지하였고, 병원성세균은 E. coli, Staphylococus sp. Bacillus sp. 가 분리되었다.
- 다양한 항생제에 내성을 가지는 것으로 확인되었으며, 이들 균을 동정한 결과 Stenotrophomonas humi, Pseudomonas libanensis 등 다양하였다.
○ 유통버섯에서의 유해미생물의 변화
- 유통버섯 종류별 자실체의 미생물상을 조사한 결과 팽이에서 세균과 곰팡이가 가장 많이 검출되었고, 큰느타리에서 가장 적게 검출되었다.
- 양송이 재배를 위한 적정 살균온도는 80℃-60분, 80℃-90분이 가장 좋았다.

1. Changes in hardness and morphological features of mushroom based on the storage strategies.
In Flammulina velutipes, the pileus and stipe were increased in length, while their hardness were decreased during the storage. In Pleurotus eryngii, the hardness of stipe was increased until two-week storage, and gradually decreased, whereas that of pileus was consistently decreased. The morphological changes were not significant. But, the thickness and length of stipes were significantly changed according to the species and the period of storage. Significant changes in color of pileus and stipe were not recognized, but partially browning reaction was found. In Flammulina velutipes, the artificial growth inhibition test using a cold-treatment improved the yield. However, this treatment did not influence the morphological features. According to the cultivars, the quality for storage was turned out to be different. Pleurotus eryngii ASI 2887 strain as well as Flammulina velutipes F4 and F10 strains represented good storage ability, while F1, F8, and F9 strains were comparatively poor.
2. Microbial diversity on cultivated mushroom during the storage
Among the microbes identified on mushroom, Ewingella americana, Pseudomonas tolaasii, and Pseudomonas fluorescens were pathogens which induced the browning reaction on fruiting bodies of stored mushroom. Chryseobacterium indologenes, Stenotrophomonas maltophilia, and Serratia grimesii, which are common microorganisms in the natural environment such as soil and water, were also frequently isolated in stored mushrooms.
○ The study for standardization of the quality and grade of various mushrooms This study was carried out to improve standardization of agricultural products and investigate quality change during preservation at various treatment. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Mushrooms notified as the standard are five items; Pleurotus ostreatus, Pleurotus eryngii,
Flammulina velutipes, Agaricus bisporus and Ganoderma lucidum. But many farmers are suffering from a strict standards. So these standard is required modification to fit farmhouse situations. The higher quality grade of mushrooms, the smaller a dimeter of mushroom pileus in the circulation Flammulina velutipes. Average pileus diameter of A grades of Flammulina velutipes was 4.1mm, B grades 4.2mm and C grades 6.5mm, respectively. The L value and a-value of pileus were 58.0∼60.1 and 0.336∼0.347. The L value and a-value of stipes were 56.2∼57.4 and 0.331∼0.335. Fruit-body weight of A, B and C grades of the circulation Agaricus bisporus were 19.3g, 12.2g and 10.4, respectively. Average pileus diameter of A grades was
43.3mm but B and C grades showed significantly each other. Average stipes length of A, B and C grades were 9.8mm, 13.1mm and 11.6mm, and stipes thick was the highest in A grades. The weight loss rate of cutting stipes of Agaricus bisporus was
12.6% in harvested mushrooms and the rate of cutting stipe length was 60.8%. The diameter of Agaricus bisporus was stored for 5 days was 3.5~4.5cm but decreased rapidly in 15 days. Pileus diameter and thick of Pleurotus ostreatus at shelf culture
were 38.9mm and 4.5mm and at bottle culture 22.2mm and 3.9mm. Stipe length was 66.7mm in bottle culture and 49.6mm shelf culture. L-value and hardness of stipes an pileus were higher at shelf culture than bottle culture. We propose a
standardization for production of Pleurotus eryngii of high quality. A grades were more 90g at average weight, more 50mm at stipe thick, 1.6∼1.7 at stipe length/stipe thick rate and 1.1∼1.2 at pileus diameter/stipe thick rate. B grades were more 45g at average weight, more 40mm at stipe thick, 1.8∼1.9 at stipe length/stipe thick rate and 1.2∼1.4 at pileus diameter/stipe thick rate.
○ Development of long-term storage technology and postharvest handling manual of export mushrooms
The market in Korea for fresh mushrooms has increased over recent years due to greater consumer awareness of products. Mushrooms have high growth potential in domestic and export markets. Further growth in the export market is presently being constrained by poor postharvest handling practices and storage practices. This report reviews current industry practices and information relating to postharvest handling of mushrooms (king oyster mushroom enoki mushroom) and , developing appropriate handling protocols and identifying research requirements.
This research report the results of a experiment of the king oyster mushroom and enoki mushroom which increase shelf-life. Quality and sensory evaluation parameters of weight loss, stem firmness, skin color, off-flavor, overall quality were monitored during storage. Pre-cooling and low temperature maintenance is requires during storage and distribution. Postharvest applications of 1-MCP, CO2 pre-treatment and MAP with microperforated film improve king oyster mushroom quality (stem color, decay, and off-odor) and shelf-life. We tried to identify physiology of mushroom after the harvest, storage method, and various techniques to optimize quality during distribution.
○ Establishment of management standard on harmful substance and damage according to the cultivation method of mushrooms
This study is carried out analysis harmful factor and to establish the safety standard for agricultural mushroom, Agaricus bisporus. As fermentation of Agaricus cultural medium, it is increased thermophilic bacteria, fluoresence Pseudomonas and actinomyces later after-fermentation in its microflora, however, filamentous fungus and pinicillia are not founded. Microorganisms, which are separated from Agaricus culture medium, are almost Bacillus and a few pathogenicity, E. coli and Staphylococus sp. Bacillus sp. During fermentation of Agaricus cultural medium, various microorganisms are separated and investigated. And the results are that they, which is Stenotrophomonas humi, Pseudomonas libanensis etc., have tolerances to diverse antibiotic excluding enrofloxacin, which became an objct of public concern that founded from chicken lately and forbidden as FDA regulation, and Ciprofloxacin. Results from investigating different mushroom fruit bodies in distribution are that Flammulina velutipes has much virus and mold while Pleurotus eringii has little. After identification for organisms from Agaricus fruit bodies, Chryseobacterium is dominance. And After checking into before and after of washing organisms, virus and mold are decreased. It is detected that there are not found more standard amount of pesticides from almost collected mushroom farm, but Dimilin and Nomolt are found around standard amount from two farms after researching pesticide residues of gathered mushrooms. Changes in density of organisms in molding ingredients are as follows. Changes of Aerobic bacteria are increased as the longer grow-out period and sharply increased after second culture. Density of Fluorescence Psuedomonas is crammed at high temperatures and compared with other treatment plot 100℃ treatment plot has extremely high density at 30min and 60min in 22th day after covering with soil. Density of thermophilic actinomyces is sharply increase from covering with soil and the highest density at 22th day of covering and rapidly decrease after first culture. Sterilizing temperature covering with soil affects quality and quantity of Agaricus. 60min, 90min at 80℃ and 30min at 100℃ treatment produce the highest mushroom yields, especially 90min at 80℃ generate the best mushroom yields in good products and fair products. 60min at 100℃ treatment products many quantities, however, this treatment has low economic feasibility for its yields are poor products. Sterilizing temperature covering with soil has an effect on generating diseases and insect pests. 60min, 90min at 80℃ and 30min 100℃ lead to low attack rate regarding other treatment plot. Although 30min at 100℃ cause low diseases and insect pests, has low Agaricus yields. And 60min at 100℃ cause high diseases and insect pests even if it bring about high Agaricus yields. Therefore proper conditions for agricultural Agaricus are 60min, 90min at 80℃.
