연구의 목적 및 내용
본 연구는 퇴행성 뇌질환에서 공통적으로 관찰되는 단백 변형 및 응집 구조물 생성으로 야기되는 신경계 변병의 새로운 치료 기반 구축을 위해, 몇 개 타깃 단백의 응집물 생성 및 조절 현상을 연구하고, 다양한 단백 수식화 및 분해 시스템 조절을 통해 이들 응집 구조물 생성을 억제하거나 분해를 촉진하는 다양한 제어기술 개발의 기반 구축을 목표로 함.
연구결과
- 본 연구에선 다음과 같은 4개 세부 주제로 나누어 지난 5년간 연구를 수행하였음.
(1) 퇴행성 뇌질환 유발 병인 단백의 변형, 응집
연구의 목적 및 내용
본 연구는 퇴행성 뇌질환에서 공통적으로 관찰되는 단백 변형 및 응집 구조물 생성으로 야기되는 신경계 변병의 새로운 치료 기반 구축을 위해, 몇 개 타깃 단백의 응집물 생성 및 조절 현상을 연구하고, 다양한 단백 수식화 및 분해 시스템 조절을 통해 이들 응집 구조물 생성을 억제하거나 분해를 촉진하는 다양한 제어기술 개발의 기반 구축을 목표로 함.
연구결과
- 본 연구에선 다음과 같은 4개 세부 주제로 나누어 지난 5년간 연구를 수행하였음.
(1) 퇴행성 뇌질환 유발 병인 단백의 변형, 응집 현상 및 조절 기전 연구
(2) 다양한 단백 수식화 및 매개 효소의 특성 및 활성 조절을 통한 단백 응집 조절 연구
(3) 세포 내 다양한 단백 분해 효소 및 분해 시스템 조절을 통한 단백 응집 조절 연구
(4) 단백 응집 매개 효소 Transglutaminase의 활성 조절을 통한 단백 응집 및 조절 현상 연구
- 지난 5년간 이들 각 세부 주제별로 여러 측면에서 다양한 단백질 응집 현상을 활발히 연구하였으며, 항목 별 주요 연구 성과로는 먼저 (1)항 퇴행성 뇌질환 병인 단백 변형에 의한 응집 및 조절 측면에서 알츠하이머병의 타우 및 아밀로이드 단백 변형 및 응집이 Dyrk1A 효소에 의해 매개 됨을 규명하였고 파킨슨병에서 루이소체 주요 구성인자인 alpha-synuclein, Synphllin-1, Parkin, PINK1, LRRK2의 다양한 변형 효과 등을 연구하였음. (2)항 단백 응집에 대한 수식화 효과 연구에서는 Phosphorylation, Ubiquitination, Sumoylation, Neddylation 등의 수식화 모드에 의한 세포 생리 효과 및 매개 효소의 특성 연구에 집중하여 이전에 규명되지 않았던 다양한 단백 봉합체·응집물 조절 및 제거 현상을 새롭게 규명하였음. 좀더 세부적으론 (가) 인산화 효과에 의한 단백 변형, 응집, 분해 효과를 연구하기 위해 ASK1, Dyrk1A, LRRK2, PINK1 등 4종 이상의 효소 효과를 집중 연구하였으며, (나) Ubiquitination 영향 분석을 위해 Parkin, CHIP, SIAH 등에 의한 유비키틴화 효과를 연구하였고, (다)항 Sumoylation 효과를 위해 PIAS family와 Pc2 등의 효과 및 alpha-synuclein, ZNF131 등 새로운 기질 및 조절 인자 등을 동정하였음. 또한 (라) Neddylation 효과는 NEDD8 첨가 효과 및 RBX1의 새로운 기질 로 RCAN1 등의 활성 제어 기전 등을 연구하였음.
- (3)항의 단백 분해 시스템 조절을 통한 응집 제어 기술 개발을 위해 Parkin 및 ASK1 등에 의한 프로테아좀의 양성 및 음성 제어 현상을 처음으로 규명하였고, 최근 새로운 제어 인자로 신경계에서 아직 정확한 기능이 규명되지 않은 텔로머레즈 단백 파트를 확인하여 그 분자 작용기전을 현재 분석 중에 있음. 마지막으로 (4)항 단백 응집 매개 효소인 TG2 연구에서는 새로운 TG2에 의한 응집 및 변형의 기질 단백과 활성 제어 인자를 3종을 동정하여 그 작용기전연구를 완료하였으며 현재 논문 발표를 준비중임
- 5년 동안의 연구 수행을 통해 2012년 6월 1일 현재 연구비 사사된 교신저자 논문을 총 22편 (SCI 등재 국제 학술지에 21편, 연구재단 등재 국내 학술지에 1편) 발표하였음. 또한 본 연구 주제와 밀접하게 연관되어 국제 및 국내 공동 연구를 통해 과제 번호가 사사되지 않은 9편의 공저자 논문을 추가로 SCI 등재 국제 학술지에 발표하였음. 이외에 국제 및 국내 학술대회에 총 58편의 초록 발표와 보건의료부분 보건복지부 표창 우수연구자 수상 실적 1회, 외국 과학자 유치 1건, 학위 배출 10 건의 추가업적을 거두었음.
연구결과의 활용계획
- 단백 구조 변형, 응집, 폴딩 현상 등 단백질 화학의 전반적 이해와 응용성 증진 기반을 제공
- 단백 응집화 현상과 아밀로이드 및 봉합체 생성의 분자 기전의 정보를 제공
- 퇴행성 뇌질환 병인 단백의 응집 구조 생성의 분자적 제어를 통한 병변의 새로운 치료 기반 기술을 구축함
- 병인 단백의 변형 제어를 통한 퇴행성 뇌질환의 새로운 치표후보물질의 발굴 및 효능 탐색을 도모함
- 참여 연구원들로 하여금 신경과학, 생리학 및 병리학 등의 기초 의학과 생화학 및 생물학 등의 기초 과학 분야에서 다양한 연구 방법 및 분석 기술을 습득토록 하여 우수 연구자로 양성함
- 단백 응집 및 아밀로이드 구조 생성으로 야기되는 퇴행성 뇌질환의 예방 및 치료기술 개발을 통한 경제 산업적 기여 효과를 창출함

The aim of current work is to study the molecular mechanism of protein modification and aggregate formation commonly observed in many neurodegenerative diseasesand their modulatory pathways. In addition, by using the diverse intracellular protein modification and digestive systems we are seeking to develop new strategies for the control of toxic aggregate formation in many neuronal cells.
Result
● The research experiments have been performed through the 4 subdivisions for past 5 years:
(1) Study of intracellular protein transformation, aggregation, and modulatory mechanism in major neurodegenerative diseases (2) Study of protein aggregations via various protein modifications, the characteristics of mediating enzymes, and their regulatory modes (3) Study of protein aggregatiion by regulating various proteases and degradation machineries of intracellular proteins and their regulations (4) Study of protein aggregation through the regulation of transglutaminase activity which is involved in protein cross-linking and aggregation
● We have intensively studied various aspects of abnormal protein aggregation, focusing on each specific subdivision. In subdivision (1), we identified that tau and amyloid protein transformation and their aggregation in Alzheimer disease are mediated by Dyrk1A in terms of protein phosphorylation. We also studied various protein transformation, including alpha-synuclein, synpillin-1, parkin, PINK1 and LRRK2 which are all important components of Lewy bodies in Parkinson disease. In subdivision (2), we focused on the effects of multiple mediating-enzymes, (i.e. phosphorylation, ubiquitination, sumoylation, and neddylation) on cellular physiology. As a result new findings related to their characteristics, various regulatory modes, newly-identified various protein aggregators have been identified. More specifically, (가) to study the effect of protein transformation, aggregation and degradation which are caused by phosphorylation, we intensively studied the effects of enzymes, such as ASK1, Dyrk1A, LRRK2 and PINK1. (나) we also studied the effects of ubiquitination by Parkin, CHIP and SIAH (다) To analyze the effects of sumoylation, we examined and clarified the effects of PIAS family and PC2. We also identified new substrates and regulatory factors, such as alpha-synuclein, ZNF131 and so on. (라) We studied the effects of protein neddylation and its regulatory mechanism for RCAN1 activity (RCAN1 is new substrate of RBX1).
● In subdivision (3), we tried to develop the aggregation-regulatory technology and identified the positive and negative regulatory modes for proteasome activity by parkin and ASK1.
Furthermore, we clarified telomerase as an additional novel regulator of proteasome, and are extensively now analyzing its functional mechanism and physiological significance. Finally in subdivision (4), we identified three new substrate proteins and regulatory factors related to TG2-related protein aggregation and transformation through the screening of TG2. Recently its molecular mechanism was completed and journal publication is now being prepared
● As of June 1 2012, I have published acknowledged-22 articles as a corresponding author. As co-author, I have also published non-cited 9 articles to SCI journals, and given 58 oral presentations in domestic and international meeting. Furthermore, I received an outstanding research award from the ministry of Health and Welfare, and invited a foreign scholar to Yonsei University. As a dissertation advisor, I have helped 10 graduate students to receive final degrees.
Expected Contribution
From our studies we are expecting the provide the mechanisms of protein structural alteration, subsequent aggregate formation, and the functional modulation. Additionally, we are hoping to link the structural alteration of target proteins, which are closely implicated to major neurodegenerative diseases, such as Parkinson, Alzheimer disease, and its links to the pathogenesis of these brain diseases. The future results obtained through this study may expand the current understanding of the molecular m, such asof protein aggregation and its role in pathogenesis. Also the new reg diseasesolecules which could stim dise or inhibit pathogenesis. Aof toxic target proteins could be utilized as therapeutic proteins and cytokines, as well as industrial enzymes. This study could be useful to understand molecular mechanisms of the cell death caused by the protofibrils and/or amyloids by aggregation-targeting proteins.
