질산염은 식물에 자연적으로 존재하는 물질로서 질소 순환계를 통해 아질산염으로 전환될 수 있다. 식물은 단백질을 합성하기 위한 질소원으로서 질산염을 필요로 하며 대기, 물, 토양 등을 통하여 질산염을 공급 받아 질소 순환계를 통하여 필요한 단백질을 합성한다. 따라서 채소류를 포함하는 식물 속에는 질산염이 존재할 수 있으며 특히 엽경 채소류에 질산염의 함량이 많다고 알려져 있다.
한편, 질산염 자체는 독성이 없으나 체내에서 질산염이 아질산으로 환원되어 우리몸에서 산소를 공급하는 헤모글로빈을 산소와 결합하지 못하게 만들어 청색증을

한편, 질산염 자체는 독성이 없으나 체내에서 질산염이 아질산으로 환원되어 우리몸에서 산소를 공급하는 헤모글로빈을 산소와 결합하지 못하게 만들어 청색증을 유발하거나 그 대사체들이 니트로사민과 같은 발암 물질을 만들 가능성이 있다고 알려져 있어 국내에서 채소류 섭취에 따른 질산염위해 여부에 대한 자료가 필요한 실정이다. 따라서 본 연구에서는 채소류 섭취를 통한 질산염 및 아질산염의 위해여부를 평가하기 위해 국내 유통 채소류 24종 800건에 대한 질산염과 아질산염의 수준을 조사하고 그 결과를 토대로 채소류 섭취에 따른 안전성 평가를 수행하였다. 채소류 중 질산염 및 아질산염의 실태조사를 위해 제외국의 문헌을 고찰하고 실험을 통하여 IC/ECD를 이용한 동시 분석법을 확립하였으며 유효성 측정을 위한 다양한 인자를 검토하였다.
확립된 분석법의 유효성 검증 결과, 검량선 범위는 질산염 3.125∼100 ㎎/㎏, 아질산염 0.3125∼10㎎/㎏, 상관 계수(r2)는 질산염 0.9990∼1.0000, 아질산염 0.9992∼0.9999, 최소검출한계(LOD)는 질산염 0.6 ㎎/㎏, 아질산염 0.3 ㎎/㎏, 최소정량한계(LOQ)는 질산염 2 , 아질산염 1 ㎎/㎏이었으며 회수율 범위는 질산염 89.9∼105.1%, 아질산염 88.6∼103.9%로 AOAC에서 제안하고 있는 가이드라인에 적합한 것으로 나타났다.
채소류 중 평균 질산염 검출 수준은 배추 1059.9 ㎎/㎏, 양배추 533.8 ㎎/㎏, 상추 1386.5 ㎎/㎏, 시금치 2123.8 ㎎/㎏, 쑥갓 2938.3 ㎎/㎏, 근대 2021.2 ㎎/㎏, 치커리 1754.9 ㎎/㎏, 파슬리 1336.4 ㎎/㎏, 무 1494.0 ㎎/㎏, 양파 36.0 ㎎/㎏, 당근 261.9 ㎎/㎏, 마늘 35.7 ㎎/㎏, 호박 357.7 ㎎/㎏, 오이 157.2 ㎎/㎏, 고추 69.8 ㎎/㎏, 가지 347.0 ㎎/㎏, 파 382.0 ㎎/㎏, 부추 1590.9 ㎎/㎏, 샐러리 2422.4 ㎎/㎏, 콜라비 796.7 ㎎/㎏, 콩나물 2.3 ㎎/㎏, 숙주나물 1.5 ㎎/㎏, 감자 206.5 ㎎/㎏, 고구마 71.3 ㎎/㎏이었다. 채소류 섭취에 따른 질산염의 1일 추정 섭취량(EDI)는 전체 연령에 대해 0.43∼0.69 ㎎/㎏ body weight/day, 아질산염의 1일 추정 섭취량은 0.01 ㎎/㎏ body weight/day로써 WHO에서 제안한 질산염의 TDI(3.7 ㎎/㎏ body weight/day)와 아질산염의 TDI(0.06 ㎎/㎏ body weight/day)보다 낮은 수준으로 우리나라 국민들이 채소류 섭취에 대한 위해 우려는 없는 것으로 평가되었다.

Nitrate is naturally occuring compound in plant that can be converted to nitrite in the nitrogen cycle and vice versa. Plant use nitrate as a nitrogen source, in order to synthesize protein, from air, water, soil and so on. Therefore, we can find nitrate in plant including vegetables. Especially, it is known that high level of nitrate found in leafy vegetables. Nitrate per se is relatively non-toxic, but its metabolite such as nitrite, nitric oxide and N-nitroso compounds, have raised concern because of implications for adverse health effects such as methaemoglobinaemia and carcinogenesis. To execute the risk assessment of nitrate and nitrite for the dietary intake of vegetables, we investigated the level of nitrate and nitrite in 24 vegetables(800 samples). For this monitoring, we developed and validated the analytical method using ion chromatography/conductivity detector of nitrate and nitrite, simultaneously.
According to the method, the range of calibration standard were 3.125∼100 ㎎/㎏ for nitrate(correlation coefficient, r2=0.9990∼1.0000), 0.3125∼10 ㎎/㎏(correlation coefficient, r2=0.9992∼0.9999) for nitrite and Limit of detection(LOD) and Limit of quantification(LOQ) were 0.6 ㎎/㎏ and 2 ㎎/㎏ for nitrate, 0.3 ㎎/㎏ and 1 ㎎/㎏ for nitrite, respectively. Also, the average recovery range were 89.9∼105.1% for nitrate, 88.6∼103.9% for nitrite. Those all parameters were thought to suitable for the guideline accredited by AOAC(Association of Official Agricultural Chemists)
The average levels of nitrate were 1059.9 ㎎/㎏ in chinese cabbage, 533.8 ㎎/㎏ in cabbage, 1386.5 ㎎/㎏ in lettuce, 2123.8 ㎎/㎏ in spinach, 2938.3 ㎎/㎏ in crown daisy, 2021.2 ㎎/㎏ in chard, 1754.9 ㎎/㎏ in chicory, 1336.4 ㎎/㎏ in parsley, 1494.0 ㎎/㎏ in radish, 36.0 ㎎/㎏ in onion, 261.9 ㎎/㎏ in carrot, 35.7 ㎎/㎏ in garlic, 357.7 ㎎/㎏ in pumpkin, 157.2 ㎎/㎏ in cucumber, 69.8 ㎎/㎏ in green pepper, 347.0 ㎎/㎏ in egg plant, 382.0 ㎎/㎏ in green onion, 1590.9 ㎎/㎏ in chinese chive, 2422.4 ㎎/㎏ in celery, 796.7 ㎎/㎏ in kohlrabi, 2.3 ㎎/㎏ in soybean sprouts, 1.5 ㎎/㎏ in green bean sprouts, 206.5 ㎎/㎏ in potato and 71.3 ㎎/㎏ in sweet potato, respectively.
The estimated daily intakes(EDI) were 0.43∼0.69 ㎎/㎏ body weight/day for nitrate and 0.01㎎/㎏ body weight/day for nitrite according to the ages. We could estimate that there are no health concern for eating various vegetables because those EDI were below the levels of TDI(3.7 ㎎/㎏ body weight/day for nitrate, 0.06 ㎎/㎏ body weight/day for nitrite) which were recommended by WHO.
