수입식품 증가 및 식품산업의 발전으로 새로운 식품첨가물의 사용이 늘어나고 있으며, 이러한 신규지정 첨가물에 대한 안전성 평가를 위해 인체노출안전기준 설정의 필요성이 대두되고 있다. 하지만 우리나라의 경우 식품첨가물에 대한 국내 인체노출안전기준이 설정되어 있지 않고 JECFA 등 제외국이 설정한 인체노출안전기준을 준용하여 사용하고 있다. 이로 인해 제외국에서 설정한 인체노출안전기준이 기관별로 다를 경우 위해평가를 수행할 때 어떤 값을 적용해야 할지에 대한 어려움이 있다. 따라서 본 연구에서는 식품첨가물에 대한 과학적이고 합리적인 국내 인체노출안전기준을 제안하고자 하였다. 또한 향후 국내 인체노출안전기준 설정의 타당성을 강화하기 위해서 식품첨가물의 인체노출안전기준 설정 체계를 마련하고, 보다 신속하고 합리적으로 인체노출안전기준을 재평가하기 위한 전략을 마련하고자 하였다.
인체노출안전기준 설정 대상 물질을 선정하기 위해 정해랑 등의 「유해물질의 인체안전기준 설정을 위한 중장기 기획 연구」의 연차별 평가 계획과 자료 확보 수준 및 평가 난이도, 정책 부서와의 협의를 통해 613종의 식품첨가물 중 ‘12년도 평가물질로 17종을 선정하였으며, 차년도 이후 평가를 위한 사전조사 대상물질로 4종을 선정하였다. 식품첨가물의 인체노출안전기준 설정 체계 마련을 위해 JECFA, EFSA(EU), 일본 식품안전위원회에서 사용되는 설정 원칙을 조사 및 분석하였다. 국내 5년간 출하량 및 수입량을 고려하여 노출량을 추정하는 Poundage Approach 방법을 적용하여 평가 우선순위를 설정하였으며, 재평가를 효율적으로 수행하는 전략을 마련하였다.
인체노출안전기준 설정 대상 물질의 위험성 조사 자료 검토를 토대로 전문가 그룹으로 구성된 실무작업 회의 및 평가를 통해 식품첨가물 17종에 대한 인체노출안전기준을 설정하였다. 알긴산 및 알긴산 염류(알긴산암모늄, 알긴산나트륨, 알긴산칼슘, 알긴산칼륨), 풀루란, 산화마그네슘, 수산화마그네슘, 황산마그네슘, 염화마그네슘, 탄산마그네슘은 “ADI 설정 불필요“로 설정되었다. 또한 소르빈산 및 그 염류(소르빈산칼륨, 소르빈산칼슘)의 ADI로 0-25 mg/kg bw/day를 설정하였다. 아산화질소 및 규조토는 “규정용도 내 사용 허용”, 코치닐추출색소는 0-5mg/kg bw/day”으로 설정하였다.

The use of new food additives has been increasing due to the increase in food imports and the development of the food industry. Therefore, the necessity of setting a health-based guidance value (HbGV) for safety assessment has been raised. However, in the case of Korea, a domestic HbGV of food additives has not been established, and values established in other nations, such as that of the Joint Food and Agriculture Organization of the United Nations/World Health Organization Expert Committee on Food Additives (JECFA), are in use. When the HbGVs set by other nations vary by institution, it is difficult to determine which value should be used.
Therefore, in this study we propose a scientific and rational HbGV for domestic use with food additives. Furthermore, the system for establishing HbGVs for food additives is provided, both to strengthen the validity of a domestic HbGV and to propose a strategy for re-evaluating the HbGV more rapidly and objectively.
For selecting materials for establishing a HbGV, 17 additives were used for the year 2012, out of 613 food additives. Selection was based on the annual assessment plan reported in “Medium and Long-term Study on the HbGV Set-up for Hazardous Materials” by Haerang Jung et al., along with the amount of collected data, the difficulty of assessment, and consultation with the policy division. Four food additives were selected for the pre-assessment survey for the following year. In order to provide a system for establishing HbGV for food additives, the principles used in the establishment of HbGV from the JECFA, the European Food Safety Authority, and the Japanese Food Safety Committee were investigated and analyzed. Assessment priority was set by the Poundage Approach that estimates the amount of additive that was shipped and imported in the past 5 years, and a strategy to efficiently perform re-assessment was established.
HbGVs for 17 food additives were established through work-group consultation and assessment by an expert group, based on the survey of the risk assessment data for the materials for HbGV establishment. Alginic acid and alginic acid salts (alginic acid ammonium, sodium alginate, calcium alginate, and alginic acid potassium), pullulan, magnesium oxide, magnesium hydroxide, magnesium sulfate, magnesium chloride, and magnesium carbonate were evaluated as “Acceptable Daily Intake (ADI) not specified”. In addition, the ADIs of sorbic acid and its salts (potassium sorbate and calcium sorbate) were set at 0– 25 mg/kg bw/day. HgGV for nitrous oxide and diatomaceous earth was “acceptable”, and the ADI of cochineal extract was set at 0– 5 mg/kg bw/day.
