연구의 목적 및 내용
물부추속 식물은 육상식물의 초기 계통에서 중요한 위치에 있으며 많은 종들이 세계적으로 멸종위기에 처해있다. 아시아의 물부추속 식물은 세 개 이상의 기원으로부터 파생된 것으로 여겨진다. 그 중 호주 서부에서 기원한 것으로 여겨지는 종들의 계통학적 유연관계는 밝혀지지 않았다. 본 연구에서는 동남아시아, 인도, 호주 북부와 서부에 분포하는 물부추속 식물의 (1) 분류학적 종의 한계를 설정하고, (2) 각 종들의 계통학적 유연관계를 밝히고, (3) 이동경로 및 분산기작을 파악하고자 하였다.

연구의 목적 및 내용
물부추속 식물은 육상식물의 초기 계통에서 중요한 위치에 있으며 많은 종들이 세계적으로 멸종위기에 처해있다. 아시아의 물부추속 식물은 세 개 이상의 기원으로부터 파생된 것으로 여겨진다. 그 중 호주 서부에서 기원한 것으로 여겨지는 종들의 계통학적 유연관계는 밝혀지지 않았다. 본 연구에서는 동남아시아, 인도, 호주 북부와 서부에 분포하는 물부추속 식물의 (1) 분류학적 종의 한계를 설정하고, (2) 각 종들의 계통학적 유연관계를 밝히고, (3) 이동경로 및 분산기작을 파악하고자 하였다.
연구결과
1. 분류학적 한계 설정
- 물부추속 식물 연구대상 지역에서 38종에 대한 연구재료를 확보하였다. 또한 남미와 북미 및 아프리카 지역에서 21종을 추가하여 총 59종에 대한 시료를 확보하였다.
- 대포자가 확보된 37종 47개 시료에 대하여 주사전자현미경을 이용하여 관찰하고, 표면구조물의 형태에 따라 구분하였다.
-연구과정 중 캄보디아의 미기록 물부추속 식물(I. coromandelina subsp. coromandelina) 의 형태적, 분자적 특성을 규명하였다.
2. 계통학적 유연관계 분석
- 핵과 엽록체 DNA의 다양한 염기서열(핵의 ITS 부위, LEAFY 유전자; 엽록체의 trnL, atpB-rbcL, trnS-psbC 등)을 이용하여 계통학적 유연관계를 파악하였다.
- 그 결과 I. phillipinensis는 호주동부-동아시아 분계군에 포함되었으며, I. laosiensis는 인도-호주북서부 분계군에 포함되었다.
3. 계통지리학적 연구
- 분자계통과 지리적 분포를 종합하여 S-DIVA 분석 결과, I. phillipinensis는 호주 동부에서 기원한 종이 분산기작에 의해 동아시아의 종들을 형성하는 중에 파생된것으로 추정된다.
- I. laosiensis는 대륙의 이동이후 산포에 의해 형성된 종으로 파악되었다.
4. 개체군의 유전적 특성 분석
- 인도 고유종인 I. dixitei의 5개 개체군에 대한 유전적 다양성과 개체군 구조를 파악하고 보전방안을 제시하였다.
연구결과의 활용계획
- 본 연구를 통해 도출되는 동남아시아, 인도, 호주에 분포하는 물부추속 식물의 계통 지리학적 연구결과는 향후 물부추속 식물전체의 계통학적 연구나, 이들 지역에 분포하는 식물의 분산기작과 이동경로를 규명하는 데 기여할 것이다.
- 물부추속 식물은 대표적인 멸종위기 수생식물이다. 본 연구에서 분석되는 유전정보는멸종위기식물의 보전전략을 수립하는데 중요한 기초자료로 이용이 가능하다.
- 향후 연구범위를 곤드와나 대륙에서 분리된 지역(아프리카, 호주, 남미, 인도)으로 확대하여 물부추속 전체의 계통지리학적 연구로 지속 발전시키고자 한다.

Phylogenetic relationships within species derived from West Australia are not revealed yet. The purposes of this study are (1) to determine taxonomic delimitation, (2) to reveal phylogenetic relationship, (3) to examine migration pathway and dispersal mechanism of Isoetes species from Southeast Asia, India, Northwest Australia.
Result
1. Taxonomic delimitation
-Samples for 38 species were collected from targeted research regions.
Moreover, samples for 21 species added from North America, South America, and Africa.
-Megaspores of 47 samples for 37 species were examined using scanning electronic microscope (SEM) and were classified based on the surface ornamentation.
-An unrecorded Isoetes plant was discovered from Cambodia and was identified as I. coromandelina subsp. coromandelina based on morphological and molecular characteristics.
2. Analysis of phylogenetic relationships
-Phylogenetic relationships were inferred using various region of nuclear and chloroplast DNA such as ITS, LEAFY, trnL, atpB-rbcL, and trnS-psbC.
-As result, I. phillipinensis and I. laosiensis had close relationship with East Austalia-East Asia clade and India-Northwest Australia clade respectively.
3. Phylogeographic study
- Statistical divergence-vicariance (S-DIVA) analysis was performed based on molecular phylogeny and geographical distribution. I. phillipinensis was inferred as a species formed during East Australian species diverged to East Asian species.
- I. laosiensis was inferred as a spcies that was formed by dispersal after continental drift.
4. Analysis of genentic characteristics for populations
-Genetic diversity and population structure for five population of Indian endemic plant (I. dixitei) were analyzed and conservation strategy was suggested
Expected Contribution
- Results of this study are basis of phylogenetic study on the whole of Isoetes species or study to reveal dispersal mechanism of other plants distributed in Southeast Asia, India, North-, and West-Australia.
- Isoetes is endangered aquatic plant. Genetic data analyzed in this project is important for establishment of conservation strategy of endangered plants.
- To understand whole phylogeography of Isoetes, we tend to expand geographical research area toward Gondwanan continents (Africa, Australia, South America, and India).
