Ⅳ. 연구개발결과

Ⅳ. 연구개발결과
1. 국내산 과일 및 채소류의 추출물 조제 및 생리활성물질 분리산유 추출물 등 27종에 대한 항산화성(DPPH 소거활성, ABTS 소거활성)을 조사한 결과깻잎 등 6종이 항산화 활성이 우수하였으며, 항산화활성이 높은 6종의 시료에 대하여 DPPH와 ABTS 50% 저해농도를 측정하였다. 이 중에서 항산화 활성이 가장 높은 산수유 열매에서 항산화성물질을 분리 중에 있다. 또한 항고혈압 활성이 높은 무청 주정추출물의 에칠아세테이트분획을 Amberlite XAD-4 resin column chromatography, Sephadex LH20 column chromatography 및 Prep-HPLC로 활성물질을 분리하였다. 분리된 활성물질물질을 LC/MS/MS로 분자량 측정과 물질의 질량 스펙트럼을 분석한 결과 플라보보이드 화합물로서 dimer형태로
존재하는 물질로 추정되었다.
2. 과일·채소류의 지방합성 저해, 항비만 등 대사질환 예방효과 구명
과일·채소 추출물 26종을 대상으로 췌장 지방분해효소 활성 저해능을 측정하였다. 췌장지방분해효소 활성 저해능이 높은 시료는 마늘종(Allium sativum L.), 단감(D iospyros kaki L.), 깻잎(P erilla frutescens) 등 7종으로 나타났다. 강한 췌장 지방분해효소 저해 활성을 보였던 7종의 시료(마늘종, 마늘, 키위, 단감, 사과, 깻잎, 매실)를 대상으로 지방 세포 내중성지방 생성 저해 효과를 측정한 결과, 매실과 키위 시료에서 대조군에 비해 모든 시료 처리농도에서 유의적으로 중성지방 생성이 억제되었다. 또한 마늘종, 마늘, 단감 및 깻잎처리군에서도 처리 농도가 높아질수록 지방세포의 중성지방 생성이 감소되었다. 국내산과일·채소 추출물 26종을 대상으로 당 조절 기능성 식품 소재를 탐색하였다. 26종의 시료 중 췌장베타세포에 안전한 시료 20종을 선발하였으며 선택적으로 췌장베타세포에 손상을 미치는 alloxan과 함께 20종의 시료를 처리하여 췌장베타세포에 대한 보호효과를 측정하였다.
Alloxan에 대한 시료의 췌장베타세포 생존율, LDH 방출량, 세포내 NAD+/NADH ratio, 인슐린 분비능 및 항산화효소 활성을 측정한 결과, 머위, 마늘종, 매실 및 쑥갓 시료가 당 조절능이 우수한 것으로 분석되었다. 항비만 및 당조절능이 우수한 시료 중 마늘종 추출물을 대상으로 비만마우스를 이용한 생체내 실험을 수행하였다. 8주간 고지방식이를 공급하여 실험동물의 비만을 유도한 후 4주간 매일 마늘종추출물을 경구투여하였다. 시험종료 시, 각 군간의 식이섭취량에 차이는 없었으나 마늘종추출물 투여군에서는 비만군에 비해 유의적으로 체중 및 지방조직 무게가 감소하였다. 또한 조직학적 검사를 통해, 마늘종추출물 투여군의 지방조직의 지방세포크기가 비만군에 비해 유의적으로 감소하였고 간 조직의 지방침착이 개선됨을 확인하였다. 4주간의 마늘종추출물 투여는 비만군에 비해 혈청 지질 지표, 인슐린 저항성 및 아디포카인 함량을 유의적으로 개선시켰다. 마늘종추출물 투여군에서는 간 조직의 지질 함량을 비만군에 비해 유의적으로 감소시켜 간 조직의 지방침착이 감소하는 것으로 파악된다. 마늘종추출물 투여군에서는 간 조직의 지질대사 관련 효소인 FAS 함량과 콜레스테롤 합성에 관여하는 HMG-CoA reductase 활성이 유의적으로 감소하였다. 마늘종추출물 섭취에 따른 간중 항산화효소 활성을 측정한 결과, 비만군에 비해 마늘종 추출물 투여군에서 SOD, GST, GPx 및 GR 효소 활성이 유의적으로 증가하였고 이에 따라 간 중의 지질과산화가 유의적으로 억제되었다. 또한 마늘종추출물 섭취에 따른 이상반응 등을 관찰한 결과 정상식이와 마늘종추출물을 투여한 실험군과 정상군 간의 통계적으로 유의적인 차이를 볼 수 없었다.
3. 과일 채소류의 대사성 대장질환 예방효과 구명
변비유발 동물모델에서 과일채소 분말의 섭취는 전반적인 변의 수분 보유력을 높여주었고 가을배추, 머위, 돌산갓, 우엉은 장의 변내 통과속도를 상승시켜 주어 변비 예방효과를 나타었다. 특히 가을배추 분말 섭취군의 경우 변비예방식품으로 잘 알려진 다시마 분말 섭취군과 같은 높은 장 통과속도를 보였다. 대장염유발 동물모델에서 과일채소 분말 섭취군은 점막상피증식이 관찰되지 않아 대장염 호전 효과가 관찰되었으며, 가을배추, 무시래기, 다시마, 신선초 분말 식이 섭취군에서 유의하게 염증세포수 감소 효과를 나타내었다. 과일채소 추출물이 대장암세포 증식억제에 미치는 영향을 조사한 결과 우엉, 신선초, 배추에서 공통적으로 대장암세포 증식을 억제하는 것으로 나타났다. 과채류 추출물의 대장암 예방효과 시험에서는 우엉→신선초→배추 순으로 암세포 증식을 억제함을 확인하였다.
4. 채소 및 과일의 항암, 항고혈압 효과 구명
국내산 채소 무시래기, 미나리, 씀바귀, 과일중 금귤, 피자두, 키위의 항암효과를 연구한 결과 무시래기 에탄올 추출물과 미나리 에탄올의 추출물은 유의적으로 유방암세포 증식을 억제하였으나 씀바귀뿌리 추출물은 억제 효과가 없었다. 과일 중 자두에탄올 추출물은 유방암세포 증식을 억제하였으나 키위, 금귤의 물추출물은 유방암세포 증식 억제효과가 없었고 유방암 세포 증식 억제효과는 무시래기가 가장 우수하였다.
무시래기에탄올 추출물의 항암기전을 분석한 결과 in vitro에서 암세포 증식을 유도하는 EGFR중 하나인 Erb3 단백질과 mRNA 발현과 Akt 활성형인 pAkt단백질과 Akt mRNA 발현을 유의하게 감소시켰다. 또한 세포 사멸을 방해하는 Bcl2 단백질과 유전자 발현을 유의하게 감소시켰으며 세포사멸을 유도하는 Bax 단백질과 mRNA 유전자 발현을 증가시켰다.
무시래기 에탄올 추출물중 생리활성 성분을 확인하기 위해 에틸아세테이트 분획물, dichloromethane, hexane 분획물중에서 에틸아세테이트 분획은 암세포이동 및 침윤성을 증가시켰으며, 암세포 증식에 혈관형성을 유도하는 angiogenesis를 유발하는 MMP2, MMP-9활성 및 mRNA발현을 억제하여 암세포 전이를 억제하였다. 항암, 암전이 억제 활성이 in vivo에서도 있는가를 확인하기 위해 Balb/C mice 에 유방암세포를 주입하여 폐암으로의 전이를 유도하고 무시래기 에틸아세테이트 분획물을 섭취시킨 결과 섭취량에 의존적으로 암세포 nodule의 형성을 유의하게 억제하였다.
항고혈압 연구에서 무시래기 건조분말의 섭취는 식이무게의 2.5%섭취수준에서 혈압을 유의하게 감소시켰으며 관련기전으로 폐조직에서 안지오텐신 전환효소 활성 억제, 소변을 통한 Na 배설의 증가를 통해 Na보유량을 감소시켜 혈압을 감소시키는 효과가 있음을 알 수 있었다.
또한 무시래기 에틸아세테이트 분획물을 체중 kg당 30mg, 90mg 섭취시킨 결과 혈압을 유의하게 감소시켰으며 섭취량이 증가하였을 때 혈압이 감소되는 시기가 더 빨랐다. 무시래기 에틸아세테이트 분획물은 폐조직에서 안지오텐신 효소 활성을 저해하였으며, NO 생성을 증가시키고 항산화 활성을 증가시켜 혈압을 감소시키는 것으로 나타났다. 그러나 뇨를 통한 Na 배설에는 영향을 주지 않았다. 무시래기 에틸아세테이트 분획물은 처리농도에 비례하여 혈전 용해능이 우수하였으며, 고혈압시 혈액응고시간을 유의하게 단축시켰으며 심근 조직의 괴사를 억제하는 효과가 유의하게 있었다.

The ethanol extracts of peeled sweet persimmon, sweet persimmon, dried persimmon, plum fruit (P runs salicina L., cv. ‘Soldam’), cornus fruit (Cornus officinalis) and dried radish greens were showed high antioxidant activity and then the extracts were further fractionated subsequently by hexane, dichloromethane, ethyl acetate and water. Antioxidant activities were investigated by DPPH, ABTS, and total phenol and flavonoid contents were determined. The ethyl acetate fractions of dried persimmon and cornus fruit were hihgest and similar to antioxidant activities of positive control. And the contents of total phenol and flavonoid were in the order as following; dried radish greens > cornus fruit > plum fruit > dried persimmon > sweet persimmon > peeled sweet persimmon.
The ethyl acetate fraction of cornus fruit ethanol extract showed a remarkable scavenging activity on the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. The antioxidative compounds were separated through preparative TLC and high performance liquid chromatography(HPLC), among which, six antioxidants were isolated, but their structure were not confirmed, yet. The Ethyl acetate fraction of dried radish greens ethanol extract was separated by Amberite XAD-4 resin column chromatography and TLC patterning.
The active fraction shown ACE inhibition was reseparated by Sephadex LH-20 column chromatography and preparative HPLC. Finally one single compound was isolated and anlayzed by LC/MS/MS and NMR. The active substance was estimated a flavonoid with molecular weight 864 but the structure of this compound was not confirmed, so further examination was needed.
2. Investigation of the effect of fruits and vegetables on antidiabetic and antiobesity
Among the tested extracts, Diospyros kaki L. showed the highest contents of total sugar and the content of total phenolic compounds was detected 17 species while there was not detected in I xeris dentate root and Allium fistulosum C. etc. We were screened 26 extracts of fruits and vegetables for pancreatic lipase inhibitory activity compared with orlistat. The strongest lipase inhibitory activities were shown by extract of Allium sativum L. stem, Allium sativum L. bulb, Apteryx australis, Diospyros kaki L. M alus P umila, P erilla frutescens and P runus mume regardless of concentrations treated. Eight extracts including Brassica juncea var. integrilolia had no effect on lipase inhibition. During the differentiation of 3T3-L1 preadipocyte to mature adipocyte, the cells were treated with carrier or 7 extracts(1, 5 and 10 ㎍/ml), which was shown to highest lipase inhibitory effects. We were quantified lipid contents secreted mature adipocytes and stained with Oil Red O. All cells treated with the Apteryx australis and P runus mume significantly decreased TG contents than the control. We also tested cytoprotective effect of ethanol extracts from 26 fruits and vegetables cultivated in Korea on alloxan-induced oxidative damage in pancreatic β-cell, HIT-T15. Alloxan significantly decreased cell viability, increased LDH leakage, and lowered NAD+/NADH ratio and insulin secretion in HIT-T15 cell. The HIT-T15 cells treated alloxan showed also significant decreases in the activities of antioxidant enzymes (SOD, GST, GPx and GR). However, the extracts such as P etasites japonicus, Allium sativum L. stem, P runus mume and Chrysanthemum coronarium, significantly increased viability of alloxan-treated cells and lowered LDH leakage. intracellular NAD+/NADH ratio and insulin secretion were also increased by the extracts, respectively. The HIT-T15 cells treated alloxan showed significant decreases in the activities of antioxidant enzymes, while the fruits or vegetables extracts significantly elevated the levels of antioxidant enzymes. We have demonstrated for the first time, to our knowledge, that oral administration of ASSE promotes body weight loss and the decrease of WAT weight by suppressing the hypertrophy of adipocyte and increasing the hyperplasia of adipocyte in fixed WAT.
Furthermore, it induced the reduction of TG, TC, LDL-C, VLDL-C and total lipid concentrations in the serum and the elevated ratio of HDL-C to TC near to the concentration levels in normal mice. Interestingly, our results showed that the consumption of ASSE favorably affected the levels of serum adipokines, such as adiponectin and leptin, and an insulin resistance index (HOMA-IR) in obese mice fed a high-fat diet. The ASSE-consumed groups were shown significant reductions of lipid droplets and lipid parameters in hepatic tissue. Hepatic FAS contents and HMG-CoA reductase activity were significantly lowered in the administration of ASSE groups compared to the obese group.
Hepatic antioxidant enzymes activities, includings SOD, GST, GPx and GR, were significantly higher in the ASSE groups than the obese group. In the indices of liver function and safety, ALT and AST of the ASSE-administered group fed normal diets were no significant compared to the normal control group.
These findings suggested that the screened fruit and vegetable could be a promising source as the effective and safe lipase inhibitors for isolating bioactive ingredients and developing functional foods. Based on these results, we suggest that Korean fruits/vegetables extracts have protective effects on the cytotoxicity and dysfunction of pancreatic β-cells against alloxan-induced oxidative stress through upregulation of antioxidant enzyme activities. Our findings indicate also that the beneficial antiobesity and lipid-lowering effects of ASSE are mainly mediated by suppressing body fat accumulation, ameliorating hyperlipidemic status, modulating serum concentrations of adipokines and regulating hepatic antioxidant defense system in high-fat diet induced obese mice. The results obtained from our study provide a potential effect of ASSE supplements in the regulation of obesity, and the ASSE may be of value to develop an effective dietary supplement to treat and prevent obesity and its related metabolic disorders.
3. Effect of fruit and vegetables on bowel disease, colitis and colorectal cancer
Food intake and FER of experimental groups as compared significantly different each other. In non-fed fruit & vegetables powder, many inflammatory cells occurs in the submucosal layer and mucosal epithelial proliferation was evident. But In fed fruit & vegetables powder, mucosal epithelial proliferation was not observed. Also, in fed chinese cabbage, kelp, radish leaves and Angelica keiskei L. groups, the number of inflammatory cells was significantly reduced compared to the non-fed group. These results means that fruit and vegetable intake is effective in preventing colitis.
One of the initial symptoms of colon cancer in rodents, aberrant crypt foci(ACF) similar to human colorectal cancer early symptoms. And Prevention experiments of human cancer with rodents in experimental cancer models caused ACF study is being carried out actively. In this study, we induced colon cancer in rat using azoxymethane(AOM) and observed the number of ACF by administering Angelica keiskei L. and burdock extracts to the rats orally. Experimental results, ACF were not observed in normal subjects and AOM alone treated group showed the highest number of ACF. In addition, the number of aberrant crypt(AC) that make up ACF was the higest. When treated with AOM and the extracts, the number of ACF significantly decreased and especially Angelica keiskei L. extract concentration treatment group showed the highest reduction in the number of ACF.
Results of all experiments, fruit and vegetable intake was effective in improving bowel disease. In particular, the radish leaves and Angelica keiskei L. are effective in preventing colitis and Burdock and Angelica keiskei L. are effective in preventing colorectal cancer.
4. The evaluation of anticancer effect
Ethanol extract from radish leaf, water parsely and plum inhibited MDA-MB-231 breast cancer cell proliferation significantly while ethanol extract from lxeris dentata root and water extract from kiwi and cumquat did not. The anticancer potential of radish leaf ethanol extract was highest.
The MDA-MB-231 cells were cultured in the presence or absence of various concentrations (100, 200, or 300 μg/mL) of ethanol extract of radish leaf significantly decreased cell proliferation after 48 h of incubation The protein and mRNA expression of ErbB2 were decreased significantly in a dose-dependent manner. The protein expression of ErbB3 was decreased significantly and mRNA expression of ErbB3 was decreased significantly in a dose-dependent manner. The protein expression of Akt was decreased significantly and the protein expression of pAkt was decreased significantly in a dose-dependent manner. The protein and mRNA expression of Bax were increased significantly. The protein expression of Bcl2 and mRNA expression of Bcl2 were increased significantly.
The MDA-MB-231 cells were cultured in the presence or absence of various concentrations (25, 50, or 100 μg/mL) of ethylacetate fraction of radish leaf extract significantly decreased cell proliferation after 48 h of incubation. The cell motility and invasiveness of MDA-MB-231 cells cells were decreased significantly in a dose dependent manner. The treatment of ethylacetate fraction of radish leaf decreased the protein and mRNA expression ofMMP-2 and MMP-9.
The oral intake of ethylacetate fraction of radish leaf extract at a dosage of 10mg/kg bw or 20mg/kg bw for 2 weeks decreased tumor nodule of pulmonary tissue in MDA-MB-231 cells injected balb/c mice .
In conclusion, we suggest that Raphanus sativus, L. inhibits cell proliferation via the ErbB-Akt pathway in MDA-MB-231 cells and metastasis in neoplastic balb/c mice induced with injection of MDA-MB-231 cells.
5. Antihypertensive effect
The systolic blood pressure(SBP)was measured in spontaneously hypertensive(SHR) and normotensive rats by the tail cuff method after feeding 4 weeks at the level of 2.5% or 5% in the diet. The supplementation of dried radish leaf powder decreased the SBP of SHR although the 5% supplementation level did not show more pronounced effect than caused by 2.5% level. The decrease in the SBP observed for dried radish leaf powder either 2.5% or 5%. However, the supplementation of dried radish leaf powder did not affect the SBP in normotensive rats. Dried powder of radish leaf exert antihypertensive effect in SHR caused by decreasing ACE activity and increasing urinary Na excretion.
The oral intake of ethylacetate fraction of radish leaf extract decreased blood pressure at a dose of 30mg/kg bw decreased blood pressure. The ethylacetate fraction of radish leaf extract inhibited ACE activity of pulmonary and increased NO production, antioxidative enzymes(glutathione peroxidase and catalase). However it did not change the urinary Na excretion. In addition, the supplementation of e increased fibrinolysis and extened blood clotting time in a dose dependent manner. The ethylacetate fraction of radish leaf extract decreased myocardial fibrosis significantly and did not affect the myocardial thickness and luminal diameter.
In conclusion, radish leaf had antihypertensive effect and protected myocardial tissue and the bioactive components presented in ethylacetate fraction
