1. 노로바이러스 Realtime RT-PCR 표준화 신속검사 키트 개발

1. 노로바이러스 Realtime RT-PCR 표준화 신속검사 키트 개발
노로바이러스 식중독 발생 원인조사, 실태조사 등에 임상·비임상 시료에서 공동으로 사용이 가능한 노로바이러스 Realtime RT-PCR 신속검사 키트를 개발하고자 하였다. 노로바이러스 Realtime RT-PCR 최적의 primer, probe set를 비교 선정하고 정량적 검출한계를 분석하였으며, 개발된 노로 바이러스 Realtime RT-PCR 표준화 키트에 지하수, 임상시료에서 분리된 노로바이러스를 적용하였다. 또한, 키트의 특이성 검증을 위해 식중독 바이러스 4종과 세균 2종의 유전자를 증폭하여 비특이적 반응여부를 확인하였다. 노로바이러스 GI, GII 모두 정량적 검출 한계는 1X101 copies까지 확인이 가능하였으며, 지하수, 임상시료에서 분리된 노로바이러스 RNA 유전자 모두 증폭이 가능하였다. 또한,노로바이러스를 제외한 다른 박테리아에서는 유전자가 증폭되지 않아 특이성에는 문제가 없는 것으로 확인되었다. 본 연구의 결과물은 언론홍보와 지방 식약청, 시·도보건환경연구원, 식품위생검사기관을 대상으로 시험법 교육을 통해 전파되도록 하였다. 임상과 비임상 노로바이러스 분석시료에서 공동으로 사용이 가능한 Realtime RT-PCR 키트 개발은 노로바이러스가 매개한 신속한 식중독 원인조사에 활용될 것이다.
2. 식중독바이러스 4종(A형 간염바이러스, 로타바이러스, 아스트로바이러스, 장관아데노바이러스)
PCR 표준양성대조군 개발
본 연구에는 A형 간염바이러스, 로타바이러스, 아스트로바이러스, 장관아데노바이러스 PCR 검출 시 size가 구분이 되는 PCR Standard control (RNA 또는 DNA) 표준양성대조군을 개발하고자하였다.
gene 합성과 in vitro transcription 과정을 거쳐 제작된 4종 바이러스의 표준양성대조군을 Conventional PCR을 통해 증폭한 결과 실제 분석시료의 PCR product size와 구분이 되는 PCR 양성대조군의 product size를 확인하였다. 본 연구에서 개발된 Conventional PCR 양성대조군은 식중독원인조사 검사지침에 포함시켜 국가의 식중독 원인조사 및 실태조사 등에 활용될 것이다.
3. 식품용수(지하수) 노로바이러스 유전자 특성 연구
2009년부터 2013년까지 9,150개 지점을 대상으로 노로바이러스가 검출된 140개 지점에 대한 유전자 분석결과 GI 그룹내에서는 GI-1이 21%로 가장 높은 비중을 차지하고 있었으며, GII 그룹에서는 GII-4가 59%로 가장 높게 나타났다. 또한, 식품용수에서의 노로바이러스 정량분석을 위해 realtime RT-PCR standard RNA을 이용하여 Duplex Realtime RT-PCR을 수행하였다. 2009년부터 2013년까지 식품용수 노로바이러스 실태조사를 통해 검출된 노로바이러스에 대한 정량적 측정에서는 최대 2,397 copies/L까지 확인되었다. 2011년부터 2013년까지 지하수에서 분리된 GII-4에 대한 variant 분석에서는 2010 Neworleans variant 33%로 가장 많은 검출율을 보였다. 본 연구의 결과는 식품용수(지하수)가 매개한 식중독 발생 추적과 예측에 연관 자료로 활용될 것이다.
4. 식품취급 조리종사자 노로바이러스 유행성 분석
본 연구에서는 2012년 10월부터 2013년 4월까지 총 2,729건의 국내의 무증상 식품취급 종사자에서 노로바이러스의 유행성을 분석하였다. 노로바이러스의 유행성 분석을 위해 전국 5개 시보건소로부터 무증상식품취급자의 직장도말 시료를 모았다. 총 2,729개의 시료중에 31건(1.1%)에서 노로바이러스 양성을 나타냈으며 12개의 genotype이 확인되었다. GII-4는 38.7%(n = 12), GII-17는 19.3%(n = 6), GII-2는 9.6%(n = 3), GI-4는 6.4%(n = 2), GI-6는 3.2%(n = 1), GI-7는 3.2%(n =
1), GI-12는 3.2%(n = 1), GI-13는 3.2%(n = 1), GI-14는 3.2%(n = 1), GII-3는 3.2%(n = 1), GII-6는 3.2%(n = 1), GII-16는 3.2%(n = 1)로 분석되었으며, GII-4는 가장 유행하고 있는 유전자형으로 확인되었다. 노로바이러스 GI genogroup은 0.3%(7/2,729), GII genogroup은 0.9%(24/2,729) 비율로 나타났다. 본 연구는 국내에서 식중독이 발생되지 않은 시설에서 무증상 식품취급종사자에 대해 노로바이러스 genotypes을 분석한 3번째 보고이다.

The goal of this study was development of realtime RT-PCR kit for rapid detection of NoVs from clinical and non-clinical samples. First, realtime RT-PCR primer and probe sets of NoVs were choose and we have analyse the LOD(Limit of Detection) of realtime RT-PCR kit. We was amplified from groundwater, clinical samples by realtime RT-PCR kit.
Also, we have confirmed non-specific amplification of food_borne viruses and bacteria for the validation of realtime RT-PCR kit. LOD(Limit of Detection) of realtime RT-PCR kit was detected for up to 1X101 copy both GI and GII. And realtime RT-PCR kit not amplified to antigens except NoVs. This NoV realtime RT-PCR kit be used to investigation for food_borne outbreaks linked to NoVs.

Development of Standardized positive control for conventional RT-PCR of hepatitis A virus, rotavirus, astrovirus and enteric adenovirus In this study, we developed the standardized positive control for the PCR detection of hepatitis A virus, rotavirus, astrovirus and enteric adenovirus with the supply of the size-distinguishable standard control(RNA). It was possible the size-distinguishable compare to real samples and standard control(RNA) through amplify results of conventional RT-PCR of standardized positive control of food_borne viruses(hepatitis A virus, rotavirus, astrovirus, Enteric Adenovirus). Development of standardized positive control for coventional PCR of hepatitis A virus, rotavirus, astrovirus and enteric adenovirus be used to products in order to investigation of food_borne outbreak.

A study on characteristics of isolated NoVs genome from groundwater From January 2009 to September 2013, we analyzed the nucleotide sequences of detected NoV strains to obtain the molecular epidemiological information on 140 sites where NoV was detected out of 9,150 sites in total. In result of genetic prevalence of NoV in groundwater, GI-1(21%) was the highest number of detected cases in the GI group, while GII-4(59%) had the highest number of detected cases in the GII group. To quantify the NoV capsid gene in these samples of under ground water, we performed duplex realtime RT-PCR using standard RNA. The result showed that the number of copies by realtime RT-PCR was 2,379/L. GII-4 variants appeared during the 2011/2003 season. Neworleans variant was the highest number of detected cases in the GII variant groups. This results is expected to contribute to preventing food poisoning caused by norovirus.
4. Epidemiology of norovirus infections in food handler population in Korea From October 2012 to April 2013, we investigated the molecular epidemiology of NV infections in an aymptomatic food handlers in Korea. We investigated the norovirus infections in the specimens collected from asymptomatic food handlers by 5 Public Health Centers in 5 cities. This study was approved by the Institute Review Board of NIFDS. Among total 2,729 samples, approximately 1.1% (31/2,729) of the samples was positive for NoV GI and GII during Oct 2012 to Apr 2009. The genotypic distribution of the 12 NoV strains was as follow: GII-4, 38.7%(n = 12); GII-17, 19.3%(n = 6); GII-2, 9.6%(n = 3); GI-4, 6.4%(n = 2); GI-6, 3.2%(n = 1); GI-7, 3.2%(n = 1); GI-12, 3.2%(n = 1); GI-13, 3.2%(n = 1); GI-14, 3.2%(n = 1); GII-3, 3.2%(n = 1); GII-6, 3.2%(n = 1); GII-16, 3.2%(n = 1). Among the genotypes, GII-4 was predominant(12/31, 39%). Seven(0.3%) of 2,729 samples were positive for the NoV GI genogroup, and 24(0.9%) of 2,729 samples were positive for NoV GII. This is the third report of the detection of NV genotypes in asymptomatic food handlers working at a nonoutbreak facility in South Korea.
