1-Methylcyclopropene (1-MCP), the gaseous ethylene action inhibitor, is an effective compound in delaying post-storage flesh softening of Fuyu persimmon fruits. Conventionally 1-MCP is treated to fruits by dissolving SmartFreshTR powder, the 1-MCP generating agent, in water in a storage room loaded with fruits. In this study, methods were worked out, in which a small portion of SmartFresh, either premixed with silica powder or sticked on adhesive tape, was enclosed directly into a MA packaged fruit box. The 1-MCP gas was generated from the SmartFresh by absorbing moisture from atmosphere of the package. These practical methods delayed the post-storage softening of Fuyu persimmon as effectively as the conventional method.
Non-astringent 'Fuyu' persimmon fruits has an excellent crispy texture due to a firm flesh. The flesh, however, become rapidly softened, if the fruits exposed to an ambient temperature after low temperature storage. Softening is the most ethylene sensitive ripening response, thus in this study the ethylene production of the fruits were investigated in relation to the flesh softening. The Fuyu fruits produced and accumulated only very small amounts of ethylene and ACC, especially during storage at low temperature. The flesh firmness was also well maintained for 4 months so long as they stored at a low temperature. Such a reduced softening rate seems to be closely related to the inhibited ethylene production at low temperature. But the longer the fruits stored at low temperature, the more they softened rapidly when exposed to an ambient temperature. Immediately after exposed to ambient temperature, the fruits showed a transitional ethylene peak concomitantly with a respiration burst. The AVG treatment could not only inhibit the transitional ethylene production, but it rather shortened the time to the initiation of climateric ethylene production. Unlike AVG, but 1-MCP reduced the respiration burst and effectively retarded the softening rate at the ambient temperature.
The effects of UV irradiation, as a hormetic stimulus, on the postharvest persimmon fruits (Diospyros kaki, cv. Fuyu) were investigated in regards to the change of carotenoid contents and flesh softening, when the UV irradiation was combined with or without the pretreatment of 1-methylcyclopropene (1-MCP) as an ethylene action inhibitor. The major carotenoid pigments in persimmon fruits were β–carotene, lycopene and β–cryptoxanthin. Of them, the lycopene was a pigment, which increased markedly in contents only after harvest. UV irradiation increased the contents of β–carotene and lycopene, enhancing the skin color to scarlet. The treatment accelerated however also the softening of fruit flesh. But the softening of UV irradiated fruits could be delayed significantly by pretreatment with 1-MCP without reducing the advantageous effect of UV irradiation on the carotenoid increase.
Seven kinds of 2-alkyl (chain length of 1-10 carbons) derivatives of 2-cyclopropene-1-carboxylic acid ethyl ester (CPE) were synthesized, and their effects on ethylene production, respiration and flesh softening during storage at room temperature were investigated in non-astringent 'Taishuu' and 'Fuyu' persimmon (Diosyros kaki) fruits. When treated gaseously on 'Taishuu' fruits in concentrations of 20ppm after harvest, the CPEs attached with short alkyl chain, i.e., methyl (MCPE) or ethyl (ECPE), did not delay and enhanced ethylene production of the fruits. However, those attached with long chain >3 carbons delayed and reduced it in proportion to their carbon number. That way, the ethylene production showed a maximal reduction in the fruits treated with octyl (OCPE) or decyl CPE (DCPE) and these fruts started to produce ethylene 2 days later in comparison to 1-MCP treated ones. MCPE and ECPE increased also fruit respiration. But the lengthening of the alkyl chain >4 carbons reduced respiration in proportion to the carbon number, showing a maximal reduction in DCPE. MCPE and ECPE did not inhibit flesh softening, while hexyl CPE (HCPE), OCPE, and DCPE did so strongly as nearly comparable to 1-MCP, an effective ethylene antagonist. The DCPE inhibited also the postharvest flesh solftening of 'Fuyu' fruits, when sprayed on the tree before harvest with a concentration of 0.4mM. In conclusion, the results suggest DCPE, which was storable in refrigerator at least for one month without any loss, is a good candidate for sprayable agent to prolong the postharvest shelf life of fruits. In addition, the different action of aklyl CPEs as agonist or antagonist in ethylene response according to their alkyl chain length was discussed on the basis of back-bonding and trans effect in ethylene binding site.
The skin blackening of ‘Fuyu’ persimmon fruits, which occurs often during storage, transport and marketing, severely affects on the fruit quality. In this study, pigments causing the skin blackening were extracted and characterized. According to the microscopic observation, the pigments were deposited mainly on the cell wall or in the intercellular space of intermediate cell existing between epidermaland tissue, where sometimes damaged cells were observed. The blackening pigment possessed physical properties of common melanin pigments in nature. In addition, black residue which acquired by serially washing blackened skin tissue with varied solvents produced benzoic acid and benzene acetic acid, which were not found in white residue of non-blackened normal skin tissue, when the residues were hydrolyzed by alkali fusion. From these results, the blackening pigments were believed to be a kind of allomelanin. Meanwhile, the skin tissue of persimmon fruits contained a large amount of flavonol, such as quercetin, kaempferol, catechin and epicatechin. Of them, catechin and epicatechin were not only easely oxidized and polymerized to black pigments by enzymatic oxidation in vitro, but from these synthetic black pigments were also produced the same alkali fusion products as those from blackened skin tissue samples. Thus it was concluded that the black allomelanin pigment deposited in skin tissue of persimmon fruits could be formed from catechin or epicatechin which leaked from intermediate cell damaged physically or functionally, probably by the oxidizing activities of polyphenol oxidase or peroxidase.
