(주)보락과 한국외국어대 연구진들은 최근에 자일리톨 발효 슬러지로부터 고내당성 고수율 만니톨 생산 균주를 얻어내었으며, 26S rRNA 염기서열 분석을 통하여 Candida magnoliae로 동정하였다. 이 균주를 NTG와 EMS 를 이용하여 인공돌연변이틀 유도하여 50% 이상의 포도당이 첨가된 복합배지에서 생장이 가능한 고내당성 균주를 선별하였으며 만니톨이 유일 탄소원인 배지에서 생장하지 않는 균주를 확인하는 과정을 거쳐 만니톨 생산이 우수한 균주를 선별하였다. 션별된 균주를 이용하여 최적 발효 조건을 확인한 결과 최적 발효 배지는 플라스크에서는 포도당 50g/L, 과당 100g/L, 효모추출물 10g/L, 황산 암모니움 2.5g/L, 이인산 칼륨 2.5g/L, 황산 마그네숨 0.4g/L로, 최적 pH는 5.0, 최적 발효 온도는 30도로 결정되었다. 5L jar 유가식 배양에서는 과당이 제외된 상기 배지 조건에서 균주의 생장을 유도한 후 243g/L의 과당을 공급하여 발효를 수행하였다. 이때 유가식 배양 수행한 이유는 션별된 균주의 경우 포도당보다 과당에 대한 친화도가 높아 과당을 균체 성장에 이용함으로 전체적인 수율을 떨어뜨리는 결과를 초래할 뿐 만 아니라 150g/L 이상의 과당에서는 과당 소비 속도와 만니톨 생산속도가 저하되었기 때문이다. 발효조를 통한 유가식 배양에서 용존 산소는 교반 속도를 500-600rpm으로 조절하여 배양초기에는 20% 이상 유지시키고 균체농도가 약 12g/L 되는 시점에서 교반속도를 200-350rpm으로 변경파시켜 용존산소를 제한하였다. 발효 전과정 동안 pH는 5로 조정하였으며 배양온도는 30℃, 통기량은 0.5vvm으로 조절하였다. 유가식배양을 통한 과당대비 만니톨 생산성 은 1.48g/L-h로 나타났으며 과당에 대한 만니톨의 수율은 75%, 만니톨의 최종 농도는 187g/L로 결정되었다. 또한 만니톨의 정제를 위하여 배양 상등액을 강산성 양이온수지, 약염기성 음이온 수지, 강염기성 음이온 수지 순으로 통과시켜 표준 물질로 사용하는 시그마사의 만니톨보다 불순물이 적은 만니톨을 생산하였으며 정제 수율은 93% 이고 99% 순도의 만니툴을 얻어내었다. 보다 안정적인 만니톨의 생산을 위하여 균체를 고정화 한 결과 Ca - alginate에서 반감기가 8000시간으로 가장 길며 과당대비 전환 수율이 65%로 가장 좋은 결과를 나타냄을 확인하였다. 또한 산업적 생산을 위한 5,000L scale-up 현장에서도 설험설 조건과 유사한 과당대비 만니톨 전환수율 65%, 1.5g/L-h의 생산성, 최종 만니톨 농도 168g/L를 확인하였다. 고수율 만니톨 생산균주의 개량을 위해 만니톨 생산에 가장 중요한 효소인 NAD(P)H dependent fructose reductase 유전자를 클로닝하였다. 유진자의 클로닝을 위하여 효소를 정제하여 N-빌단과 내부의 아미노산 서열을 확인하여 이로부터 탐침자를 합성하였고, 이를 이용하여 총 852개의 염기서열을 확인하였다. 이를 대장균에서 발현하여 그 활성을 확인하였고 만니톨 고생산성 균주의 유전체에 삽엽하여 기존의 균주보다 높은 생산성과 과당대비 만니톨 수율이 높은 3개의 균주를 분리하였다.

Mannitol has been produced by chemical hydrogenation of fructose. This method yields almost equal amounts of mannitol and a by-product, sorbitol. The resulting need to separate mannitol from its isomer, sorbitol, coupled with the very low conversion yield of the process, results in a high cost of production. To 0γercome these problems, many approaches to produce mannitol via microbiological techniques are being studied. Since microorganisms can specifically produce mannitol from glucose or fructosε mannitol can be isolated and purified much more easily. Until now, however, microbiological production of mannitol on an industrial scale has been prevented by low yield and productivity, and by the presence of by-products, such as glycerol and ribitol. Recently, we isolated a novel microorganism able to produce mannitol when grown in the presence of fructose and glucose as its carbon source. The isolated strain was identified as Candida magnoliae HH-01 because it has identical sequence in the D1/D2 domain of 26S rDNA and similar carbon source utilization pattem with Cmagnoliae. C. magnoliae HH-01 showed the highest mannitol yield ever reported for a mannitol-producing microorganism. Above 150g/L fructose, however, vo1umetric mannitol production rate and yield decreased because of a lag period in mannitol production and the formation of side products, respectively. In fed-batch culture, the volumetric production rate and mannitol yield from fructose were maximum at a controlled fructose concentration of 50 g/L. To Íncrease the mannitol prouction, we constructed optimal growth medium and growth condition. Under these conditions, a final mannitol production of 187 g/L, which corresponded to a yield of 75%, was obtained from 243 g/L fructose in 120h. And we a1so accomplished the scale-up for commerce production at 5,000L pilot fermentor. to yie1d a final mannitol production of 168g/L, which corresponded to a yie1d of 65 % and a productivity of 1.5g/L-h. Ca-alginate was selected as an immobilization matrix. Half life of immobilized enzyme was 8,000h and the conversion yield from fructose to mannitol was 65%. Production of mannitol in C. magnoliae is catalyzed by mannitol dehydrogenase, an enzyme that converts fructose to mannitol. For cloning of this enzyme, NADPH-dependent mannitol dehydrogenase was purified to homogeneity from C. mαgnoliαe. And N-terminal and internal amino acid sequence were obtained. NADPH-dependent mannitol dehydrogenase, containing a single open reading frame of 852 bp, was cloned using PCR primers based on ammo acid sequences. The NADPH-dependent mannitQil dehydrogenase gene from C. magnoliae was expressed in E. coli BL21 and C. magno/iae. And we selected three C. magnoliae recombinants superior to wild one.
