Ⅱ. Objectives and necessity of the research
Many kinds of antibiotics are being used to take care and prevent animal diseases in livestock farms. However, it is a serious problem regarding human health concerns (food safety) created by mis/abuse of antibiotics and inducing pathogene resistant to clinical drugs. Moreover, the side effects of exposure to fluoroquinolones on the central nervous systems of younger children have been reported for years. The safety use guidelines and maximum residue limits (MRLs) for enorfloxacin and danofloxacin in pork meat has been fixed by Korean Food and Drug Administration (KFDA) based on the necessary data's obtained for other countries, but the residual dissipation pattern of these antibiotics in the pork in Korea has not ben checked yet. Considering the potential problems that may arise due to the presence of some antibiotics in animal products, it is necessary to develop analytical method(s) for monitoring residue levels in chicken, eggs, pork, beef and milk in order to propose guidelines for safety usage and to establishment the MRL values in animal food products. Therefore, a study was carried out in order to (1) develop an economical and environment-friendly analytical method for antibiotic residue analysis using SFE/HPLC and (2) investigate residual dissipation patterns of fluoroquinolones in pig tissues by combination with charcoal as a functional additive.
Ⅲ. Content and extent of the research
In the present study, the optimal conditions for the determination of enorfloxacin, ciprofloxacin (the metabolite of enorfloxacin), and danofloxacin residues in pig tissues that includes muscle, lung, kidney and plasma have been investigated. Furthermore, the optimal conditions (time, temperature, pressure, the ratio of modifier) for SFE method and clean-up procedure were investigated to determine the antibiotic residues in pig. LODs, LOQs and recoveries of enorfloxacin, danofloxacin and ciprofloxacin were individually tested for the validity of the analysis procedure. The antibiotic residues were determined from the pig tissues at 1, 2, 3, 5, and 7 days after the final treatment based on the MRL values. Moreover, for evaluation of the effect of charcoal as a functional additive to the antibiotics, introduction of a combination of 1% charcoal, 70% of suggested amounts of antibiotics by KFDA in the feeds was done and fed to pig. The residues in blood was investigated.
Ⅳ. Application plan of research results
Multi-residue extractions and analysis of enrofloxacin, danofloxacin and ciprofloxacin were successfully done by using SFE/HPLC in pig tissues (muscle, lung and kidney) in the present study. The residues in the muscle, lung and kidney tissues were found to be present at a values less than MRL levels after 3 days of antibiotic treatment. The use of proper doses of antibiotics based on the established MRL values by KFDA in combination with charcoal as a functional additive to the antibiotics could help the rapid excretion of the residual antibiotics from the animal body.
