Ⅳ. 연구개발 결과
1. 싱싱회 경과시간 판정지표의 검색
넙치, 농어, 방어, 숭어, 조피볼락, 참돔, 홍민어 총 7종의 활어를 싱싱회로 제조하여 빙장 중 파괴강도와 ATP 관련물질의 변화를 조사하였다. 넙치는 즉살 직후 넙치의 파괴강도는 1.34±0.04 kg였으며, 시간의 경과와 더불어 파괴강도는 빠르게 상승하여 6시간 후의 파괴강도는 1.58±0.10 kg을 나타내었다. 이후 파괴강도는 감소되기 시작하였으며, 저장 16시간에 1.31±0.13 kg으로 즉살 직후에 비하여 다소 높은 값을 나타내었으나 유의적인 차이는

1. 싱싱회 경과시간 판정지표의 검색
넙치, 농어, 방어, 숭어, 조피볼락, 참돔, 홍민어 총 7종의 활어를 싱싱회로 제조하여 빙장 중 파괴강도와 ATP 관련물질의 변화를 조사하였다. 넙치는 즉살 직후 넙치의 파괴강도는 1.34±0.04 kg였으며, 시간의 경과와 더불어 파괴강도는 빠르게 상승하여 6시간 후의 파괴강도는 1.58±0.10 kg을 나타내었다. 이후 파괴강도는 감소되기 시작하였으며, 저장 16시간에 1.31±0.13 kg으로 즉살 직후에 비하여 다소 높은 값을 나타내었으나 유의적인 차이는 없었다(P>0.05).
저장 16시간 이후부터 55시간까지의 파괴강도 값은 유의적인 차이는 없었으며(P>0.05), 즉살 직후부터 6시간까지는 파괴강도 값이 증가하였으나 그 이후에는 시간이 경과함에 따라서 파괴강도 값이 감소하는 것을 확인할 수 있었다.
넙치육의 ATP 관련물질 총 함량은 약 9.20 μmole/g이며, 즉살 직후의 ATP함량은 7.67 μmole/g이였으며, 저장기간 동안 서서히 감소하여 파괴강도 값이 최대로 상승한 저장 10시간에서는 6.65 μmole/g의 ATP 함량을 나타내었다.
또한, 생선회 감칠맛의 주체가 되는 IMP 함량은 빙장 중에 서서히 증가하였으며, 즉살 직후에는 0.23 μmole/g, 저장 10시간에는 1.26 μmole/g로 증가하였다. 저장 24시간 이후에는 IMP의 함량이 5.47 μmole/g이였으며, 저장40시간에는 7.38 μmole/g로 급격한 증가를 보이고 있다.
농어는 빙장 5시간 저장 동안은 파괴강도 값이 증가하였다가 그 이후에 급격히 감소하는 경향을 나타내었다. 즉, 즉살 직후에는 1.42±0.09 kg의 파괴강도 값이 저장 5시간에 1.61±0.07 kg의 최대 파괴강도를 나타내었다.
ATP 관련물질의 총 함량은 6.47~6.55 μmole/g이였으며, 저장 기간동안 총함량의 변화는 거의 없었다. 그러나 ATP 함량은 즉살 직후에는 5.30 μmole/g이였으며, 저장 10시간에는 1.60 μmole/g, 저장 15시간에는 0.52μmole/g, 저장 25시간에는 0 μmole/g의 함량을 나타내었다.
붉은 살 생선의 대표적인 어류인 방어는 즉살 직후는 흰살 생선인 넙치등과는 달리 파괴강도 값은 1.25±0.14 kg으로 매우 낮았으며, 저장 중파괴강도의 상승은 그리 크지 못하였으며 저장 7.5시간까지는 즉살직후의 파괴강도 값과 유의적인 차이가 없었으며(P>0.05), 그 이후에는 파괴강도가 급격하게 감소하였다. ATP 관련물질의 총 함량은 약 7.29~7.30 μmole/g 이였으며, 저장기간 동안 총 함량의 변화는 거의 없었으나, ATP 함량은 즉살직후에는 6.13 μmole/g이였으며, 저장 10시간에는 0.43 μmole/g의 함량을 나타내었다.
참숭어는 즉살 직후의 파괴강도 값은 1.28 ±0.16 kg이였으며, 빙장 5시간동안 저장 후에는 1.47±0.18 kg으로 최대 파괴강도를 나타내었다. 다른 어종과 마찬가지로 ATP 관련물질의 총 함량은 약 5.00 μmole/g으로 빙장 중변화는 거의 없었으나 ATP 함량은 즉살 직후에는 3.58 μmole/g이였으며, 저장6시간에는 1.46 μmole/g, 저장 9시간에는 1.10 μmole/g, 저장 15시간에는 0μmole/g의 함량을 나타내었다.
조피볼락은 넙치와 참돔 등의 흰 살생선의 저장 중 파괴강도의 변화와 유사하게 즉살 초기에는 파괴강도 값이 1.26±0.13 kg이였으나, 저장 5시간에는 최대의 파괴강도 값인 1.52±0.13 kg으로 증가하였다. 또한 저장 10시간 이후에는 파괴강도가 감소하였으며, 저장 30시간 경과 후에는 파괴강도의 변화가 나타나지 않았다. ATP함량은 빙장 중 서서히 감소하였다. 즉살 직후
참돔의 파괴강도는 1.48±0.79 kg였으며, 시간의 경과와 더불어 파괴강도는 빠르게 상승하여 5시간 후의 파괴강도는 1.81±0.12 kg을 나타내었다.
파괴강도는 10시간 이후에 급격히 감소되기 시작하였으며, 저장 20시간에 0.85±0.12 kg으로 나타났다. 빙장 중 참돔 싱싱회의 ATP 관련물질의 총 함량은 7.95~8.00 μmole/g이였으며, ATP 함량은 빙장 중 감소하였다. 즉살한 홍민어의 파괴강도는 1.50 kg으로 나타났으며, 저장 5시간까지 육질의 단단함이 증가하였으며, 빙장 중 육의 ATP 관련물질의 총 함량은 약 5.31μmole/g이였으며, ATP 함량의 변화는 다른 어종과 마찬가지로 저장기간 동안 감소하였는데, 즉살 직후 4.80 μmole/g, 6시간 저장 후에는 3.70 μmole/g, 24시간 저장 후에는 0.30 μmole/g의 ATP 함량을 나타내었다.
어종별 싱싱회의 빙장 중 파괴강도와 ATP 함량과의 상관관계를 살펴본 결과, 모든 어종에서 싱싱회로 유통되는 시간인 10~15시간 이내까지는 파괴강도는 증가하였으며, ATP 함량은 어종별로 함량의 차이는 있으나 지속적인 감소 경향을 나타내었다.
그러므로 싱싱회의 유통시간은 파괴강도와 밀접한 관계가 있으며 파괴강도는 앞에서 언급한 바와 같이 ATP를 소모하면서 수축하게 되므로, 싱싱회의 경과시간의 판정 지표로써 어육의 파괴강도 및 어육 중 ATP함량으로 선정하였다.
2. 현장에서 루미노메타(Lumitester)에 의한 싱싱회 근육 중의 ATP 함량 측정방법 확립 및 개발방향 제시
가. 최적의 추출용매 선정
추출용매로써는 Glycine buffer, Tris-HCl buffer, Citrate buffer, Tris-malate buffer, Tricine buffer, Phosphate buffer, Borate buffer, 증류수, Perchloric acid (PCA), Trichloroacetic acid (TCA)를 농도별, pH별로 제조하여, 이들 buffer에 8 μmol의 ATP 표준물질과 어육추출액을 루미노메타로 측정하여 가장 높은 RLU값을 나타내는 추출용매를 선정하고자 하였다.
우선 농도 0.1 M, pH 6.8로 조정하여 각 추출용매를 제조하여 ATP 표준물질과 반응시켜 RLU를 측정한 결과, Tris-HCl (pH 6.8), Tris-malate buffer, Tricine, 증류수, 20 % TCA, 10 % PCA에서는 ATP 표준물질과 반응시 RLU가 나타나지 않았으며, Glycine buffer (pH 6.8)는 362,732 RLU, Citrate buffer (pH 6.8)와 Phosphate buffer (pH 6.8)는 각각 225,652 RLU와 70,660 RLU를 나타내었으며, Borate buffer (pH 6.8)는 676,402 RLU으로 나타났다.
위 결과를 토대로, ATP 표준물질을 이용한 루미노메타에서 RLU값을 얻지 못한 추출용매를 제외한, 농도별, pH별로 조제된 phosphate buffer, Citrate buffer, Borate buffer 5 mL에 어육 0.5 g을 첨가하여 소형균질기를 이용하여 40초 동안 균질화 한 후, 여과시킨 후 반응시켰다.
Phosphate buffer는 0.1 M에서 ATP 표준물질과 마찬가지로 4배까지 희석하였으나, 기계의 최대치를 초과하였으며, 0.3 M에서는 각각 1,007,841±16,596 RLU(pH 6.0), 1,857,031±85,782 RLU(pH 6.5), 2,153,742± 91,502 RLU(pH 6.8), 3,036,936±230,834 RLU(pH 7.0)를 나타내었으며, pH 7.5에서는 4배까지 희석하였으나 최대치를 초과하였다. 0.5 M에서도 0.3 M 농도의 phosphate buffer와 마찬가지로 pH가 상승할수록 높은 RLU값을 나타내었으나, 1 M에서도 기타 농도와 유사한 결과가 나타났으며, pH 7.5에서는 최대치를 초과하였다.
Citrate buffer는 phosphate buffer와 비슷한 경향으로 pH가 상승할수록 RLU값도 높아졌으나, 0.3 M, 1 M에서는 4배까지 희석하였으나 최대치를 초과하였다. 반면, 0.1 M에서는 pH 6.0에서는 449,140±112,356 RLU, pH 6.5에서는 898,817±104,022 RLU, pH 6.8에서는 1,286,510±63,258 RLU, pH 7.0에서는 1,553,825±74,680 RLU를 나타내었다. 그리고 0.5M의 농도에서는 pH별(6.0, 6.5, 6.8, 7.0, 7.5)로 각각 86,722±7,682 RLU, 310,099±2,423 RLU, 482,411±38,216 RLU, 589,265±44,292 RLU, 644,175±17,272 RLU를 나타내었다.
앞서 ATP 표준물질과의 반응에서도 살펴본 바와 같이 Borate buffer는 0.1 M에서만 pH별로 buffer가 조제되었으며, 그 결과 pH 별로 각각 560,236±85,345 RLU(pH 6.0), 896,948±101,379 RLU(pH 6.5), 1,317,381±106,916 RLU(pH 6.8), 1,881,960±50,527 RLU(pH 7.0)으로 나타나 phosphate buffer 나 glycine buffer에 비하여 낮은 RLU를 나타내었다.
이상의 결과를 살펴볼때, 현장에서 희석단계가 나뉘어지면 사용상의 문제점과 농도별, pH별로 가장 잘 반응한 phosphate buffer를 선정하였으며, 측정된 값들의 편차와 적은 희석단계 등을 고려하여, 0.5 M phosphate buffer(pH 6.8)를 루미노메타로 어육중의 ATP 함량을 측정하기 위한 최적의 추출용매로 선정하였다.
나. 최적의 추출방법 선정
본 연구의 목적은 현장에서 신속․정확하게 싱싱회 경과시간을 판정하는 것으로 어육에서 ATP를 추출하는 방법으로 일정량의 어육과 추출용매를 혼합하여 비닐팩에서 손으로 주물러 추출하는 방법을 선정하여, 사용육의 양과 추출액의 양을 조절하여 조사하였으며, 추출시간과 추출방법에 의한 어육 중의 ATP 함량을 조사하여 최적의 추출방법으로 선정하고자 하였다.
어육의 양이 증가할수록 RLU값은 증가하였으며, 추출방법에서도 균질기를 이용하는 방법과 비닐팩을 이용한 방법이 싱싱회 경과시간에 따른 ATP 함량의 감소경향이 비슷하였으며, 부위별에 따라 ATP 함량은 큰 차이를 보이지 않았다. 현장에서의 신속, 정확한 방법을 통하여 싱싱회의 경과시간을 판정하기 위한 방법으로 실험자가 손쉽게 어육에서 ATP를 추출하여 측정할 수 있는 방법을 모색하였다. 그 결과로써, 비닐팩에 어육과 추출용매를 혼합하여 손으로 주물러서 추출하고자 하였으나 실험자간의 오차 등이 확인되어, 추출방법을 비닐팩을 이용한 방법(A), 시험관을 이용한 방법(B), 막자사발을 이용한 방법(C), 소형균질기를 이용한 방법(D)으로 실험자를 달리하여 어육에서 추출된 ATP를 루미노메타로 확인한 결과, 손으로 주물러서 측정하는 방법을 실험자간의 차이를 살펴보면, 각각 709,162±47,637 RLU, 842,081±39,051 RLU로 나타나, 실험자간의 차이가 유의적인 차이를 나타내었으며, 시험관을 이용한 방법은 365,598±80,413 RLU, 422,458±10,417 RLU으로 나타나, A 추출방법보다는 편차는 작았으나, 전체적인 RLU 값은 낮았다. 막자사발을 이용한 방법에서는 413,892±118,989 RLU, 371,115±136,591 RLU로 재현성이 매우 낮게 나타났다.
마지막으로 소형균질기를 이용한 방법은 1,375,477±21,456 RLU, 1,326,508±22,857 RLU로 실험자간의 오차 및 재현성이 높았으며, 기타 추출방법보다 높은 RLU값을 나타내었다. 그러므로 신속․정확한 판정을 위하여 소형균질기를 이용하여, 소형균질기 유리 셀에 어종별 육 0.5 g, buffer 10 mL를 취해 40초간 15,000 rpm으로 균질화시키고 filtering 하여 0.5 mL를 kit에 넣어 30초간 흔든 후 1분 방치하여 루미노메타로 RLU값을 측정하는 방법을 최적의 추출방법으로 선정하였다.
최적의 추출용매와 추출방법을 이용하여 넙치, 농어, 방어, 숭어, 조피볼락, 참돔, 홍민어 총 7종의 어종을 크기별로 구입하여 싱싱회로 제작한 후, 경과시간에 따른 ATP 함량을 조사한 결과, 어종별로 초기 RLU값이 차이를 보였으며, 싱싱회 경과시간이 길어질수록 ATP 함량은 급격히 감소하는 경향을 나타내었다.
3. 현장에서 경도계에 의한 육질의 단단함의 측정방법 확립 및 개발방향 제시
휴대용 경도계를 이용하여 파괴강도 값을 측정하기 위해서 원기둥 모양과 반구 모양의 plunger의 적합성 여부를 확인하고자 하였다. 넙치 싱싱회를 제조하여 육표면에 두가지 모양의 도구를 이용하여 파괴강도 값을 측정한 결과, 원기둥 모양을 사용한 파괴강도 값은 저장 10시간까지 약간 증가하였다가 감소하는 경향을 나타냈으나, 반구모양을 사용하여 측정한 파괴강도 값의 변화는 저장기간 동안 서서히 감소하는 경향을 나타내었다.
휴대용 경도계의 원기둥을 이용하여 조피볼락 싱싱회의 파괴강도를 측정한 결과, 저장 3시간에는 0.49±0.05 kg의 파괴강도를 나타내었으며, 저장 10시간에는 0.50±0.05kg의 파괴강도를 나타내어 파괴강도의 값의 편차가 크지만 저장 10시간 이후보다는 다소 높은 값을 나타내었다. 저장 10시간까지는 평균 0.5±0.1kg을 나타내었으며, 그 이후에는 평균 0.4 kg의 파괴강도 값을 나타내었다.
참돔은 저장 기간 동안 서서히 감소하는 파괴강도 값을 나타내었으며, 저장 3시간에는 0.40±0.01 kg이였으나, 저장 10시간에는 0.42±0.04 kg의 파괴강도를 나타내었다. 그리고 10시간 이후에는 0.32~0.42 kg사이의 파괴강도 값을 나타내었다.
이상의 결과에서 휴대용 경도계를 이용한 파괴강도값 측정은 레오메타를 이용한 경우와 마찬가지로 10시간까지는 파괴강도 값이 일정하게 유지되거나 다소 증가하였다가 10시간 이후에는 감소하는 경향을 보이고는 있으나, 실험값의 편차가 크며 실험자의 누르는 힘의 차이 등에 의하여 고도로 숙련된 자에 의해서 실험이 행해져야 하는 문제점 등이 있다.
4. 싱싱회 경과시간에 따른 RLU값의 수치화
가. ATP 함량의 HPLC법의 결과와 루미노메타(Lumitester)결과의 상관성 분석
1,200 g, 800 g, 600 g 크기의 넙치를 싱싱회로 제작하여 HPLC와 루미노메타를 이용하여 어육 중의 ATP 함량을 측정한 결과, 크기가 클수록 다소 높은 RLU값을 나타내었으며, HPLC로 분석한 어육 중의 ATP 함량과 루미노메타로 분석한 ATP 함량은 싱싱회 경과시간에 따라 거의 유사하게 감소된 것을 확인할 수 있었다. 넙치의 크기별에 따른 RLU값의 변화와는 초기값이 다소 차이를 보였으나, 저장 10시간 경과시에는 비슷한 RLU값을 나타내었다. 1,200 g의 넙치의 초기 RLU값은 2,154,360±5,335 RLU를 나타내었으며, 이때 HPLC로 분석한 ATP 함량은 9.81 μmol/g이다. 10시간 경과시에는 각각 1,616,368±2,817 RLU, 7.53 μmol/g으로 나타났으며, 18시간 경과 시에는 694,464±33,443 RLU, 1.84 μmol/g으로 나타났다.
800 g에서는 초기 RLU값이 1,872,930±21,867 RLU이며, HPLC로 분석한 ATP 결과는 8.13 μmol/g으로 나타났다. 저장 10시간 경과 시에는 1,138,013±16,955 RLU, 4.65 μmol/g로 나타났다. 600 g에서는 초기 RLU값이 1,594,359±15,903 RLU로 나타났으며, 이때 HPLC로 분석한 ATP 결과는 7.89 μmol/g이였으며, 10시간 경과 시에는 922,900±13,161 RLU, 4.03 μmol/g로 나타났으며, 기타 어종에서는 유사한 결과가 나타났다.
그러므로 싱싱회의 경과시간을 실험실에서 고가의 장비인 HPLC를 이용하여 분석하는 결과와 현장에서 소형균질기를 이용하여 어육에서 ATP를 추출한 후, 루미노메타를 이용하여 분석한 결과와의 상관성이 매우 높으므로, 루미노메타를 이용하여 현장에서 신속․정확하게 싱싱회의 경과시간을 판정 할 수 있을 것으로 판단된다.
나. 파괴강도의 레오메타 결과와 휴대용경도계 결과의 상관성 분석
일정 형태의 틀을 사용하지 않고 넙치 싱싱회 육에 바로 측정하여 상관성을 조사한 결과, 일정 크기의 틀을 이용한 결과와 비슷하게, 물성측정기에서 측정한 값에 1 kg 정도 낮게 나타났으며, 두 값의 상관성이 r=0.69로 측정되었다(Fig. 3-4-3). 그러나 앞서 밝힌 바와 같이 현장에서 휴대용 경도계를 이용하여 싱싱회의 육에 대한 파괴강도를 측정시 실험자간의 누르는 힘이 다르며, 무엇보다 숙련된 기술을 요하게 된다.
이상의 결과로 살펴볼 때, 싱싱회의 경과시간을 판정할 수 있는 판정지표는 육질의 단단함, 즉 파괴강도와 ATP 함량으로 선정하였다. 본 연구의 목적이 현장에서 신속․정확하게 싱싱회의 경과시간을 판정하기 위하여 파괴강도는 휴대용 경도계, ATP 함량은 루미노메타를 이용한 방법을 제시하여, 어종별 싱싱회 경과시간에 따른 각각의 값들을 조사하였다. 그러나 휴대용 경도계를 이용한 파괴강도의 측정은 실험자간의 누르는 힘과 숙련도가 다르며, 경과시간에 따른 각 값들의 편차가 매우 적어 싱싱회 경과시간을 판정하기에는 다소 어려움이 있을 것으로 판단된다.
그러므로 싱싱회의 경과시간의 판정은 앞서 제시한 최적의 방법으로 어육에서 ATP를 추출하여 루미노메타를 이용하여 측정된 ATP 함량으로 가능할 것이다.
다. 싱싱회 경과시간에 따른 어종별 RLU값의 수치화
싱싱회로 가장 많이 제조되는 넙치, 농어, 숭어, 조피볼락, 참돔의 크기에 따른 ATP관련물질의 함량 차이를 조사한 결과, 어종의 종류와 크기에 따라서 ATP 관련물질의 총 함량은 다소 차이가 있었으며, ATP 함량도 그 차이가 있었다.

"A study into the technical development of an establishment's food safety system in order to promote the demand for fish using materials for sliced raw fish" was conducted by the Ministry of Maritime Affairs and Fisheries on 7 types of fish that are popular throughout Korea, including the olive flounder, black rockfish, red seabream, yellowtail, and seabass. The study determined that their freshness and chewy texture expired in 10 hours. Accordingly, they proposed guidelines for the gutting, processing, packaging, and transporting of fish. As a result, the raw fish industry has been revitalized with numerous processing factories being set up to produce the fish that will be sold in discount stores throughout the country.
The raw fish industry's mission was to promote the demand for fish by using materials for sliced raw fish (which were being over-supplied), and also to contribute to the livelihoods of fishermen and peoples' health in the process. In particular, we were able to prevent Vibrio septicemia and food poisoning (which is the main reason why people avoid eating sliced raw fish during the summer), and provided our customers with reliable and safe fish at a cheap price.
As mentioned before, unlike the processed raw fish in Japan, Koreans prefer chewy raw fish, which is why the expiration time for raw fish is set at 10 hours (in order to preserve that texture). However, in order to maximize profits, some discount stores are selling fish that are five days past their processed date.
Consumers who buy such outdated fish with its accompanying bad texture often decide to never purchase raw fish again. This lack of enforcement of the rules of expiration dates has been a major obstacle for attracting more customers and the raw fish industry as a whole. Yet, part of the problem also has to do with the lack of a reliable evaluation system for checking the expiration status of raw fish, making it difficult for the government to come up with an effective solution. Thus, our objective is to gain the trust of consumers, promote demand for raw fish, and contribute to the revitalization of maritime industries by developing an easy, on-the-spot method for evaluating the expiration status of fish.
To develop an index for evaluating the freshness of fish, we studied the changes in ATP related materials throughout the progression of time using a lumitester and HPLC, and we also used a Rheometer to measure the changes in meat texture. In order to make this index useful in the field, we established a method for using lumitesters for measuring ATP levels, and a portable hardness tester for measuring the level of destruction. We also selected an optimal solvent for measuring ATP levels with a lumitester, and we established a quick and accurate way to sample products in the field. Using our selected solvent and sampling methods, we produced shinshinghoe for each type of fish and studied how their ATP levels changed as time progressed. We also used a durometer in order to measure changes in the texture of the fish according to their type, size, storage temperature, and storage time. From the results attained from each of the measurement methods mentioned above, we did a correlation analysis to evaluate how they are related and find a numerical formula for the expiration time of fish. The results are as follows.
1. Searching for an evaluation index for raw fish.
We produced raw fish for 7 types of fish including olive flounder, black rockfish, red seabream, yellowtail, seabass, flathead mullet, and red drum in order to measure the changes in their breaking strength and ATP levels during their storage. The breaking strength level for the olive flounder was 1.34±0.04 kg right after being slaughtered, and it increased rapidly to reach 1.58±0.10 kg in 6 hours. Later, the breaking strength level started to slow down, and although it reached a fairly high level of breaking strength of 1.31±0.13 kg at 16 hours of storage, in comparison to the level right after its slaughter, there were no notable differences (P>0.05).
The breaking strength levels between 16 hours of storage and 55 hours of storage did not show notable differences (P>0.05), showing that the level of breaking strength decreased after rapidly increasing during the first six hours of storage.
The total content of ATP related materials in the flesh of the olive flounder was approximately 9.20 μmole/g. The ATP levels immediately after its death were 7.67 μmole/g, and then it slowly decreased to reach 6.65 μmole/g 10 hours after being stored (when the breaking strength intensity level was at its peak). Also, the IMP content, which is what gives fish their flavor, slowly increased during storage. It recorded a level of 0.23 μmole/g right after slaughter, but rose to 1.26 μmole/g during 10 hours of storage. In 24 hours of storage, the IMP content soared to 5.47 μmole/g, and after 40 hours, it reached 7.38 μmole/g.
The breaking strength levels for seabass tended to rise during the first 5 hours of storage and then rapidly decreased afterwards. The breaking strength level was 1.42±0.09 kg after slaughter and peaked at 1.61±0.07 kg 5 hours into storage.
The total content of ATP related materials was 6.47~6.55 μmole/g and there were hardly any changes in total ATP levels throughout storage. However, the ATP content which was 5.30 μmole/g immediately after slaughter, became 1.60 μmole/g for 10 hours of storage, 0.52 μmole/g for 15 hours of storage, and 0 μ mole/g for 25 hours of storage.
The breaking strength level for the yellowtail (a popular red-flesh fish) was very low at 1.25±0.14 kg(unlike white-flesh fish such as olive flounders), and did not increase very much throughout storage. Up to 7.5 hours of storage, the breaking strength level rose to reach the 1.24±0.23 kg mark, and then decreased rapidly afterwards. The total content of ATP materials was 7.29~7.30 μmole/g, and there were no major changes in total content during storage. Still, the ATP content was 6.13 μmole/g right after slaughter, and marked 0.43μmole/g after 10 hours of storage.
The breaking strength level for the flathead mullet immediately after death was 1.28±0.16kg, and peaked at 1.47±0.18 kg after 5 hours of storage. Like the other types of fish, its total content of ATP related materials was about 5.00 μmole/g and the total content did not change much throughout storage. The ATP content was 3.58 μmole/g right after slaughter, 1.46 μmole/g after 9 hours of storage, 1.10 μmole/g after 15 hours of storage, and 0 μmole/g after 15 hours of storage.
Like other white-flesh fish such as the olive flounder and red seabream, the breaking strength level of the black rockfish was 1.26±0.13 kg immediately after death and peaked at 1.52±0.13 kg after 5 hours of storage. Also the breaking strength level decreased after 10 hours of storage, and did not show any change whatsoever after 30 hours of storage. The ATP content slowly decreased during storage. The breaking strength level of the red seabream was 1.48±0.79 kg right after death, and increased rapidly to reach 1.81±0.12 kg after 5 hours of storage.
The breaking strength level then rapidly decreased after 10 hours and reached 0.85±0.12 kg after 20 hours of storage. The total content of ATP related materials for the seabream was 7.95~8.00μmole/g right after death, and it decreased throughout storage.
The breaking strength level for the red drum was 1.50 kg and the texture of the meat became harder during the first five hours of storage. The total content of ATP related materials was 5.31μmole/g and like the other fish, its ATP content decreased during its storage time. The ATP level was 4.80 μmole/g right after being slaughtered, 3.70 μmole/g after 6 hours of storage, 0.30 μmole/g after 24 hours of storage.
Observing the correlation between the breaking strength level and ATP content for each type of fish, we could see that all of them showed increasing breaking strength levels during the initial 10-15 hours of storage, while the ATP content continuously decreased (although the content levels varied across each fish type).
Thus, the expiration time of raw fish is closely related to its breaking strength level, which contracts as it consumes the ATP (as mentioned previously). For these reasons, we selected breaking strength level and ATP content as the index for evaluating the expiration time of raw fish.
2. Establishing a method for measuring ATP content in the field using the lumitester
A. Selecting an optimal solvent
As possible candidates, we produced Glycine buffers, Tris-HCl buffers, Citrate buffers, Tris-malate buffers, Tricine buffers, Phosphate buffers, Borate buffers, Water, Perchloric acids (PCA), and Trichloroacetic acids (TCA) for each level of density and pH. Using a lumitester, we measured these buffers with 8 μmol of standard ATP material and fish meat extracts in order to select the buffer with the best results.
Firstly we produced each buffer set at density 0.1 M and pH 6.8 and measured the RLU (relative light unit) in reaction to the standard ATP material.
Tris-HCl (pH 6.8), Tris-malate buffer, Tricine, Water, 20% TCA, and 10% PCA did not exhibit any RLU reaction against the ATP material. The Glycine buffer (pH 6.8) marked 362,732 RLU, while the Citrate buffer (pH 6.8) and the Phosphate buffer (pH 6.8) each showed 225,652 RLU and 70,660 RLU, respectively. The borate buffer (pH 6.8) achieved a 676,402 RLU.
Based on the results above, we excluded the buffers that did not achieve a RLU response in the previous test and produced phosphate buffers, Citrate buffers, and Borate buffers at different densities and pH levels. We then homogenized them with 5 mL of fish meat using a small homogenizer and then tested them after filtering.
At 0.1 M, we were able to dilute the Phosphate buffer up to 4X 0.1 M (as with the ATP standard), but it exceeded the maximum level of the machine. At 0.3 M we observed 1,007,841±16,596 RLU(pH 6.0), 1,857,031±85,782 RLU(pH 6.5), 2,153,742±91,502 RLU(pH 6.8), and 3,036,936±230,834 RLU(pH 7.0). At pH 7.5, we were able to dilute it up to 4X but it exceeded the maximum level.
At 0.5 M, the phosphate buffer showed higher levels of RLU as the pH increased (as with the density set at 0.3 M). Results for the 1 M testing were similar, and at 7.5 pH, we exceeded the maximum limit.
The results for the Citrate buffer test were similar to the phosphate buffer, as the RLU increased with higher pH levels. We diluted it up to 4X at 0.3 M and 1 M, but it exceeded the maximum limit. On the other hand, at 0.1 M, we observed 449,140±112,356 RLU at pH 6.0, 898,817±104,022 RLU at pH 6.5, 1,286,510±63,258 RLU at pH 6.8, and 1,553,825±74,680 RLU at pH 7.0. Also, at 0.5 M density, for each of pH level (6.0, 6.5, 6.8, 7.0, 7.5) we noticed 86,722±7,682 RLU, 310,099±2,423 RLU, 482,411± 38,216 RLU, 589,265±44,292 RLU, 644,175±17,272 RLU, respectively.
As with the responses observed for the standard ATP materials, Borate buffers for different pH levels were only produced for the 0.1 M test. The results we observed here were 560,236±85,345 RLU(pH 6.0), 896,948±101,379 RLU(pH 6.5), 1,317,381±106,916 RLU(pH 6.8), 1,881,960±50,527 RLU(pH 7.0), showing a lower RLU relative to the phosphate buffers and glycine buffers.
From these results, we selected phosphate buffers to be the most optimal for field use where the dilution levels are varied since they responded well to each of the density and pH levels. Also, in consideration to the steady measurements and lower dilution levels, we selected the 0.5M phosphate buffer (pH 6.8) to be the optimal solvent for measuring the ATP levels in fish through the Lumitester.
B. Selecting the optimal sampling method
The objective of our research is to find a quick, on-the-spot and accurate evaluation system for testing ATP levels in fish. We first considered the method of putting the solvent and fish meat into a plastic bag and mixing them together by hand, and then we experimented by putting different amounts of meat and solvents to see if it was the best way for testing ATP levels.
The level of RLU went up as the amount of fish meat increased, and the results for this method on the level of decline for ATP throughout storage time were similar to the results attained from the method using a homogenizer. Also, ATP content did not vary much throughout the various parts of the fish meat.
We searched for a raw fish evaluation method that would be quick and convenient, yet also accurate. The method involving the plastic bag was devised as a result of a brainstorming session, but there were discrepancies amongst different people conducting the tests, so we added different test methods conducted by a couple of people each to compare their results (through the lumitester and ATP content).
The different tests were: (A) Using a plastic bag and squeezing them with our hands (B) Using a test tube (C) Using a pestle and mortar (D) using a small homogenizer.
For the plastic bag method, the test showed differing results at 709,162±47,637 RLU and 842,081±39,051 RLU, showing a significant discrepancy.
The test tube method marked 365,598±80,413 RLU and 422,458±10,417 RLU showing more consistent results than method A, but the RLU was low here. The pestle and mortar method marked 413,892±118,989 RLU and 371,115±136,591 RLU showing low consistency. Finally, the homogenizer method registered the most consistent results at 1,375,477±21,456 RLU and 1,326,508±22,857 RLU exhibiting high repeatability. Also, the RLU level was higher than that of the other sampling methods.
Thus, for quickest and most accurate method for sampling was determined to be the homogenizer method. We put 0.5 g of each type of fish with 10 mL of the buffer solvent, and homogenized the two for 40 seconds at 15,000 rpm. After homogenization, the mixture was washed with 10 mL of the buffer solvent, filtered, and then put into a 0.5 mL kit to be shaken for 30 seconds and then left alone for 1 minute (afterwards it was ready for ATP testing via a lumitester).
For the purposes of the test, we produced each type of fish (the olive flounder, black rockfish, red seabream, yellowtail, seabass, flathead mullet, and red drum) in different sizes and then added them to the raw fish in order to test them via our optimal solvent and sampling method. Observing their ATP content over time, there were differences amongst the RLU levels depending on fish type, and ATP content decreased rapidly as more time passed.
3. Establishing a method for measuring fish meat texture (hardness) on the filed using a hardness tester
In order to measure the level of breaking strength using a portable hardness tester, we first wanted to check the appropriateness of using a cylindrical or a half-spherical adaptor. We produced shinshinghoe with olive flounders and measured its level of breaking strength using both the cylindrical and half-spherical tools. The level of breaking strength measured gained from the cylindrical tool method showed that the level slightly increased for the first 10 hours of storage and then decreased. The half-spherical tool method showed that the breaking strength levels slowly decreased while being in storage. Using the cylindrical portable hardness tester to measure the breaking strength level of black rockfish raw fish, we found out that the breaking strength level was 0.49±0.05 kg after 3 hours of storage, 0.50±0.05 kg after 10 hours of storage.
This showed that there was still a big gap between the two levels of breaking strength, but they were still higher than the measurement attained from black rockfish that went beyond 10 hours of storage. Up to 10 hours of storage, the average level was 0.5±0.1 kg, and afterwards the average level of breaking strength was 0.4 kg.
The red seabream exhibited breaking strength levels that decreased slowly over storage time. It reached breaking strength levels of 0.40±0.01 kg after 3 hours of storage, 0.42±0.04 kg after ten hours of storage, and 0.32~0.42 kg beyond the 10 hour mark.
As with the use of the Rheometer, the method of measuring breaking strength levels using the portable hardness tester showed levels that were constant or slightly increasing during the first 10 hours, and then a decrease afterwards. Yet, there were big discrepancies between the measurements due to differences in the amount of force that test conductors placed on the subject and thus this method requires highly experienced people to carry out the tests.
4. The statistical analysis of RLU levels for the Shinshinghoe throughout time
A. Analyzing the correlation between the lumitester results for ATP content with the results from the HPLC method
We produced olive founder raw fish pieces sized at 1,200 g, 800 g, and 600 g and tested their ATP content using both the HPLC and the lumitester. The RLU levels tended to be quite high, as the size of the meat increased. By observing the ATP content measured by HPLC and the lumitester, we were able to confirm that both of them decreased similarly over time. There were initial differences between the two in terms of variations in RLU levels due to olive flounder size, but their RLU measurement leveled out after reaching the 10 hour mark. The initial RLU level of the 1,200 g olive flounder was 1,187,505±152,069 RLU, and the ATP content measure by HPLC at this time was 9.81 μmol/g.
After 10 hours, they each recorded 712,314±2,560 RLU, 7.53 μmol/g, and after 18 hours they were measured at 694,4642±33,443 RLU, 1.84 μmol/g.
The initial RLU level for the 800 g fish was 1,872,930±21,867 RLU, and the HPLC result was 8.13 μmol/g. After 10 hours, it reached 1,138,013±16,955 RLU and 4.65 μmol/g. For the 600 g fish, the initial RLU level was 1,594,359±15,903 RLU, and the corresponding HPLC result was 7.89 μmol/g. After 10 hours the measurements read 922,900±13,161 RLU, 4.03 μmol/g, and the results were similar for all the other fish types.
The analysis results for the expiration time for raw fish using expensive lab equipment such as the HPLC displayed ahigh correlation to the results attained by using a portable homogenizer and a lumitester. Thus, it seems safe to conclude that a quick field-testing method using the lumitester is also reliable and accurate.
B. Analyzing the correlation between the breaking strength level results from the Rheometer and the hardness tester
We measured the flesh of the olive flounder without setting a specific framework in terms of size, and the results were similar to measurements that had a framework for size where the measurements were about 1 kg lower than the results measured by a hardness tester. Their correlation were calculated at r=0.69 (Fig. 3-4-3). But, as mentioned before, the pushing force varies across people conducting the test, so experience will be required for people to accurately measure the level of breaking strength for raw fish using a portable hardness tester.
From the results above, we have selected the hardness of the flesh (or level of breaking strength), and ATP content as the measuring index for evaluating the expiration of raw fish. In order to complete our objective of finding a quick and accurate method that can be used in the field, we suggested the use of a portable durometer (for measuring the level of breaking strength), the lumitester method, and tested them according to each class of fish (for measuring ATP content). However, it will be difficult to evaluate the expiration of raw fish using a portable durometer, since pushing force and experience of the conductor will vary between different test conductors. Thus, the best method overall is the lumitester method measuring ATP content.
C. The statistical analysis of RLU levels across raw fish storage
Investigating the levels of ATP material in olive founders, seabass, flathead mullet, black rockfish, and red seabream according to size, the total ATP related material levels and ATP content did vary across different sizes. Therefore, we deduced the following RLU levels for raw fish through time, in consideration to fish size and type.

If the results of this research on evaluating the expiration status of raw fish are practiced in the field, we will be able to establish a methodical distribution system that could give raw fish a reputation for being cheap, hygienic, and tasty (and naturally raise the demand for it). It would also improve the culinary culture of eating sashimi, and contribute to the revitalization of marine industries as a whole.
Thus, we must apply the new evaluation method for raw fish that we have described, as a means of regulating simultaneously the processing factories, the distribution chains, and stores and also to use it as a way of promoting the reliability of raw fish quality for the globalization of this culinary culture.
