Ⅳ. 연구개발 결과 및 활용에 대한 건의
1. 연구개발 결과
가. 곤충의 효소를 이용한 과채류 중 잔류농약 검출키트 개발 및 현장검증
1) 집파리 머리 균질화에 사용되는 완충용액 중 Triton X-100의 농도가 1% pH 8-9에서 효소는 가장 높은 활성을 보였으며 집파리는 성충시기에 AChE 활성이 가장 높으며 우화 후 4∼ 5일 경과한 후에 효소 활성이 높게 나타났다.
2) Affinity Chromatography를 사용하여 정제를 진행하였고, 그 결과 집파리와 꿀벌의 정제 정도는 230 ∼ 310배로
Ⅳ. 연구개발 결과 및 활용에 대한 건의
1. 연구개발 결과
가. 곤충의 효소를 이용한 과채류 중 잔류농약 검출키트 개발 및 현장검증
1) 집파리 머리 균질화에 사용되는 완충용액 중 Triton X-100의 농도가 1% pH 8-9에서 효소는 가장 높은 활성을 보였으며 집파리는 성충시기에 AChE 활성이 가장 높으며 우화 후 4∼ 5일 경과한 후에 효소 활성이 높게 나타났다.
2) Affinity Chromatography를 사용하여 정제를 진행하였고, 그 결과 집파리와 꿀벌의 정제 정도는 230 ∼ 310배로 측정되었으며, SDS-PAGE와 capillary electrophoresis를 통해서 순도를 검정하였다.
3) 억제제 특이성 검증에서 집파리의 경우는 BW284C51이 가장 특이적으로 AChE를 억제시켰고, 꿀벌에서는 ethopropazine보다는 BW284C51에서 더 특이적인 억제반응이 나타났다.
4) 정제한 집파리와 꿀벌 AChE를 가지고 농약 8종에 대한 억제도를 측정한 결과 각각의 농약에 대한 억제도가 효소원에 따라 억제 종도가 다르게 나타났다. 또한 정제한 효소와 조추출한 효소와의 억제도 차이는 크지 않은 것으로 나타났다.
5) 집파리 유래 AChE는 초원심분리 방법에 의한 분자형태가 잘 분리되었으며, 분자형태별 농약 억제실험에서는 조금의 억제도의 차이가 나지만 기존의 대만 kit나 SRS계통의 효소원에서 나타나는 민감도에는 미치지 못했다.
6) 상추에 농약을 살포한 후 날자 별로 잔류 농약을 추출하여 감수성 집파리로부터 추출한 곤충 효소를 이용하여 농약 저해도 측정 실험을 진행한 결과 효소에 대한 농약의 저해도는 시간이 지남에 따라 적어져 갔으며 채소에서의 잔류 농약 검증 실험은 농약 살포 4일 전으로 하는 것이 정확성이 높으며 현실성이 있는 것이다.
나. 곤충 AChE 유전자 분리, 돌연변이 유도 및 미생물에서 대량 생산
1) AChE gene cloning
한국화학연구소로부터 분양 받은 집파리로부터 분리한 유전자(hm)를 분석한 결과, OFR (open reading frame) 2,076bp이었으며, signal peptide가 80 residues, mature protein 부분이 612 residues이었다. 경상대 계통의 muature AChE gene은 gm으로 명명하였으며 1,836bp이었고 hm과 매우 유사하였다. 두 strain의 아미노산 비교에서 단지 세 개의 residues만이 차이를 보였다. 클로닝된 화학연구소 계통(pTFullHm)과 경상대 계통 (pTGm)을 유전자은행 (Genebank)에 등록하였다 (Accession number: pTFullHm (AY134873), pTGm (AF533335)). 이 두 계통을 기존의 감수성 계통이라고 Cooper 등에 의하여 보고되었던 AChE와 비교한 결과, Cooper 등은 감수성 계통과 저항성 계통의 차이를 82번째 아미노산인 I →M, 180번째 아미노산인 V → L, 262번째 아미노산인 G → A, 327번째 아미노산인 F → Y로 4군데의 아미노산이 변형되었을 때 감수성 계통에서 저항성 계통으로 돌연변이가 일어난다고 보고하였는데, 한국화학연구소로부터 분양 받은 집파리의 경우와 경상대로부터 분양 받은 집파리는 4군데 중 180번째 아미노산인 V을 제외하고는 나머지 3군데에서 모두 point mutation이 일어나 저항성 계통일 가망성이 많은 것으로 판명되었다
2) Site-directed mutagenesis
A262G mutant를 얻기 위해 A262G mega primer (72bp)를 합성하였고, 이 primer를 이용하여 얻은 PCR product의 염기 서열을 분석하였다. 그 결과 A262가 G로 변환되었음을 확인하였다. 이를 pTA262G라 명하였다. Y327f mutant를 얻기 위해 Y327F mega primer를 만들었고, 이를 이용하여 염기서열을 분석한 결과 Y327이 성공적으로 F로 치환된 것을 확인하였다. 이를 pTY327F라 명하였다. 또한 single point mutagenesis뿐만 아니라 double mutant (A262G and Y327F)를 제조하여, 이를 pTGF라 명명하였다.
3) AChE gene expression in E. coli and Pichia pastoris pTHm과 pTGm의 AChE 유전자를 발현시키기 위하여, 발현 벡터로는 Invitrogn 사의 pRSET-B vector를 사용하였고 대장균으로는 BL21 (DE3)를 사용하였다. 위와 같은 발현 과정을 통해 나온 재조합체를 각각 pRHm과 pRGm이라 명명하였다. pRHm과 pRGm을 다시 발현 호스트인 대장균에 각각 형질 전환하여 형질 전환체를 얻었다. 이런 형질 전환체들은 insoluble fraction에서 한국화학연구소 계통의 집파리 AChE, 경상대 계통 집파리 AChE가 각각 발현되는 것을 확인하였다. 그러나 이들 발현체들은 효소의 활성을 가지지 않아서 대장균에서의 발현 시스템은 적당하지 않은 것으로 판단되었다. P. paxtoris를 발현 호스트로 사용하고, 발현 벡터로는 pPICZaA를 사용하여 효소의 활성을 가지는 재조합체 pPHm (화학연구소 계통 집파리 AChE), pPA262G (pTA262G), pPY327F(pTY327F) 그리고 pPGF (pTGF)를 얻었다.
4) Optimum pH and temperature
발현된 네 가지 AChEs와 한국화학연구소 집파리로부터 분리된 AChE(Resistant)와 일본으로부터 분양 받은 국제적으로 공인된 감수성 계통의 집파리로부터 분리된 AChE (Sensitive)와 비교하여 적정 pH와 온도를 측정했다. 모든 AChEs는 pH 7.5∼8.2사이에서 최대활성을 보였고, 35∼40℃에서 최적 활성을 나타내었다.
5) 농약에 대한 inhibition assay
두개의 서로 다른 유기인계 살충제인 (그림 22), trichlorphon 과 chlorpyrifos에 대하여 발현된 AChEs의 inhibition activity를 측정한 후 Sensitive의 inhibition activity를 측정하였다. 측정된 값을 Sensitive ratio로 환산한 결과 약간의 variation은 보였으나 GF가 두 가지의 유기인계 살충제 모두에 대하여 가장 sensitive한 것으로 나타났다. 다시 말하여, trichlorphon에 대한 GF의 relative sensitivity는 72.4 %를 회복하였고 chlorpyrifos에 대하여는 82.5 %의 회복을 나타내었다. 또한 single point mutants 보다 double mutants에서 농약에 대한 민감도가 높아짐을 확인 할 수있었다.
6) Over-expression of AChE in Baculovirus expression system 농약에 대한 inhibition assay 실험에서 복합적인 mutants가 효소의 민감도에 영향을 미치는 것을 확인하였다. 그래서 triple mutant (pTIGF)를 만들었고 이를 대량 발현하기 위하여 곤충의 발현 시스템을 이용하기로 결정하였다. 이와 같은 발현 시스템으로 얻은 재조합체를 pVIGF라 명명하였고, 10% SDS-PAGE를 수행하여 이 재조합체 단백질이 대량 발현 되는 것을 확인하였다.
다. 꿀벌 AChE를 이용한 잔류농약 진단 키트 개발
1) 꿀벌 AChE 추출 조건 조사
꿀벌 AChE 추출 조건은 0.1M sodium phosphat buffer (pH 8.60 Triton X-100 0.5%함유(v/v))로 결정되었고, 균질화 시간은 7분이 적당하며, 적정 원심분리 조건은 14,450g로 나타났다. AChE 활성 측정조건 실험에서 활성은 꿀벌의 두부에서 가장 높았고, 적정온도는 40℃, pH는 8.6, 버퍼의 농도는 0.1M, 발색용액의 농도는 0.33mM 그리고 기질 농도는 0.5mM이 가장 적당한 것으로 나타났다. 꿀벌 AChE의 Km값은 1.0×10-4M이었고, 효소의 안정성 검사에서 효소 추출 후 25일이 지난 후 까지도 효소활성에는 큰 차이가 없었다. 카바메이트계 농약과의 상호작용 연구에서 농약 농도 0.06∼3.5ppm수준에서 8가지 농약(bendiocarb, carbaryl, carbofran, isoprocarb, methiocarb, methomyl, oxamyl, propoxur)은 꿀벌 AChE를 50%수준까지 저해정도를 보였다. 이 정도의 효소의 민감도라면, 유기인계나 카바메이트계 잔류 농약의 총량 검정은 충분히 가능한 것으로 판단되었다.
2) 잔류농약(카바메이트계) 추출법(Ⅰ)
잔류농약 추출 적정용매는 메탄올로 나타났고, 잔류농약의 회수율은 8가지 농약(bendiocarb, carbaryl, carbofran, isoprocarb, methiocarb, methomyl, oxamyl, propoxur)에 있어서 대개의 경우 80%이상을 나타냈다. 농약과 효소와의 반응 시간은 10분이 적당한 것으로 나타났고, 또한 포장에서의 복합적으로 잔류된 농약의 검출 실험에서도 본 개발 효소가 유용한 것으로 나타났다.
3) 잔류농약(카바메이트계) 추출법(Ⅱ)
카바메이트계 농약에 대한 정량 실험을 통해서, 딸기, 배와 쌀에 농약의 일정 농도(1.0ppm)을 처리하여 잔류 농약 추출 후 저해율을 조사한 결과 bendiocarb, carbaryl, carbofuran과 methiocarb는 80% 이상의 저해율을 나타냈고, isoprocarb, methomyl, oxamyl과 propoxur는 30% 이상의 저해율을 나타냈다. 또한 위의 8가지 농약의 50%저해율 농도로 혼합 후 작물에 처리한 다음에, 저해율의 회수율을 조사한 결과 80%이상의 회수율이 나타났다.
4) 효소의 표준화
꿀벌 두부 조추출물을 동결 건조한 후 냉장고 (0∼4℃)에 보관하면서 효소 활성 측정 결과 9개월이 지나도 67%의 활성을 유지함을 확인하였다.
5) AChE 흡착 DISC 제조기술 검토
색의 변화에 있어서는 Disc쪽 보다는 용액 쪽이 더 선명하게 나타났고, 용액을 이용한 multi-plate(MP)법에서의 적정 기질농도는 0.8mM, 발색제는 0.6mM로 나타났다. 최적 효소량은 경제성과 실효성을 비교했을 때 효소 50ul가 적당한 것으로 나타났고, 발색제량은 50ul, 기질량은 10ul, 추출용매(메탄올)는 100ul 그리고 효소와 기질과의 최적 반응시간은 5분으로 나타났다.
6) 잔류량 측정의 정확성, 분석시간 등 경제적 타당성 평가
기존의 방법보다 더 적은 초자기구 및 용매의 사용과 소량의 유기용매와 시약의 사용에 의한 환경오염과 실험자의 안정성 위해가 거의 없는 것으로 판단된다.
7) 정형화된 측정법 및 잔류허용기준과이 비교
MP법을 이용하여 잔류농약량을 측정한 결과 카바메이트계 8 가지 농약(bendiocarb, carbaryl, carbofran, isoprocarb, methiocarb, methomyl, oxamyl, propoxur) 모두 MRL 수준 이하에서도 검출이 가능한 것으로 나타났다.
8) 카바메이트계 농약별 AChE 검정/GC, HPLC 결과와의 비교 MP분석법이 기존의 기기분석법과 비교해 보았을 때 검출한계가 높게 나왔지만, 각 작물별 MRL 기준 이하에서 검출 가능한 것도 있었고, 대만 kit보다 더 간편하게 포장에서 이용할 수 있는 장점이 있다고 판단되었다.
2. 활용에 대한 건의
가. 곤충의 효소를 이용한 과채류 중 잔류농약 검출키트 개발 및 현장검증 곤충으로부터 유래한 AChE를 잔류농약 검출 kit의 효소원으로 이용할 경우 본 연구에서 실험한 곤충(담배나방, 누에, 꿀벌, 집파리) 중에서는 집파리와 꿀벌이 가장 적정한 효소원이라는 결과를 얻었다. 또한 경제성과 노력을 고려할 때 효소제조 전처리 과정이 간편 할수록 이점이 있는데 본 연구를 통하여 증명된 바와 같이 효소의 순수 정제라는 과정과 AChE의 분자형태에 따라 살충제에 대한 효소의 민감도가 달라지는 것은 아니라는 것이 밝혀짐으로서 앞으로 연구될 과제에 도움이 될 것이라 기대한다. 그리고 본 실험에서 사용한 집파리 AChE가 생화학적인 결과와 유전자 분석 결과로 볼 때 살충제 저항성 계통임이 밝혀짐에 따라 샘플 선택의 중요성이 다시 한번 더 입증되어 앞으로의 실험에 경종을 울리는 하나의 계기가 될 것으로 판단한다.
나. 곤충 AChE 유전자 분리, 돌연변이 유도 및 미생물에서 대량 생산 한국화학 연구소와 경상대 계통의 집파리 AChE를 유전자 클로닝을 수행하면서 경험한 것을 바탕으로 곤충 유래 AChE의 유전자 클로닝 시스템을 정립했기 때문에 앞으로의 유전자원 확보와 보존이라는 연구방향에 하나의 도움이 될 수 있을 것이라 판단된다. 살충제와 효소간의 상호 반응 연구에 있어, 곤충의 살충제에 대한 저항성 기작의 하나로 효소의 아미노산 순서의 변화가 될 수 있다는 것을 증명함으로 인해서, 효소의 유전자 조작은 물론 효소와 살충제 구조와의 관계에 대한 연구가 활발해 질 것으로 기대된다. 또한 곤충 유래 AChE를 잔류 농약 검출 키트로 이용할 때 이용되어 질 소모적인 효소의 량을 생각할 때 대량발현 시스템이 구축되어져 있으므로 경제적으로 많은 도움이 될 것으로 기대된다.
다. 꿀벌 AChE를 이용한 잔류농약 진단 키트 개발
생산현장과 판매현장에서 농산물 중의 잔류농약을 간단하고 신속히 검색하여 부적합한 농산물을 가려내 소비자에게 안전한 농산물만을 공급할 수 있는 방안의 일환으로, 꿀벌로부터 추출․정제한 AChE를 이용하여 농산물 시장이나 백화점, 판매현장에서 간단하게 활용할 수 있도록 kit를 개발하였다. 이는 기존에 사용되고 있는 대만 kit보다 간편하고 경비도 절약되는 kit로, 이를 통하여 저공해 농작물 재배 및 시설원예 농작물에 대한 출하 전 검사를 실시하여 품질인증 및 가격 차별화의 계기를 부여하고 품질 인증 및 가격차별화에 의한 농가소득증대와 안정적인 농업기반 조성에 이바지하며 판매 장소로 진단 키트 검사소에서나 도매(경매)전, 또는 백화점 등에서 유통되기 전에 검사를 수행하여 안전 농산물만을 판매 할 수있다. 한편 농산물 검사 기관 및 검역기관에서는 시간과 경비가 많이 드는 정밀 검사 방법을 실시하기 전의 검색단계로 활용하여 효과적인 농산물에 대한 잔류농약검사로 가능하게 할 수 있다.

Acetylcholinesterase (AChE, EC 3.1.1.7) is a universal key enzyme for neurotransmission at cholinergic synapses by catalyzing hydrolysis of the neurotransmitter acetylcholine (Eldefrawi, 1985; Soreq and Zakut, 1993) . From a view of agricultural industry, insect AChE has been a primary target by organophosphorus and carbamate insecticides, which are structurally analogous to that of acetylcholine. Upon the binding of these insecticides to the enzyme, insect AChE became inactivated due to the phosphorylation or carbamoylation of the serine residue in the active site (Corbett, 1974; Matsumura, 1985; Sussman et at., 1991). While hydrolysis occurs rapidly in the acylated enzyme by an authentic substrate acetylcholine, its rate becomes very slow in the phosphorylated or carbamoylated enzyme, essentially inactivating AChE. Due to the inactivated AChE, the post-synaptic membrane remains depolarized and synaptic transmission fails to work, resulting in paralysis and death of insects by organophosphorus and carbamate insecticides (Boublik et at., 2002). Consequently, the primary toxicity of insecticides comes from Irreversible inhibition of AChE.
Since cholinergic transmission is universally conserved among species, short-term exposure to insecticides causes ecotoxicological problems in environment and threatens to all animals including human and livestock. To its surprise, WHO estimates annual insecticides-related deaths account for a total of 220,000 persons (Veil, 1992). Many of organophosphorus and carbamate insecticides used in Korea, can cause in human and/or other susceptible animals a characteristic delayed neuropathy, in which clinical and morphological signs of axonal degeneration develop at least one week after insecticide exposure (Johnson, 1975; Abou-Donia and Lapadula, 1990). It is therefore necessary to detect the residual level of insecticides on the agricultural products to prevent insecticides-related death. Detection of these toxic chemicals can be chemically achieved by the combination of gas chromatography and mass spectrometry. But, this technique is expensive, time consuming and not suitable in field. Alternatively, AChE can be used as a sensing molecule for detecting these compounds. Insect AChE is more sensitive to a broad range of these toxic chemicals than the vertebrate enzyme (Villatte et al., 1998) and dipterans show higher sensitivity to organophosphorus and carbamate insecticides than that of hemipterans or coleopterans (Toutant, 1989). In particular, housefly (Musca domestica) AChE was known to be inactivated by organophosphorus and carbamate insecticides (Boublik et al., 2002), providing a candidate for detecting residual organophosphorus insecticides. And also the honeybee (Apis mellifera) has the same merit like the housefly as a sensing element for detecting insecticides. Due to those reasons described as above, this study was performed.
Acetylcholinesterase (AChE) was purified from the head of the housefly (M. domestica) and the honeybee (A. mellifera) by affinity chromatography. It resolved as a single peak by capillary electrophoresis (CE), in an overall yield of 33% representing 311-fold purification with a specific activity of about 107.2umol/min/mg in the housefly and in an overall yield of 37% representing 237-fold purification with a specific activity of about 32.9umol/min/mg .
These were a true AChE, with hydrolyzing more acetylthiocholine (ATC) than S-butyrylthiocholine (BTC) and propionylthiocholine (PTC) as a substrate at the same concentration. And also, approximately 90 and 78% of AChE activity was inhibited by BW284C51 and eserine, respectively, whereas only 6% of AChE activity was inhibited by ethopropazine at the same concentration of 0.1uM in the housefly AChE. In the case of the honeybee, AChE activity was inhibited more by BW284C51 than ethopropazine. The purified AChE appeared to prefer pH in the range was from 7.5 to 9.0 and optimum temperature in the prefer from 20℃ to 45℃. Optimum concentrations of ions for enzyme activity in the housefly. The were 0.01M Na+ and 0.1M K+ AChE from the housefly was successfully separated into the membrane-bound form and the soluble form. In in vitro inhibition assay with organophophates, no significant deference was obsecvcel between the crude and the purified AChE. Although the deference between the membrane-bound form and the soluble form of AChE was revealed in sensitivify to insecficides negligible, it was neligible.
The major target of insecticides is AChE which is inhibited by organophosphorus and carbamates. However, intensive use of these insecticides led to the resistance of AChE. So, it is necessary to completely understand these insecticides and control the insecticides to minimize the shock of resistance and conserve these limited resources for the control of insect (John et al., 2000). One of the insecticide resistance mechanism is mutations in the AChE gene. Two M. domestica AChE genes, which are cloned in this study, show the relationship between mutations and their resistance to insecticides. Newly cloned two AChE genes are named hm and mgm. Hmof 2076 bp contains 612 amino acids of a mature protein (67 kDa) and 80 residues of a signal peptide. And mgmof 1836 bp codes for 612 amino acids of a mature protein. Unlike other 'sensitive' strains, where enzyme activity of AChE is sensitively inhibited by insecticides, AChEs from two houseflies were 'not sensitive' to insecticides. DNA sequence data suggested that both AChEs are almost identical to the sensitive strains in their amino acid sequences with exceptions for three amino acids Ala-262, Tyr-327 and Ala-366. Site-directed mutagenesis of two residues (A262G and Y327F) into the corresponding residues of the sensitive strains, followed by expression in Pichia pastoris produced three different mutant AChEs; two single mutants A262G, Y327F, and one double mutant, GF (A262G/Y327F). These mutants restored sensitivity to the insecticides to large extents, showing that residues at 262 and 327 are the key structural elements to determine the sensitivity by the organophosphorus insecticides, trichlorphon and chlorpyrifos. Besides these residues, amino acids at 234 and 236 in the conserved sequence FGESAG among AChEs are proposed to play a role in modulating sensitivity by organophosphorus insecticides. To over-express sensitive AChE, triple mutant (pVIGF) was cloned and expressed in Sf9 cells using the baculovirus expression system.
A simple bioassay for insecticide residues was performed with the Ellman's method to detect carbamate insecticides through enzyme inhibition measurements. The principle of the bioassay is based on a modification of the Ellman's method using the honeybee (A. mellifera) head AChE as the enzyme, acetylthiocholine iodide (ATChI) as the substrate and 5,5'-dithio-bis(2-nitrobenzoic acid) (DTNB) as the color reagent. The optimum condition for enzyme activity was found to be 0.5mM ATChI, 0.33mM DTNB, pH 8.6 and 40℃. The enzyme was fairly stable at 0 ℃ ∼ 4℃ for 3 weeks as a crude extract and this enzyme could be also stored for at least 4 months when freeze-dried. On the basis of this method, the ID50 value of eight carbamate insecticies and the detection recovery percent of eight carbamate insecticides from various fruits and vegetables were established. Also, we found that AChE was inhibited by eight carbamate insecticides at 0.1ppm level.
