외래 유전자를 삽입시킨 형질전환 마우스의 생산 기술은 고등동물에서 타생물체의 유전자를 도입시켜 발현시킬 수 있음을 입증한 것으로 이를 응용한 무한한 잠재적 가능성을 제시하고 있다. 이러한 방법은 생물학, 의학 등의 기초 및 응용 분야에 널리 이용되고 있으며, 가축에 있어서는 수의 질병의 모델, 의학적 유용 물질의 생산 및 유용 단백질의 변형을 위해 포유동물을 위주로 하여 활발한 실험들이 각국의 정부, 국공립연구기관, 사립연구소에서 막대한 지원을 바탕으로 생산 기술 확립을 위하여 박차를 가하고 있다. 닭을 비롯한 가금류의 경우에서는 최근에 이르러 활발한 연구가 진행되고 있지만 기초 자료 부족과 접근 방법에 있어서의 어려움으로 아직까지 외래 유용 유전자를 도입시켜 산업화된 형질전환 닭은 보고되고 있지 않다. 따라서 국외 선도 그룹과의 격차가 거의 없어 이에 대한 지속적인 지원과 연구는 단시일 내에 국내 기술 수준을 세계적 수준으로 끌어올릴 수 있는 몇몇 안 되는 분야중 하나일 것이다.

1. Establishment of chicken embryonic germ (EG) cells
Embryonic germ (EG) cell lines established from primordial germ cells (PGCs) are undifferentiated and pluripotent stem cells. To date, EG cells with proven germ-line transmission have been completely established only in the mouse with embryonic stem (ES) cells. We isolated PGCs from 5.5-day-old (stage 28) chicken embryonic gonads and established a putative chicken EG cell line with EG culture medium supplemented with stem cell factor (SCF), leukemia inhibitory factor (LIF), basic fibroblast growth factor(bFGF), interleukin-11 (IL-11), and insulin-like growth factor-I (IGF-I). These cells grew continuously for 10 passages (4 months) on a feeder layer of mitotically active chicken embryonic fibroblasts. After several passages, these cells were characterized by screening with the Periodic acid-Shiff's reaction, anti-SSEA-1 antibody, and a proliferation assay. The chicken EG cells maintained characteristics of gonadal PGCs and undifferentiated stem cells. When cultured in suspension, the chicken EG cells successfully formed an embryoid body and differentiated into a variety of cell types. The chicken EG cells were injected into stage X blastodermal layer and produced chimeric chickens with various differentiated tissues derived from the EG cells. Chicken EG cells will be useful for the production of transgenic chickens and for studies of germ cell differentiation and genomic imprinting.
2. The production of gemline chimeras by transferring chicken EG cells
This study reported for the first time production of chicken germline chimeras by transfer of embryonic germ (EG) cells into recipient embryos of different strain. EG cells were established by culturing gonadal tissue cells of stage 28 White Leghorn (WL) embryos with I/I gene. Gonadal primordial germ cells (gPGCs) seeded with gonad stromal cells began to colonize 7 days after culture following significant (p<0.0001) increase in cell population. Colonized gPGCs 10-15 days after primary culture were then subcultured with chicken embryonic fibroblast monolayer for EG cell preparation. As experimental treatments, EG cells subcultured once or twice were transferred to stage 17 Korean Ogol chicken (KOC) embryos with i/i gene. After managed for sexual maturation up to 6 months, a testcross analysis by artificial insemination was conducted for evaluating germline chimerism. As results, EG cells transfer yielded total 2 germline chimeras out of 15 sexually matured chickens (13.3%). The efficiency of germline transmission in the chimeras was 1.5-14.6% in EG cells. In conclusion, chicken germline chimeras could be produced by transfer of EG cells derived from cultured gPGCs to recipient embryos and this might greatly contribute to establishing various innovative technologies in the field of avian transgenic research for growth improvement, disease resistance, and bioreactor production.
3. PCR Amplification for Sexing
Primers for sexing were designed based on W-chromosome specific regions. Primer set of CES#1/CES#2 amplified a fragment of 416bp and primer set of nCES#1/nCES#2 amplified a fragment of 348bp in female of both Ogol chicken for donor White Leghorn for recipient and but not in male. These primers were designed based on Xho I-repetitive sequence of W-chromosome. On W-chromosome of chicken, an 1,100bp (Eco RI family) sequence and a 717bp (Xho I family) sequence are repeated 6,000 and 14,000 times respectively. Primer set of QSEX#1/QSEX#2 amplified a fragment of 396bp on the region of W-chromosome, which is putative functional gene because this region contains splicing acceptor sequence and splicing donor sequence but the exact function is unknown. In dilution test of female and male genomic DNA, 1 ng of female genomic DNA could be detected but, even 300 ng of male genomic DNA could not be detected by PCR. These results show that these primers for sexing are very useful to distinguish between male and female of donor embryo. Although not confirmed yet, the mixed gPGCs of male and female embryo may affect the development of recipient gonads or germ cells after microinjection of manipulated gPGCs. Therefore, sexing of donor embryo is very important and indispensable pre-step for gPGCs manipulation.
It is very difficulty to be isolated genomic DNA from blood of donor embryo at stage 28 for sexing, because the amount of blood from 5.5-day-old embryo is very small and DNA extraction is time-consuming process. Therefore, Whole-blood direct PCR was performed. In whole-blood direct PCR method, blood sample from embryo was just boiled before PCR thermocycles for sexing and there was no DNA extraction step. Blood sample was boiled at 97℃ for 5 minutes and at 55℃ for 5 minutes five times to break down cell membrane and release genomic DNA in sterilized PCR tube and then PCR thermocycles was performed, which resulted in the specific band only in female embryo.
4. Production of transgenic stocks by using pantropic retrovirus (LacZ & hGH)
This study was conducted to produce the transgenic stocks by using pantropic retroviral vector containing Lac Z as a reporter gene or human growth hormone (hGH) gene and to increase the efficiency of transgenesis through several technical approaches. For the expression and titering of hGH-retroviral vector, the concentration of hGH were compared in chicken embryonic fibroblast (CEFs) and NIH3T3 cells by ELISA. The hGH was produced 178.8±76.4ng/ml and 152.3±44.0ng/ml in CEFs and NIH3T3, respectively. The retrovirus then was injected into blood vessel of 2.5-day-old embryos and incubated until further next experiments. For screening of foreign transgene, polymerase chain reaction (PCR) and Southern blotting was conducted at 6, 12-days, and hatch after transfer of retrovirus. As a result of PCR and Southern blotting, hGH gene was detected in several embryonic tissues including the embryonic gonads, regardless of the embryonic developmental stages. Of total 29 analyzed, 19 individuals were positive to hGH gene sequence with 65.5% of transgenic efficiency. Thus, transgenic aves can be efficiently produced by using pantropic retroviral vector and increase the germline transmission of transgene by the microinjection of highly concentrated retroviral vector into blood vessel.
